Abstract
PURPOSE: To obtain a protein particularly suitable for injection free from endotoxin, by subjecting a useful protein containing endotoxin to chromatography, using a metal chelate carrier.
CONSTITUTION: A useful protein containing endotoxin is subjected to chromatography using a with a metal chelate carrier having metallic ions combined complex- forming group, e.g. an insoluble carrier comprising iminodiacetic acid-metallic ions combined with epoxy activated sepharose of the formula, to adsorb the imidazole groups or thiol groups in the side chain of the protein on the metallic ions with coordination bonds. the endotoxin passes through the column without adsorption. The desired protein free from the endotoxin can be obtained in high yield by severing the coordination bonds.
