Claims (1)
- 【特許請求の範囲】 1.微視的物質、より詳しくは生物学的物質、細胞結合体、単細胞または細胞 構成要素を基板表面上で凍結保存および低温処理する方法であって、 特徴として、包被溶媒を含む少なくとも1個の貯蔵容器中に上記物質を導入す る段階と、 少なくとも1個の微小液滴配置手段によって、温度T1を有する上記基板表面 を形成している温度制御された基板ターゲットに上記少なくとも1個の貯蔵容器 から時間的、位置的に制御された状態で包被された物質を案内する段階と、 ターゲットの温度を貯蔵温度T2あるいは処理温度T3に調整しながら、上記 基板ターゲット上に上記物質を固定的に接着させる段階と、 上記のように処理された基板ターゲットを貯蔵および/または処理する段階と 、からなる上記冷凍保存および低温処理する方法。 2.上記物質の案内は微小液滴噴出装置によって行われ、また上記ターゲット に対する微小液滴の動きを1回の発射毎に移動させ、および/または上記微小液 滴噴出装置に対する上記ターゲットの位置を移動させることによって上記基板表 面に予め設定された間隔もしくはパターンで上記物質を接着させることを特徴と する請求の範囲1に記載の方法。 3.1またはそれ以上のターゲット領域に複数の微小液滴噴出装置によって同 時にまたは順次に物質が噴射されることを特徴とする請求の範囲1または2に記 載の方法。 4.上記温度T1が水の氷点以上であり、温度T2とT3が−2℃乃至−27 3℃の範囲内であることを特徴とする請求の範囲1乃至3の何れかに記載の方法 。 5.微小液滴噴出装置による装填処理に先立って、上記基板ターゲットの表面 は部分的にあるいは全面に亘ってゲル、分子層、液体あるいは高分子、あるいは それらの複数または組合わせで被覆されることを特徴とする請求の範囲4に記載 の方法。 6.上記の温度T1、T2およびT3が−5℃乃至−273℃の範囲内である ことを特徴とする請求の範囲1乃至3の何れかに記載の方法。 7.上記基板ターゲットの表面に向かって飛翔中の上記物質と微小液滴は光学 的あるいは電気的に感知され、上記物質を伴った上記微小液滴の衝突位置は上記 感知によって得られたデータに関して可変であることを特徴とする請求の範囲1 乃至6の何れかに記載の方法。 8.上記装填処理された基板ターゲットの貯蔵および/または処理が液体窒素 または類似の低温液体またはそれのガス相のもとで行われることを特徴とする請 求の範囲1乃至7の何れかに記載の方法。 9.全体の方法あるいはその方法の個々の作業ステップは常圧とは異なる力の 下で実行されることを特徴とする請求の範囲1乃至8の何れかに記載の方法。 10.貯蔵および/または処理に先立って、上記処理された基板ターゲットは さらに薄い層で被覆され、この薄い層は微小液滴噴出装置によって供給された凍 結した溶液で形成され、あるいは例えば噴霧、ガス相におけるスパッタリングあ るいは真空蒸着すること、によって生成される金属層とすることができる薄い層 であることを特徴とする請求の範囲1乃至9の何れかに記載の方法。 11.低温状態における上記処理段階において、物質は除去され、および/ま たは細胞物質は光学的切除手段、凍結エッチング(freeze etchin g)に類似した方法、あるいは真空下における蒸発によって物理的に切除される ことを特徴とする請求の範囲1乃至10の何れかに記載の方法。 12.低温状態において、合成、遺伝子あるいは細胞源であり、ある物質が供 給および/または置換され、あるいは解凍中または解凍後に開口細胞をその生命 を保ちつつ閉鎖する物質が供給されることを特徴とする請求の範囲11に記載の 方法。 13.上記物質が適応した状態の下で凍結乾燥されることを特徴とする請求の 範囲1乃至12の何れかに記載の方法。 14.細胞を有するまたは細胞を有しない、同じまたは異なった組成の数滴の 微小液滴が互いに低温的に固定されるか並置されることを特徴とする請求の範囲 1乃至13の何れかに記載の方法。 15.上記基板表面に形成された1つの層が上記細胞またはその固定状態にあ る部分を含んでいる上記微小液滴を切除することを特徴とする請求の範囲1乃至 14の何れかに記載の方法。 16.適当な位置で凍結される前および/または上記包被溶液の凍結点より高 い温度で上記ターゲットを加熱することによる解凍の後、上記細胞は電場の力に よって操作され、または上記包被溶液中に浮遊した状態で保持されることを特徴 とする請求の範囲1乃至15の何れかに記載の方法。 17.基板表面上の微視的物質、より詳しくは、生物学的物質、細胞形成物、 単細胞あるいは細胞構成要素を凍結保存および低温で処理する装置であって、特 徴として、時間的、位置的に制御された状態で包被された生物学的物質を含む包 被溶液の液滴を上記基板表面を形成する温度制御された基板上に噴出する微小液 滴噴出装置を有することを特徴とする冷凍保存と低温で処理するための装置。 18.上記装置は上記供給された物質を貯蔵および/または操作する手段を含 んでいることを特徴とする請求の範囲17に記載の装置。 19.上記基板ターゲットは、より詳しくは微小電極、電子的感知素子あるい は電子的感知成分、窪み、突出部、貫通孔あるいは溝、あるいは波状、柱状、突 起、土塁状の微細構造を含む表面構造を有し、これらは上記表面の大きさを一般 にマイクロメートルまたはサブマイクロメートルの範囲に拡大していることを特 徴とする請求の範囲17乃至18に記載の装置。 20.上記微小液滴噴出装置および/または半導体材料、特にシリコンを基礎 とする半導体材料からなる上記基板ターゲットを有することを特徴とする請求の 範囲17乃至19の何れかに記載の装置。 21.噴射期間中、上記基板ターゲットは水の氷点以上の温度T1に保たれて おり、噴射後は冷却され、且つ−5℃乃至−273℃の範囲内に温度T2に温度 制御され、上記基板ターゲットは時間的に指定された温度プログラムに従って冷 却および/または加熱可能であり、例えば、より具体的には液体窒素を含む冷却 槽で、または上記冷却槽のガス雰囲気で冷却されることを特徴とする請求の範囲 17乃至20の何れかに記載の装置。 22.上記処理手段が、ミクロトームカッター、微小ドリル(microdr illing)や研磨装置(フライス盤)(milling machine) 、レーザービーム、あるいは他の素子からなり、これらの処理手段によって低温 状態にある上記物質を機械的、熱的または光学的に除去または切除することを特 徴とする請求の範囲17乃至21の何れかに記載の装置。 23.上記基板温度と微小液滴の温度が互いに独立して調整され、これらの温 度は噴射される微小液滴用の液滴溶液の凍結点以上よりもはるかに高いことを特 徴とする請求の範囲17乃至22の何れかに記載の装置。 24.医学的、生物学的、製薬的あるいは他の目的に使用することができる細 胞バンク(bank)のために細胞を凍結保存し、あるいは医学的診断学、環境 工学および薬剤研究における試験チップ、センサー、スクリーニング素子として 基板を生産するための請求の範囲1乃至23の何れかに記載の方法あるいは装置 の使用法。
