Abstract

PURPOSE: To prepare a polysaccharide in high yield, by inoculating a bacterial medium consisting of methanol as a main carbon source with a specific bacterium belonging to the genus Pseudomonas so that the polysaccharide is accumulated, followed by collecting it from the medium.
CONSTITUTION: Pseudomonas sp. ID-196, a bacterium belonging to the genus Pseudomonas is inoculated on a bacterial medium consisting of methanol as a main carbon source to accumulate a polysaccaride in high yield. The culture solution is diluted by 10W20 times and filtered or centrifuged to remove solid bodies or materials, and twice as much acetone as the solution by weight is added to it to give a crude polysaccharide. The polysaccharide is dissolved in water again and subjected to the operation of protein removal, e.g., heat treatment, precipitation of isoelectric point, etc., and centrifuged to give a supernatant liquid, to which acetone is added to deposit a precipitate. The precipitate is dissolved in water, desalted by a ratio of water dialysis to give a white spongy purified polysaccharide.
