Abstract
PURPOSE:To improve the productivity of the titled substance, by extracting DNA having the genetic information related to the biosynthesis of the titled substance from bacteria, having the ability to produce the titled substance, and belonging to the genus Brevibacterium, incorporating the DNA into a vector, and introducing the recombined gene into the bacteria. CONSTITUTION:Chromosomic deoxyribonucleic acid (DNA) carrying the genetic information related to the biosynthesis of L-glutamic acid is extracted from bacteria, e.g. Brevibacterium lactofermentum ATCC 13869, belonging to the genus Brevibacterium, and having the ability of produce the L-glutamic acid and then treated with a restriction endonuclease. A plasmid or phage obtained from bacteria capable of donating the DNA or a variant strain deficient in the ability to produce the L- glutamic acid derived from the donor bacteria is then treated with the endonuclease and mixed with the chromosomic DNA to give a combined substance. The resultant combined substance is then introduced into the bacteria or a variant strain having the L-glutamic acid requirement thereof by the transforming method to give the aimed transformed strain.
