Claims (1)
- 【特許請求の範囲】 1.DNAプロウイルスの形状であるときに宿主細胞ゲノム中に挿入される ことができ、DNAプロウイルス中に(a)調節因子によって誘導可能である調 節されたプロモーターと、(b)(a)におけるプロモーターの転写制御下にあ る選択された遺伝子(単数又は複数)とを与える配列を含むパッケージ可能なR NAゲノムを含む、第1レトロウイルスに基づくレトロウイルスベクターであっ て、第1レトロウイルスの5’LTRプロモーター機能の代わりに、調節された プロモーター(a)がプロウイルスの5’末端反復配列(LTR)中に存在し、 選択された遺伝子(単数又は複数)(b)がLTRの間に配置されている上記レ トロウイルスベクター。 2.調節されたプロモーター(a)が第2レトロウイルスから調節タンパク 質によって誘導可能である、請求項1記載のレトロウイルスベクター。 3.選択された遺伝子が第1治療遺伝子である、請求項1又は請求項2に記 載のレトロウイルスベクター。 4.調節されたプロモーターが第2レトロウイルスのU5とR、又はそれら の機能部分を含む、請求項2又は請求項3に記載のレトロウイルスベクター。 5.第1レトロウイルスがMLVである、請求項1〜4のいずれかに記載の レトロウイルスベクター。 6.第2レトロウイルスがHIVである、請求項2〜5のいずれかに記載の レトロウイルスベクター。 7.構成プロモーターと、第2治療遺伝子又は選択可能なマーカー遺伝子と の発現カセットをさらに含む、請求項1〜6のいずれかに記載のレトロウイルス ベクター。 8.同じ又は異なる転写単位上に存在する複数個の治療遺伝子を含む、請求 項1〜7のいずれかに記載のレトロウイルスベクター。 9.プロモーターの制御下にある、請求項1〜8のいずれかに記載のレトロ ウイルスベクターに関してパッケージ可能なRNAゲノムをコードするDNA構 築体。 10.プロモーターが第1又は第2レトロウイルス以外のソースに由来する 、請求項9記載のDNA構築体。 11.プロモーターがCMVプロモーターである、請求項9又は請求項10 記載のDNA構築体。 12.選択された遺伝子が存在せず、構築体が選択された遺伝子(単数又は 複数)が挿入されうる挿入部位を有する、請求項9〜11のいずれかに記載のD NA構築体。 13.請求項9〜12のいずれかに記載のDNA構築体によってトランスフ ェクトされたパッケージング細胞ラインを含み、請求項1〜8のいずれかに記載 にレトロウイルスベクターを産生することができるレトロウイルスベクター産生 系。 14.ターゲット細胞の感染又は形質導入の遺伝子療法のための、請求項2 〜8のいずれかに記載のレトロウイルスベクターの使用。 15.請求項14記載の方法から生じるターゲット細胞。
