Abstract

NEW MATERIAL:An endotoxin-sensitive substance. A protein having an ultraviolet absorption at 280nm and having the property to hydrolyze the thrombin and the enzyme substrate of the known clotting enzyme of horseshoe crab by the action of endotoxin originated from Gram-negative bacteria.
USE: Substance for the detection and determination of endotoxin.
PROCESS: Amebosite obtained from the limph of horseshoe crab blood is extracted with hypotonic extractant to obtain amebosite.lizate, which is subjected to the affinity chromatography using a carrier such as heparin-bonded agarose derivative and eluted with a buffer solution containing 0.154MW0.5M NaCl to obtain an endotoxin-sensitive substance as a protein fraction. When the endotoxin-sensitive substance and the amebosite.lizate are subjected to the affinity chromatography, and endotoxin is added to the fraction obtained above in combination with the fraction obtained by the buffer solution containing ≤0.154M NaCl, then coagulation factor Xa and the enzume substrate of the known clotting enzyme of horseshoe crab are hydrolyzed.
