Claims (1)
- 【特許請求の範囲】 1.トランスジェニック高等植物の予め選ばれた貯蔵器官における未変性着色カ ロテノイドの蓄積を、双方の植物を同じ条件下に生育した場合に同じ基準の非形 質転換植物の前記貯蔵器官における前記未変性着色カロテノイドの蓄積に比べて 増大させる方法であって、前記形質転換植物を前記予め選ばれた貯蔵器官の成熟 度まで生育し、前記トランスジェニック植物のゲノムが(i)キメラポリペプチ ド結合体をコードするDNAセグメントを(ii)前記トランスジェニック植物 の予め選ばれた貯蔵器官における前記キメラポリペプチド結合体の貯蔵器官増強 発現を引き起こすプロモーターDNAセグメントに作用上結合したものを含み、 前記キメラポリペプチド結合体が、C末端が非高等植物フィトエンシンターゼ酵 素のN末端にペプチド結合されるN末端色素体シグナルペプチド部分を有する工 程を含む方法。 2.前記予め選ばれた貯蔵器官が、根、種子、塊茎及び果実からなる群より選ば れる請求項1記載の方法。 3.前記植物が、ジャガイモ、トマト、ニンジン、メロン、カボチャ、レッドグ アバ、パッションフルーツ、マンゴ、レッドパパイヤ、アボカド、チェリー、タ ンジェリン、マンダリン、パーム、キュウリ、アプリコット、ピーチ及びトウモ ロコシからなる群より選ばれる請求項1記載の方法。 4.前記色素体シグナルペプチド部分が、タバコRUBISCO、ペチュニアE PSPシンターゼ及びコショウPSY遺伝子シグナルペプチドからなる群より選 ばれる請求項1記載の方法。 5.前記プロモーターが根又は塊茎増強プロモーターであり、前記予め選ばれた 貯蔵器官が各々根又は塊茎である請求項1記載の方法。 6.前記根増強プロモーターが、mas、アブラナエクステンシン、トウモロコ シメタロチオネン様タンパク質、ASF−2結合部位含有rbcS−3A及びC aMV35Sプロモーターの塩基対−90〜+8からなる群より選ばれる請求項 5記載の方法。 7.前記塊茎増強プロモーターがパパチンプロモーターである請求項5記載の方 法。 8.前記キメラポリペプチド結合体が、エルウィニア・ハービコーラのcrtB 遺伝子によってコードされるフィトエンシンターゼ酵素を含む請求項1記載の方 法。 9.トランスジェニック植物の予め選ばれた貯蔵器官における未変性着色カロテ ノイドの蓄積を、双方の植物を同じ条件下に生育した場合に同じ基準の植物の前 記貯蔵器官における前記未変性着色カロテノイドの蓄積に相対して増大させる方 法であって、 (a)トランスジェニック植物組織をトランスジェニック植物に再生し、前 記トランスジェニック植物組織が(i)キメラポリペプチド結合体をコードする DNAセグメント及び(ii)前記トランスジェニック植物組織から再生された トランスジェニック植物の前記予め選ばれた貯蔵器官における前記キメラポリペ プチド結合体の貯蔵器官増強発現を引き起こすプロモーターDNAセグメントに 作用上結合したゲノム的組込み発現ベクターを含む組換えDNA分子で形質転換 される植物組織であり、前記キメラポリペプチド結合体が、C末端が非高等植物 フィトエンシンターゼ酵素のN末端にペプチド結合されるN末端色素体シグナル ペプチド部分を有する工程;及び (b)該トランスジェニック植物を前記貯蔵器官の成熟度まで生育する工程 : を含む方法。 10.前記貯蔵器官が、根、種子、塊茎及び果実からなる群より選ばれる請求項9 記載の方法。 11.前記植物が、ジャガイモ、トマト、ニンジン、メロン、カボチャ、レッドグ アバ、パッションフルーツ、マンゴ、レッドパパイヤ、アボカド、チェリー、タ ンジェリン、マンダリン、パーム、キュウリ、アプリコット、ピーチ及びトウモ ロコシからなる群より選ばれる請求項9記載の方法。 12.前記色素体シグナルペプチド部分が、タバコRUBISCO、ペチュニアE PSPシンターゼ及びコショウPSY遺伝子シグナルペプチドからなる群より選 ばれる請求項9記載の方法。 13.