Abstract

PURPOSE: To produce an artificial antigen containing a choline residue by reacting a peptide containing a free amino group with a compound obtained by the reaction of choline with a dicarboxylic acid anhydrides and to obtain an antibody of higher titer against acetylcholine using the same.
CONSTITUTION: A compound as given by the formula I (where X- represents monovalent anion such as C1 and n is 2W10) is obtained by the reaction of choline with glutamic anhydride or the like as given by the formula II (where n is 2W10) in a solvent of dimethylformamide or the like mear the room temperature with a tertiary amine such as pyridine as reaction accelerator. Then, the compound as given by the formula I is dissolved in an aqueous phosphate buffer solution at pH4W7 and a peptide such as ox serum albumin or the like having a free amino group is added thereto. Then, the mixture is brought into reaction overnight with 1-ethyl-3-(3- dimethylaminopropyl)carbodiimide or the like as a reaction accelerator. The reaction product is purified by dialysis and freeze-dried to yield an artificial antigen. The antigen thus obtained is injected into a rabbit or the like and the blood is extracted to produce an antibody.
