Claims (1)
- 【特許請求の範囲】 1.a)エフェクタードメイン、 b)第一のタンパク質の結合ドメインに近接して配置された第一の共優性の細胞 内シグナル部分、および c)第二のタンパク質の結合ドメインに近接して配置された第二の共優性の細胞 内シグナル部分 を含む、共優性で媒介される毒素。 2.第一および第二の細胞内シグナル部分が分解シグナルである、請求の範囲1 の共優性で媒介される毒素。 3.第一および第二の細胞内シグナル部分がトランスロケーションシグナルであ る、請求の範囲1の共優性で媒介される毒素。 4.トランスロケーションシグナルが核局在化シグナルである、請求の範囲3の 共優性で媒介される毒素。 5.第一の細胞内シグナル部分が分解シグナルでありかつ第二の細胞内シグナル 部分が核局在化シグナルである、請求の範囲1の共優性で媒介される毒素。 6.a)エフェクタードメイン、 b)第一のタンパク質の結合ドメインに近接して配置された第一の共優性の分解 シグナル、および c)第二のタンパク質の結合ドメインに近接して配置された第二の共優性の分解 シグナル を含む、細胞内分解シグナル依存性でリガンドで調節される毒素。 7.a)エフェクタードメイン、 b)第一のタンパク質の結合ドメインに近接して配置された第一の共優 性のトランスロケーションシグナル、および c)第二のタンパク質の結合ドメインに近接して配置された第二の共優性のトラ ンスロケーションシグナル を含む、細胞内分トランスロケーションシグナル依存性でリガンドで調節される 毒素。 8.第一および第二のトランスロケーションシグナルが核局在化シグナルである 、請求の範囲7の細胞内トランスロケーションシグナル依存性でリガンドで調節 される毒素。 9.エフェクタードメインがサイトゾル特異性である、請求の範囲8の細胞内ト ランスロケーションシグナル依存性でリガンドで調節される毒素。 10.エフェクタードメインが核特異性である、請求の範囲8の細胞内トランス ロケーションシグナル依存性でリガンドで調節される毒素。 11.a)エフェクタードメイン、 b)細胞表面結合ドメイン、 c)サイトゾル貫通ドメイン、 d)第一のタンパク質の結合ドメインに近接して配置された第一の共優性の細胞 内シグナル部分、および e)第二のタンパク質の結合ドメインに近接して配置された第二の共優性の細胞 内シグナル部分 を含む、共優性で媒介される毒素。 12.第一および第二の細胞内シグナル部分が分解シグナルである、請求の範囲 11の共優性で媒介される毒素。 13.第一および第二の細胞内シグナル部分がトランスロケーションシ グナルである、請求の範囲11の共優性で媒介される毒素。 14.トランスロケーションシグナルが核局在化シグナルである、請求の範囲1 3の共優性で媒介される毒素。 15.エフェクタードメインがサイトゾル特異性である、請求の範囲14の共優 性で媒介される毒素。 16.エフェクタードメインが核特異性である、請求の範囲14の共優性で媒介 される毒素。 17.第一の細胞内シグナル部分が分解シグナルでありかつ第二の細胞内シグナ ル部分が核局在化シグナルである、請求の範囲11の共優性で媒介される毒素。 18.第一のタンパク質および第二のタンパク質の双方を含有することが既知の 細胞の分裂を選択的に鎮めるもしくは排除する方法であって、その方法が a)i)エフェクタードメイン、 ii)第一のタンパク質の結合ドメインに近接して配置される第一の共優性の分解 シグナル、および iii)第二のタンパク質の結合ドメインに近接して配置される第二の共優性の分 解シグナル を含む、細胞内分解シグナル依存性でリガンドで調節される毒素をコードするD NA発現構築物を提供すること、ならびに b)段階a)からのDNA構築物を、細胞内分解シグナル依存性でリガンドで調 節される毒素の発現に適切な条件下に殺されるべき細胞内に導入すること、 を含む方法。 19.第一のタンパク質および第二のタンパク質を含有することが既知の細胞が 原核細胞である、請求の範囲18の方法。 20.第一のタンパク質および第二のタンパク質を含有することが既知の細胞が 真核細胞である、請求の範囲18の方法。 21.第一のタンパク質および第二のタンパク質の双方をサイトゾルに含有する ことが既知の真核細胞の分裂を選択的に鎮めるもしくは排除する方法であって、 その方法が a)i)サイトゾル特異的なエフェクタードメイン、 ii)第一のタンパク質の結合ドメインに近接して配置された第一の共優性の核局 在化シグナル、および iii)第二のタンパク質の結合ドメインに近接して配置された第二の共優性の核 局在化シグナル を含む、細胞内トランスロケーションシグナル依存性でリガンドで調節される毒 素をコードするDNA発現構築物を提供すること、ならびに b)段階a)からのDNA構築物を、細胞内デグロン依存性でリガンドで調節さ れる毒素の発現に適切な条件下に殺されるべき細胞内に導入すること、 を含む方法。 22.第一のタンパク質および第二のタンパク質の双方をサイトゾルで欠くする ことが既知の真核細胞の分裂を選択的に鎮めるもしくは排除する方法であって、 その方法が a)i)核特異的なエフェクタードメイン、 ii)第一のタンパク質の結合ドメインに近接して配置された第一の共優性の核局 在化シグナル、および iii)第二のタンパク質の結合ドメインに近接して配置された第二の共優性の核 局在化シグナル を含む、細胞内トランスロケーションシグナル依存性でリガンドで調節される毒 素をコードするDNA発現構築物を提供すること、ならびに b)段階a)からのDNA構築物を、細胞内トランスロケーションシグナル依存 性でリガンドで調節される毒素の発現に適切な条件下に細胞内に導入すること、 を含む方法。 23.第一のタンパク質および第二のタンパク質の双方を含有することが既知の 真核細胞の分裂を選択的に鎮めるもしくは排除する方法であって、その方法が a)i)エフェクタードメイン、 ii)細胞表面結合ドメイン、 iii)サイトゾル貫通ドメイン、 iv)第一のタンパク質の結合ドメインに近接して配置された第一の共優性の細胞 内シグナル部分、および v)第二のタンパク質の結合ドメインに近接して配置された第二の共優性の細胞 内シグナル部分 を含む、多ドメインタンパク質融合物を提供すること、ならびに、 b)段階a)の多ドメイン融合物を標的細胞に接触させること、 を含む方法。 24.第一および第二の細胞内シグナル部分が分解シグナルである、請求の範囲 23の方法。 25.第一および第二の細胞内シグナル部分がトランスロケーションシ グナルである、請求の範囲23の方法。 26.トランスロケーションシグナルが核局在化シグナルである、請求の範囲2 3の共優性で媒介される毒素。 27.第一の細胞内シグナル部分が分解シグナルでありかつ第二の細胞内シグナ ル部分が核局在化シグナルである、請求の範囲23の方法 28.1種の予め決められた細胞内リガンドに特異的な、条件付きの、細胞内リ ガンド依存性の毒性を表す融合タンパク質であって、その融合タンパク質が a)エフェクタードメイン、および b)タンパク質の結合ドメインに近接して配置される細胞内シグナル部分、 を含むタンパク質。 29.1種の予め決められた細胞内リガンドに特異的な、条件付きの、細胞内リ ガンド依存性の毒性を表す融合タンパク質であって、その融合タンパク質が a)エフェクタードメイン、 b)細胞表面結合ドメイン、 c)サイトゾル貫通ドメイン、および d)タンパク質の結合ドメインに近接して配置される細胞内シグナル部分、 を含むタンパク質。
