Abstract

PURPOSE: Vessels containing a culture solution for measuring microorganisms are sealed in vessels, then sterilized with heat under pressure, thus preparing readily and safely the culture solution for measuring microorganisms.
CONSTITUTION: Culture solutions for measuring microorganisms, such as stantard agar culture medium, EC medium for examining colon bacilli, physiological salt solution for the dilution in measurment of the cell numbers of general living bacteria or lactic bacteria, is charged in a prescribed amount into vessels of synthetic resin, metal, or glass or enameled ware, such as bags, dishes, cups, or tubes, and sealed in order to prevent recontamination. Then, they are sterilized by heating under pressure in a sterilizer and cooled. This method provides extremely simple process for sterilizing the culture solution and eliminates the sterilization processes for each object, lid, bag, and solution respectively.
