Claims (1)
- 【特許請求の範囲】 1.(a)図1に示される核酸配列あるいは(b)その配列の一部であって、図1に 示されるアミノ酸配列の細胞外ドメイン、膜貫通部分、または細胞内ドメインを コードする配列あるいは(c)その配列の一部であって、図1に示されるアミノ酸 配列の細胞外ドメイン、膜貫通部分および細胞内ドメインをコードする配列を含 む実質的に精製された組換え核酸分子。 2.a)図1の192〜1610のヌクレオチド; b)図1の1611〜1697のヌクレオチド; c)図1の1698〜2738のヌクレオチド;または d)図1の192〜2738のヌクレオチド を含む、請求項1に記載の核酸。 3.a)図1に示されるアミノ酸配列を含有するか;あるいは b)細胞外ドメイン、膜貫通部分、または細胞内ドメインを含有する(a) のタンパク質の一部を含有するか;あるいは c)細胞外、膜貫通および細胞内領域を含有する(a)のタンパク質の一部 を含有するか; d)a)、b)、またはc)のタンパク質と機能的に等価であるか;あるい は e)a)、b)、またはc)のタンパク質と実質的な類似性を有する、 実質的に精製されたタンパク質。 4.図1の20〜492のアミノ酸を含有する、請求項3に記載のタンパク質。 5.請求項3または4に記載のタンパク質をコードする核酸分子に、作動的に 連結させた発現調節配列を含有する、発現ベクター。 6.請求項1または2に記載の核酸分子に、作動的に連結させた発現調節配列 を含有する、請求項5に記載の発現ベクター。 7.即時性遺伝子プロモーターを含有する、請求項5または6に記載のベクタ ー。 8.前記即時性遺伝子プロモーターがfosプロモーターまたはjunプロモーター である、請求項7に記載のベクター。 9.請求項5〜8のいずれかにおいて定義される発現ベクターを保有する、細 胞。 10.請求項5〜8のいずれかに記載の発現ベクターを保有する、微生物。 11.PC12細胞株またはNIH 3T3細胞株由来の細胞で、請求項7または8にお いて定義されるベクターを保有する、請求項9に記載の細胞。 12.即時性遺伝子プロモーターの制御下で、発現可能な検出マーカーをコー ドする遺伝子をさらに含有する、請求項11に記載の細胞。 13.無血清培地中で成長因子依存的であり、そして請求項6〜8のいずれか に記載の発現ベクターを含有する、繊維芽細胞株。 14.請求項1、5または6に記載の核酸分子にハイブリダイズするヌクレオ チド配列であり、そして該配列が正常な筋よりも除神経筋においてより高いレベ ルで発現されるチロシンキナーゼをコードする、ヌクレオチド配列。 15.請求項3または4に記載のタンパク質に結合するリガンドを同定するプ ロセスであって、 a)無血清培地中では生存しない成長因子依存性細胞を、請求項1におい て規定される核酸または請求項5〜8のいずれかにおいて規定される発現ベクタ ーで形質転換する工程; b)そのような形質転換細胞を潜在的Dmk結合リガンドの存在下、無血清 培地中で培養する工程;および c)無血清培地中で該形質転換細胞の生存を支持するようなリガンドを同 定する工程、 を包含するプロセス。 16.請求項15に記載の方法で同定されるリガンド。 17.健常人と比較した患者中のDmkタンパク質の発現レベルの差が、患者の 障害が一次的または二次的にDmk代謝と関連し得ることを示唆する場合において 、患者のサンプル中の請求項3または4において定義されるDmkタンパク質の発 現レベルと、健常人由来の比較用サンプル中の請求項3または4において定義さ れるDmkタンパク質の発現レベルとを比較する工程を包含する、患者における筋 肉障害または他の障害のエクスビボ診断法。 18.前記障害が特発性ねじれ失調症である、請求項17に記載の方法。 19.筋肉障害または他の障害を患うヒトあるいは動物の身体を処置する方法 における用途のための、請求項3または4において同定されるタンパク質、ある いは該タンパク質のペプチドフラグメントまたは誘導体。 20.チロシンキナーゼレセプター遺伝子の少なくとも一部をクローニングす る方法であって、 (i)cDNA分子の収集物をテンプレートとして用い、そしてa)配列番号3 に基づく縮重プライマー;およびb)配列番号5、配列番号7、配列番号9、配列 番号11および配列番号13からなる群から選択される配列 に基づく縮重プライマーを包む、オリゴヌクレオチドプライマーの組み合わせ、 または、以下の配列:c)配列番号4;ならびにd)配列番号6、配列番号8、配列 番号10、配列番号12および配列番号14からなる群から選択される配列を有 するプライマーを含むオリゴヌクレオチドの組み合わせを用いたポリメラーゼ連 鎖反応によって、チロシンキナーゼをコードする核酸配列を増幅する工程;なら びに; (ii)適切なベクターに該増幅核酸をクローニングする工程、 を包含する方法。 21.請求項20に記載の方法によりクローン化された実質的に精製された核 酸。 22.請求項21に記載の核酸分子を保有する細胞。 23.請求項21に記載の核酸分子を保有する微生物。 24.ATCC寄託番号75498を有する微生物。
