Claims (1)
- 【特許請求の範囲】 1.生物学的材料中の感染性化合物(IC)、特にタンパク質性感染性化合物を 分離および/または検出および/または定量する方法であって、該化合物は、感 染因子の特異構成性化合物、内因性または外因性の完全または不完全な感染因子 を含む構造体、それらの代謝物、該感染因子のある種の特異性を示すこれらの感 染因子の組み立て体、および上述の化合物によるかまたは異常様式で発現される 遺伝子の発現によって生物において特異的に誘導される化合物からなる群より選 択され、β2GPI/IC複合体(ここでβ2GPIはβ2糖タンパク質Iであり、かつICは 感染性化合物であり、該複合体は適切な手段によって同定され得、かつ (a)(β2GPI)n/IC;(β2GPI)nは生物学的材料中に存在するβ2糖タンパク 質Iであり、そしてICはウイルス化合物VICまたは非ウイルス化合物nVICである、 (b)(β2GPI)a/nVIC;(β2GPI)aは生物学的材料中に添加されるβ2糖タン パク質Iであり、そしてnVICは非ウイルス感染性化合物である、 からなる群より選択される)が、分離および/または検出および/または定量さ れることに特徴づけられる、方法。 2.前記ICが、β2GPIに結合する物質を使用して、β2GPI/IC複合体を認識す ることによって、分離および/または検出および/または定量されることに特徴 づけられる、請求項1に記載の方法。 3.前記β2GPI/IC複合体が、該複合体が形成される媒体において、該複合体 を支持体へ前もって付着させずに、該媒体を付着させてまたは付着させないで、 分離および/または検出および/または定量されることに特徴づけられる、請求 項1または2に記載の方法。 4.前記β2GPI/IC複合体に含まれる要素の1つが、支持体上に保持されるこ とに特徴づけられる、請求項1または2に記載の方法。 5.前記支持体が固体支持体であることに特徴づけられる、請求項4に記載の 方法。 6.使用される前記支持体が、膜、マイクロタイタープレート、または顕微鏡 スライドであることに特徴づけられる、請求項5に記載の方法。 7.前記β2GPI/IC複合体が、複合体のβ2GPI要素によって前記支持体に保持 されることに特徴づけられる、請求項4から6のいずれか一項に記載の方法。 8.前記β2GPI/IC複合体が、複合体のIC要素によって前記支持体に保持され ることに特徴づけられる、請求項4から6のいずれか一項に記載の方法。 9.前記IC要素が、前記β2GPI/IC複合体が形成される前に前記支持体に付着 され、次に、該複合体が形成され、そして次に、該複合体の該β2GPI要素が分離 および/または検出および/または定量されることに特徴づけられる、請求項8 に記載の方法。 10.物理的、化学的、または生化学的に支持体へ付着される、生物学的材料 のICが、前記複合体のβ2GPIに結合する物質を使用して、分離および/または検 出および/または定量されることに特徴づけられる、請求項8または9に記載の 方法。 11.前記β2GPI/IC複合体が、該複合体のβ2GPI部分またはIC部分のいずれ かに結合する化合物によって支持体に保持され、そこで該支持体に付着される複 合体が、生物学的材料から分離され、そして該複合体が、複合体の他方の部分の 認識により分離および/または検出および/または定量されることに特徴づけら れる、請求項1または4に記載の方法。 12.前記複合体が、複合体のβ2GPI部分に結合する化合物によって前記支持 体に保持されることに特徴づけられる、請求項11に記載の方法。 13.前記複合体が、複合体のIC部分に結合する化合物によって前記支持体に 保持されることに特徴づけられる、請求項11に記載の方法。 14.β2GPIに結合する化合物が、β2GPIに特異的である抗体、別のタンパク 質、およびβ2GPIに結合する生物学的または化学的化合物からなる群より選択さ れることに特徴づけられる、請求項2に記載の方法。 15.β2GPIを認識するタンパク質がHIV2 p26 RODタンパク質であることに特 徴づけられる、請求項14に記載の方法。 16.前記複合体のICが、HIV1、HIV2、HSV、HBV、およびウイルス起源の粒子 またはタンパク質からなる群に属することに特徴づけられる、請求項1に記載の 方法。 17.前記複合体のICが、細菌、寄生虫、マイコプラズマ、カビ、および異常 動物細胞からなる群に属することに特徴づけられる、請求項1に記載の方法。 18.支持体に付着される前記β2GPI/IC複合体のICが、感染性、特異酵素反 応、蛍光または放射性標識トレーサー、標識プローブとのハイブリダイゼーショ ンまたはポリメラーゼ連鎖反応(PCR)による核酸の検出、あるいはICに特異的 である抗体の使用によって、分離および/または検出および/または定量される ことに特徴づけられる、請求項7に記載の方法。 19.使用されるβ2GPIの形態が、純粋形態であるかまたは純粋形態ではない β2'GPIであることに特徴づけられる、請求項1から18のいずれか一項に記載 の方法。 20.前記ICとの複合体を組み立てるために使用されるβ2GPIが標識され、該 複合体が、該標識により検出および/または定量されることに特徴づけられる、 請求項1から19のいずれか一項に記載の方法。 21.前記β2GPIが、酵素、放射性産物、または蛍光産物によって標識される ことに特徴づけられる、請求項20に記載の方法。 22.前記β2GPI要素が、前記β2GPI/IC複合体が形成される前に前記支持体 に付着され、次に、該複合体が形成され、そして次に、該複合体のIC要素が検出 および/または定量されることに特徴づけられる、請求項6に記載の方法。 23.前記複合体の要素の少なくとも1つが、動物起源であるか、あるいは遺 伝子工学および/または化学工学によって生成されることに特徴づけられる、請 求項1から22のいずれか一項に記載の方法。 24.請求項4または11のいずれかに記載の方法を実施するための固体支持 体であって、それが(β2GPI)n/IC複合体をその構成性要素の少なくとも1つ によって、直接または間接に、特異的または非特異的に保持するのに適している ことに特徴づけられる、固体支持体。
