Abstract

PURPOSE: To accurately determine proteinic matter (toxohormone L) having fat- mobilizing activity by an experiment not on a living body but in a test tube.
CONSTITUTION: From such canceous-cell secretion as abdominal dropsy or fluid in the thoracic cavity of a cancer patient or cancer-carrying animal, the fractions precipitated by an ammonium sulfate fractionation of 40W70% saturation are collected and treated on the ion exchanger chromatography by the gradient elution technique. As the ion exchanger, DEAE cellulose is specially preferable. Then the fractions having fat-mobilizing activity are lyophilized and filtered by using a hydrophilic gel. From the fractions thus obtained, toxohormone L can be separated and refined within a certain pH range by the isoelectric point cataphoresis. A proper number of fat-tissue slices are added to a Krebs-Ringer carbonic acid buffer solution, and to this a toxohormone L aqueous solution and a Krebs-Ringer carbonic acid buffer solution containing 5% albumin are added to determine the liberated fatty acid by the Dole's method.
