Claims (1)
- 【特許請求の範囲】 1.C-11位に結合した担体を有する、式: で示される化合物のキニジン−担体免疫原性抱合体。 2.式: [式中、x=1または2] で示されるキニジン誘導体およびその塩。 3.キニジン誘導体が、式: [式中、x=1または2] を有する、請求項1に記載のキニジン−誘導体担体免疫原性抱合体。 4.担体が、ポリ(アミノ酸)、タンパク質、炭水化物、ラテックス粒子、お よび糖タンパク質から成る群から選択される、請求項1に記載のキニジン−担体 免疫原性抱合体。 5.担体が、キーホール・リムペット・ヘモシアニン(KLH)、γ−グロブリ ン、およびカボチャ種子グロブリン(PSG)から成る群から選択される、請求項 1に記載のキニジン−担体免疫原性抱合体。 6.請求項1に記載の免疫原性抱合体に対して製造される抗キニジン抗体。 7.キニジン誘導体が、キニジン分子のC-11位においてリンカーと結合してい る、キニジン−リンカー−リポーター免疫原性抱合体。 8.-NH(CH2)mO (CH2CH2O)n (CH2)mNH- [式中、m=2または3、およびn=1〜10] から成るリンカーを介して、キニジンがリポーターに結合している請求項7に記 載の抱合体。 9.n=1または2である請求項8に記載の抱合体。 10.キニジン誘導体が、式: [式中、x=1または2] で示される請求項7に記載の抱合体。 11.リポーターが、ラテッスク粒子、発蛍光団、化学発光物質、発色団、放 射性物質および酵素から成る群から選択される請求項7に記載の抱合体。 12.リポーターがラテックス粒子である請求項7に記載の抱合体。 13.構造式:Pr-NH (CH2)mO (CH2CH2O) n(CH2) mNH2[式中、Prはアミン保 護基、m=2または3、n=1-10] を有するモノ保護親水ジアミン試薬。 14.n=1または2である請求項13に記載のモノ保護親水ジアミン試薬。 15.アミン保護基がtert−ブトキシカルボニル(t-Boc)である請求項13 に記載のモノ保護親水ジアミン試薬。 16.下記段階: (a)水性/有機媒体中、水酸化ナトリウムの存在下に、ジ−tert−ブチルジ カーボネートと過剰親水ジアミンとを反応させる; (b)媒体の有機溶媒を、媒体の水性相から分離する; (c)水性相のpHを下げることによって、本質的に全ての残留アミン基にプ ロトン付加する; (d)水不混和性有機溶媒を水性相に加えて、段階(c)の生成物から非塩基 性物質を除去する; (e)塩基を加えて水性相のpHを上げて、残留アミン基の少なくとも半分が非 プロトン付加であるようにする; (f)段階(e)の水性相に塩を加える; (g)水性相を水不混和性有機溶媒で抽出する; (h)有機相を水性相から分離する;および (i)有機相から溶媒を除去して、所望の生成物を得る; を含んで成る、請求項13に記載のモノ−t-Boc保護親水ジアミン試薬の合成方 法。 17.下記段階: (a)構造式:Pr-NH (CH2)mO (CH2CH2O) n (CH2) mN H2 [式中、Prはtert−ブトキシカルボニル] を有するモノ保護親水ジアミンリポーター試薬、試験サンプル、および凝集剤を 混合し;続いて、 (b)既知の凝集イムノアッセイフォーマットを実施する; を含んで成る、試験サンプル中のキニジンの存在および/または量を検査するイ ムノアッセイ。