前記プロモーターが根又は塊茎増強プロモーターであり、前記予め選ばれた 貯蔵器官が各々根又は塊茎である請求項9記載の方法。 14.前記根増強プロモーターが、mas、アブラナエクステンシン、トウモロコ シメタロチオネン様タンパク質、ASF−2結合部位含有rbcS−3A及びC aMV35Sプロモーターの塩基対−90〜+8からなる群より選ばれる請求項 13記載の方法。 15.前記塊茎増強プロモーターがパパチンプロモーターである請求項13記載の方 法。 16.前記キメラポリペプチド結合体が、エルウィニア・ハービコーラのcrtB 遺伝子によってコードされるフィトエンシンターゼ酵素を含む請求項9記載の方 法。 17.トランスジェニック植物の予め選ばれた貯蔵器官における未変性着色カロテ ノイドの蓄積を、双方の植物を同じ条件下に生育した場合に同じ基準の非形質転 換植物の前記貯蔵器官における前記未変性着色カロテノイドの蓄積に比べて増大 させる方法であって、 (a)予め選ばれた貯蔵器官において着色カロテノイドを蓄積する植物の組 織を(i)キメラポリペプチド結合体をコードするDNAセグメント及び(ii )前記トランスジェニック植物組織から再生されたトランスジェニック植物の前 記予め選ばれた貯蔵器官における前記キメラポリペプチド結合体の貯蔵器官増強 発現を引き起こすプロモーターDNAセグメントに作用上結合した組込みベクタ ーを含む組換えDNA分子でゲノム的に形質転換することによりトランスジェニ ック植物組織をつくり、前記キメラポリペプチド結合体が、C末端が非高等植物 フィトエンシンターゼ酵素のN末端にペプチド結合されるN末端色素体シグナル ペプチド部分を有する工程; (b)前記トランスジェニック植物組織を、貯蔵器官を有する前記トランス ジェニック植物に再生させる工程;及び (c)前記トランスジェニック植物を前記予め選ばれた貯蔵器官の成熟度ま で生育する工程: を含む方法。 18.前記貯蔵器官が、根、種子、塊茎及び果実からなる群より選ばれる請求項17 記載の方法。 19.前記着色カロテノイドがカロテン又はキサントフィルである請求項17記載の 方法。 20.前記植物組織が、ジャガイモ、トマト、ニンジン、メロン、カボチャ、レッ ドグアバ、パッションフルーツ、マンゴ、レッドパパイヤ、アボカド、チェリー 、タンジェリン、マンダリン、パーム、キュウリ、アプリコット、ピーチ及びト ウモロコシからなる群より選ばれる植物から得られる請求項17記載の方法。 21.前記色素体シグナルペプチド部分が、タバコRUBISCO、ペチュニアE PSPシンターゼ及びコショウPSY遺伝子シグナルペプチドからなる群より選 ばれる請求項17記載の方法。 22.前記プロモーターが根又は塊茎増強プロモーターであり、前記予め選ばれた 貯蔵器官が各々根又は塊茎である請求項17記載の方法。 23.前記根増強プロモーターが、mas、アブラナエクステンシン、トウモロコ シメタロチオネン様タンパク質、ASF−2結合部位含有rbcS−3A及びC aMV35Sプロモーターの塩基対−90〜+8からなる群より選ばれる請求項 22記載の方法。 24.前記塊茎増強プロモーターがパパチンプロモーターである請求項22記載の方 法。 25.前記キメラポリペプチド結合体が、エルウィニア・ハービコーラのcrtB 遺伝子によってコードされるフィトエンシンターゼ酵素を含む請求項17記載の方 法。 26.非形質転換植物のように根においてカロテン又はキサントフィルを蓄積する 形質転換ニンジンの根においてカロテン又はキサントフィルの蓄積を増大させる 方法であって、 (a)根においてカロテン又はキサントフィルを蓄積するニンジンの組織を (i)キメラポリペプチド結合体をコードするDNAセグメント及び(ii)前 記トランスジェニックニンジン組織から再生されたトランスジェニックニンジン 植物の根における前記キメラポリペプチド結合体の根増強発現を引き起こすma sプロモーターDNAセグメントに作用上結合した組込みベクターを含む組換え DNA分子でゲノム的に形質転換することによりトランスジェニック ニンジン組織をつくり、前記キメラポリペプチド結合体が、C末端がエルウィニ ア・ハービコーラ フィトエンシンターゼ酵素のN末端にペプチド結合されるN末 端RUBISCOシグナルペプチド部分を有する工程; (b)前記トランスジェニック植物組織を、根を有するトランスジェニック ニンジン植物に再生させる工程;及び (c)前記トランスジェニックニンジン植物を該根の成熟度まで生育する工 程: を含む方法。 