Abstract
PURPOSE: To obtain an antitumoral protein polysaccharide WAK, by culturing protein polysaccharide WAK-producing bacteria such as Aspergillus oryzae A155, Aspergillus oryzae IFO4176 or Aspergillus oryzae NK.
CONSTITUTION: Protein polysaccharide WAK-producing bacteria are cultured in a culture medium containing embryos of crop such as rice or wheat. The culture product obtained is extracted with, for example, hot water. To the extract is added a metal salt such as CaCl2 to separate nucleic acids. To this solution is added CH3OH and the precipitate formed is dissolved in water. This solution is mixed again with CH3OH to form a precipitate. This precipitate is washed successively with 70% C2H5OH and 95% C2H5OH, and then dried to crude powder. Then, this crude powder is fractionated to obtain an eluate containing antitumoral components by a 0.1W0.4 M-NaCl concentration gradient method using, for example, DEAE cellulose and then title WAK is collected by concentration, salt removal and gel filtration of the eluate through, for example, Sephadex G-200.
