Abstract

PURPOSE: To prepare a slow-releasing formulation containing a physiologically- active substance and having controlled eluting rate, by dispersing a physiologically- active substance in the mixed solution of a pH-soluble polymer and a monomer polymerizable at low temperatures to give a formulation and polymerizing the formulation at low temperature with radiation of light, etc..
CONSTITUTION: A polymer or a copolymer soluble at pH of 1W10.0, e.g. Eudragit E, etc., is dissolved in a monomer polymerizable at low temperature, e.g. 2-hydroxyethyl methacrylate, etc., to give a transparent and homogeneous mixed solution. A formulation is provided by dispersing or homogeneously dissolving a physiologically-active substance, e.g. aspirin, KCl, etc., in the above solution, and irradiated with ionizing rays, e.g. light, γ-rays, etc., at ≤0°C for polymerization of the monomer polymerizable at low temperatures to give a slow-releasing formulation having the pH-controlled elution rate of the physiologically-active substance.
EFFECT: Elution of the active substance is suppressed at the pH of the gastric juice, and accelerated at the intestinal juice part. Consequently the elution of the active substance can be controlled and the administration frequency of the active substance can be reduced.
