Claims (1)
- 【特許請求の範囲】 1. 配列番号51および該配列の相補配列であるオリゴヌクレオチド。 2. 配列番号53および該配列の相補配列であるオリゴヌクレオチド。 3. 配列番号54および該配列の相補配列であるオリゴヌクレオチド。 4. 配列番号55および該配列の相補配列であるオリゴヌクレオチド。 5. 配列番号56および該配列の相補配列であるオリゴヌクレオチド。 6. 配列番号57および該配列の相補配列であるオリゴヌクレオチド。 7. 配列番号87および該配列の相補配列であるオリゴヌクレオチド。 8. 被験サンプル中の標的HGBV−Cヌクレオチドを検出する方法であって 、 a.1以上のHGBV−C特異的オリゴヌクレオチドもしく は該ヌクレオチドの相補配列と標的HGBVヌクレオチドとを接触させる段階、 ならびに b.被験サンプル中における標的の有無を検出する段階 を有してなる方法。 9. 前記オリゴヌクレオチドが、配列番号51、配列番号53、配列番号54 、配列番号55、配列番号56、配列番号57および配列番号87ならびにこれ ら配列の相補配列からなる群から選択される請求項8に記載の方法。 10. 前記オリゴヌクレオチドが、配列番号2、配列番号3、配列番号4、配 列番号5、配列番号6、配列番号9、配列番号10、配列番号11、配列番号1 3、配列番号13、配列番号14、配列番号15、配列番号16、配列番号17 、配列番号18、配列番号19、配列番号20、配列番号21、配列番号22、 配列番号23、配列番号24、配列番号60、配列番号61、配列番号62、配 列番号63、配列番号64、配列番号65、配列番号66、配列番号67、配列 番号68、配列番号69、配列番号70、配列番号71、配列番号72、配列番 号73、配列番号74、配列番号75、配列番号76、配列番号77、配列番号 78、配列番号79、配列番号80、配列番号 81、配列番号82、配列番号83、配列番号84、配列番号85および配列番 号86ならびにそれら配列の相補配列からなる群から選択される請求項8に記載 の方法。 11. 被験サンプル中の標的HGBV−Cヌクレオチドを検出する方法であっ て、 a.配列番号51、配列番号53、配列番号54、配列番号55、配列番号5 6、配列番号57および配列番号87ならびにこれら配列の相補配列から成る群 から選択される1以上のオリゴヌクレオチドと標的HGBVヌクレオチドとを接 触させる段階、ならびに b.被験サンプル中における標的の有無を検出する段階 を有してなる方法。 12. 段階(a)を行う前に、前記標的HGBV−Cヌクレオチドを固相に付 着させる請求項8に記載の方法。 13. 被験サンプル中のGB型肝炎ウィルスの5’末端cDNAを増幅する方 法であって、 a.ランダムプライマーを用いた逆転写を行う段階、 b.第1段階のPCRにおけるセンスおよびアンチセンスプライマーとしてH GBV−Cの他のオリゴヌクレオチドプライ マーを用いることで、段階(a)から得られたcDNAを増幅して、増幅cDN Aを得る段階、 c.被験サンプル中のHGBV−Cのアンプリコンの有無を検出する段階 を有してなる方法。 14. 前記段階(b)の増幅をポリメラーゼ連鎖反応によって行う請求項13 に記載の方法。 15. 段階(a)を行う前に、前記被験サンプルを固相に付着させる請求項1 3に記載の方法。 16. 前記検出段階が、測定可能信号を発生させることができる検出可能標識 を利用する段階を有する請求項13に記載の方法。 17. 前記検出可能標識を固相に付着させる請求項16に記載の方法。 18. 被験サンプル中のGB型肝炎ウィルスの5’末端cDNAを増幅する方 法であって、 a.ランダムプライマーを用いた逆転写を行う段階、 b.第1段階のPCRにおけるセンスおよびアンチセンスプライマーとして他 のオリゴヌクレオチドプライマーを用いるこ とで、段階(a)から得られたcDNAを増幅して増幅cDNAを得る段階であ って、前記センスプライマーが配列番号51、53および56からなる群から選 択され、前記アンチセンスプライマーが配列番号54、55、57および87か らなる群から選択される段階、 c.被験サンプル中のアンプリコンの有無を検出する段階 を有してなる方法。 19. 前記段階(b)の増幅をポリメラーゼ連鎖反応によって行う請求項18 に記載の方法。 20. 段階(a)を行う前に、前記被験サンプルを固相に付着させる請求項1 8に記載の方法。 21. 前記検出段階が、測定可能信号を発生させることができる検出可能標識 を利用する段階を有する請求項18に記載の方法。 22. 前記検出可能標識を固相に付着させる請求項18に記載の方法。 23. 標的HGBVを有することが疑われる被験サンプルにおけるHGBV− Cを検出する方法であって、 a.センスプライマーとしての配列番号51、配列番号56 および配列番号53からなる群から選択される1以上のオリゴヌクレオチドなら びにアンチセンスプライマーとしての配列番号54、配列番号55、配列番号5 7および配列番号87からなる群から選択される1以上のオリゴヌクレオチドと 前記被験サンプルを接触させ、同配列を増幅して第1段階の反応生成物を得る段 階、 b.前記段階(a)の1以上のオリゴヌクレオチドと配列番号51、配列番号 53、配列番号54、配列番号55、配列番号56、配列番号57および配列番 号87からなる群から選択される第2のオリゴヌクレオチドとを、前記第1段階 の反応生成物と接触させる段階(ただし、第2のオリゴヌクレオチドは、使用さ れる第1のオリゴヌクレオチドに対して3’の位置にあり、前記第1のオリゴヌ クレオチドに対して反対のセンスのものである)、 c.前記HGBV標的を検出する段階 を有してなる方法。 24. 前記増幅をポリメラーゼ連鎖反応によって行う請求項23に記載の方法 。 25. 段階(a)を行う前に、前記被験サンプルを固相に付 着させる請求項23に記載の方法。 26. 前記検出段階が、測定可能信号を発生させることができる検出可能標識 を利用する段階を有する請求項23に記載の方法。 27. 前記検出可能標識を固相に付着させる請求項26に記載の方法。 28. 配列番号35、配列番号36、配列番号37、配列番号38、配列番号 39、配列番号40、配列番号41、配列番号43、配列番号44、配列番号4 5、配列番号46、配列番号47、配列番号48、配列番号49および配列番号 50ならびにそれら配列の相補配列からなる群から選択されるGAPリガーゼ連 鎖反応に有用なプライマー。 29. 被験サンプル中のHGBV−C標的を検出するのに有用な試験キットで あって、配列番号51、配列番号53、配列番号54、配列番号55、配列番号 56、配列番号57および配列番号87ならびにそれら配列の相補配列からなる 群から選択される1以上のオリゴヌクレオチドの入った容器を有してなる試験キ ット。
