Claims (1)
- 【特許請求の範囲】 1.選択された器官または組織に帰巣する分子を得るための方法であって、以下 の工程を包含する、方法: a.被験体に多様な分子のライブラリーを投与する工程; b.該選択された器官または組織のサンプルを採集する工程;および c.該選択された器官または組織に帰巣する分子を同定する工程。 2.前記多様な分子のそれぞれがタグに結合されている、請求項1に記載の方法 。 3.前記タグが支持体である、請求項2に記載の方法。 4.前記ライブラリーがファージディスプレイライブラリーである、請求項1に 記載の方法。 5.前記分子がペプチドである、請求項4に記載の方法。 6.前記ライブラリーが、核酸ライブラリーおよび化学的ライブラリーからなる 群より選択される、請求項1に記載の方法。 7.前記選択された器官または組織が、脳および腎臓からなる群より選択される 、請求項1に記載の方法。 8.前記選択された器官または組織が腫瘍である、請求項1に記載の方法。 9.脳に選択的に帰巣するペプチドであって、該ペプチドが以下のアミノ酸配列 を有する、ペプチド: X1SRX2(配列番号45)、 ここでX1およびX2はそれぞれ約1個から約10個の独立に選択されたアミノ酸 である。 10.前記ペプチドがアミノ酸配列CLSSRLDAC(配列番号3)を有する、請求項 9に記載のペプチド。 11.前記ペプチドが、CNSRLHLRC(配列番号1)およびCNSRLQLRC(配列番号5 )からなる群より選択されたアミノ酸配列を有する、請求項9に記載のペプチド 。 12.脳に選択的に帰巣するペプチドであって、該ペプチドが以下のアミノ酸配 列を有する、ペプチド: X3VLRX4(配列番号46)、 ここでX3は、存在しないかまたは約1から10個の独立に選択されたアミノ酸で あり、そしてX4は、約1個から約20個の独立に選択されたアミノ酸である。 13.前記ペプチドが、CVLRGGRC(配列番号4)およびWRCVLREGPAGGCAWFNRHRL (配列番号16)からなる群より選択されたアミノ酸配列を有する、請求項12に 記載のペプチド。 14.脳に選択的に帰巣するペプチドであって、該ペプチドがCENWWGDVC(配列 番号2)およびCGVRLGC(配列番号6)からなる群より選択されたアミノ酸配列 を有する、ペプチド。 15.脳に選択的に帰巣するペプチドであって、該ペプチドがCKDWGRIC(配列番 号7);CLDWGRIC(配列番号8);およびCTRITESC(配列番号9)からなる群より 選択されたアミノ酸配列を有する、ペプチド。 16.腎臓に選択的に帰巣するペプチドであって、該ペプチドがCLPVASC(配列 番号21);CGAREMC(配列番号22);およびCKGRSSAC(配列番号23)からなる群よ り選択されたアミノ酸配列を有する、ペプチド。 17.腎臓に選択的に帰巣するペプチドであって、該ペプチドがCWARAQGC(配列 番号24);CLGRSSVC(配列番号25);およびCTSPGGSC(配列番号26)からなる群よ り選択されたアミノ酸配列を有する、ペプチド。 18.乳ガンに選択的に帰巣するペプチドであって、該ペプチドがCGRECPRLCQSS C(配列番号48)、CGEACGGQCALPC(配列番号49)、およびCNGRCVSGCAGRC(配列番 号50)からなる群より選択されたアミノ酸配列を有する、ペプチド。 19.黒色腫に選択的に帰巣するペプチドであって、該ペプチドがWGTGLC(配列 番号52)およびCLSGSLSC(配列番号55)からなる群より選択されたアミノ酸配列 を有する、ペプチド。 20.黒色腫に選択的に帰巣するペプチドであって、該ペプチドがCSVANSC(配列 番号53)、CSSTMRC(配列番号54)、およびCIEGVLGGC(配列番号56)からなる群よ り選択されたアミノ酸配列を有する、ペプチド。 21.選択された器官または組織に帰巣する分子であって、該分子が以下の工程 を包含するインビボパンニングによって同定される、分子: a.被験体に多様な分子のライブラリーを投与する工程; b.該選択された器官または組織のサンプルを採集する工程;および c.該選択された器官または組織に帰巣する分子を同定する工程。 22.前記選択された器官または組織が、脳、腎臓、腫瘍、および脈管形成の血 管系からなる群より選択される、請求項21に記載の分子。 23.前記腫瘍が、乳ガンおよび黒色腫からなる群から選択される、請求項22 に記載の分子。 24.部分を選択された器官または組織に指向させる方法であって、以下の工程 を包含する、方法: a.該部分を請求項21に記載の分子に結合させて、該部分および該分子を含 む複合体を形成させる工程;および b.該複合体と該選択された器官または組織とを接触させる工程であって、こ こで請求項21に記載の該分子が該選択された器官または組織に選択的に結合し 、これにより該部分を該選択された器官または組織に指向させる、工程。 25.前記接触工程がインビトロで行われる、請求項24に記載の方法。 26.前記接触工程がインビボで行われる、請求項24に記載の方法。 27.前記部分が検出可能な標識を含む、請求項24に記載の方法。 28.前記部分が細胞毒性因子を含む、請求項24に記載の方法。 29.前記部分が室のあるマイクロデバイスを含む、請求項24に記載の方法。 30.標的分子の存在を同定する方法であって、該標的分子は選択された器官ま たは組織に選択的に帰巣する分子に特異的に結合し、該方法は以下の工程を包含 する、方法: a.該標的分子を発現すると疑いのある器官または組織のサンプルを得る工程 ; b.該サンプルと請求項21に記載の分子とを、請求項21に記載の該分子と 該標的分子との特異的結合を可能にする条件下で接触させる工程;および c.請求項21に記載の該分子の特異的結合を検出する工程であって、該特異 的結合が該サンプル中の該標的分子の存在を同定する、工程。 31.標的分子を実質的に単離する方法であって、該標的分子は選択された器官 または組織に選択的に帰巣する分子に特異的に結合し、該方法は以下の工程を包 含する、方法: a.該選択された器官または組織のサンプルを得る工程であって、該器官また は組織が該標的分子を発現している、工程; b.該サンプルと請求項21に記載の分子とを、請求項21に記載の該分子と 該標的分子との特異的結合を可能にする条件下で接触させ、複合体を形成させる 工程;および c.該複合体から該標的分子を実質的に単離する工程。 32.請求項31に記載の方法により得られる、実質的に単離された標的分子。 33.脈管形成血管系を含有する器官または組織をインビボで標的化する方法で あって、該器官または組織とαV含有インテグリンに特異的に結合する分子とを 接触させる工程を包含する、方法。 34.前記器官または組織が腫瘍である、請求項33に記載の方法。 35.前記分子が、RGD含有ペプチド、およびαV含有インテグリンに特異的に結 合する抗体からなる群より選択される、請求項33に記載の方法。 36.前記αV含有インテグリンがαVβ3インテグリンである、請求項33に記 載の方法。 37.前記分子がCDCRGDCFC(配列番号57)である、請求項33に記載の方法。
