Abstract

PURPOSE: To prepare HBs antigen which is an antigen of the hepatitis B virus, in high purity and with a small-sized apparatus, by the ion exchange chromatographic treatment of blood plasma or serum.
CONSTITUTION: Blood plasma or blood serum is treated with an anion exchange material (e.g. DEAE-Sepharose CL-6B: product of Pharmacea Co.) to remove the plasma components other than the protein components mainly composed of KBs antigen and γ-globulin. The solution containing the plasma proteins is treated with a cation exchange material (e.g. CM-Sepharose CL-6B) to adsorb and remove the protein components mainly composed of γ-globulin. The purified HBs antigen can be separated from the fraction which is not adsorbed to the ion exchange material. The antigen contains extremely small amount of HB viruses.
