Abstract
PURPOSE: To secure the simple and high-reproducibility measurement for the glycosyl hemoglobin in the blood, by giving the analysis to the pre-processed sample through the chromatography method using the specified developer.
CONSTITUTION: The sodium azide of about 40W1,000ml/l is added to the blood in which the erythrocyte is subjected to hemolysis, and thus the contained glycosyl hemoglobin and the hemoglobin are modified to methemoglobin each to obtain azido methoglycosyl hemoglobin (a) and azido methemoglobin (b) respectively which are used for the sample of the liquid chromatography. In other words, the sodium azide aqueous solution (preferably pH6.6W7.1) of about 400W1,000ml/l is used for the 1st developer of the chromatogrphy in order to elute component (a). Then the 2nd developer (such as the phosphoric acid buffer solution) which contains no sodium azide is used to elute component (b). Then the components isolated and eluted are measured by the spectro-photometer.