27.(a)キメラポリペプチド結合体をコードするゲノム構造遺伝子を有しかつ (b)同じ基準の非トランスジェニック植物の前記貯蔵器官における未変性着色 カロテノイドの蓄積に相対して予め選ばれた貯蔵器官における未変性着色カロテ ノイドを過剰蓄積し、前記キメラポリペプチド結合体が、C末端が非高等植物フ ィトエンシンターゼ酵素のN末端に結合されるN末端色素体シグナルペプチド部 分を有するトランスジェニック植物。 28.前記貯蔵器官が、根、種子、塊茎及び果実からなる群より選ばれる請求項27 記載のトランスジェニック植物。 29.ニンジンである請求項28記載のトランスジェニック植物。 30.(a)キメラポリペプチド結合体をコードするゲノム構造遺伝子を有しかつ (b)同じ基準の非トランスジェニック植物の前記貯蔵器官における蓄積に相対 して根、種子、塊茎及び果実からなる群より選ばれる予め選ばれた貯蔵器官にお ける未変性着色カロテノイドを過剰蓄積し、前記キメラポリペプチド結合体が、 C末端が非高等植物フィトエンシンターゼ酵素のN末端に結合されるN末端色素 体シグナルペプチド部分を有するトランスジェニック植物。 31.前記色素体シグナルペプチド部分がRUBISCOシグナルペプチドである 請求項30記載のトランスジェニック植物。 32.フィトエンシンターゼ酵素を示す前記非高等植物がエルウィニア・ハービコ ーラ のcrtB遺伝子でコードされる請求項30記載のトランスジェニック植物。 33.前記非トランスジェニック植物が、ジャガイモ、トマト、ニンジン、メロン 、カボチャ、レッドグアバ、パッションフルーツ、マンゴ、レッドパパイヤ、ア ボカド、チェリー、タンジェリン、マンダリン、パーム、キュウリ、アプリコッ ト、ピーチ及びトウモロコシからなる群より選ばれる請求項30記載のトランスジ ェニック植物。 34.前記着色カロテノイドが、カロテン及びキサントフィルからなる群より選ば れる請求項30記載のトランスジェニック植物。 35.(a)キメラポリペプチド結合体をコードするゲノム構造遺伝子を有しかつ (b)同じ基準の非トランスジェニックニンジンの蓄積に相対して根におけるカ ロテン又はキサントフィルを過剰蓄積し、前記キメラポリペプチド結合体が、C 末端がエルウィニア・ハービコーラフィトエンシンターゼ酵素のN末端に結合さ れるN末端RUBISCOシグナルペプチド部分を有するトランスジェニック植 物。 36.同じ基準の非トランスジェニック植物に相対して未変性着色カロテノイドを 過剰蓄積するトランスジェニック植物又はそれに由来するハイブリッドに発芽さ せることができ、種子から発育させた植物の予め選ばれた貯蔵器官におけるキメ ラポリペプチド結合体の貯蔵器官増強発現を引き起こすプロモーターに作用上結 合されるキメラポリペプチド結合体をコードするゲノム遺伝子を含み、前記キメ ラポリペプチド結合体が、C末端が非高等植物フィトエンシンターゼ酵素のN末 端に結合されるN末端色素体シグナルペプチド部分を有するトランスジェニック 植物種子。 37.前記未変性着色カロテノイドが、カロテン及びキサントフィルからなる群よ り選ばれる請求項34(?)記載のトランスジェニック植物種子。 38.同じ基準の非トランスジェニック植物に相対してβ−カロテンを過剰蓄積す るトランスジェニック植物又はそれに由来するハイブリッドに発芽させることが できるトランスジェニック植物種子。 39.ニンジン種子である請求項38記載のトランスジェニック植物種子。
