Abstract

PURPOSE:To enable high sensitization through the suppression of nonsingular agglutination, by a method wherein an antigen is treated into a condition to sensitively react to latex particles, and a liquid to be measured, containing the antigen, is caused to react to a latex reagent, in which the latex particles, being so sensitively reacted with the antigen, are treated with a serum containing the antigen, to optically measure a change in turbidity. CONSTITUTION:The latex reagent is prepared such that, after an antigen is treated into a condition to sensitively react to a polystyrene latex and the like, a latex, to which so treated antigen reacts sensitively, is added to an antiserum for the treatment, excess antiserum is removed, and the resultant liquid is resuspended into a buffer liquid. Said treatment enables the effective suppression of nonsingular agglutination. After a phosphatic buffer liquid is added to said latex reagent, a detecting substance, containing an antigen, is added for mixture to immediately record a change with the elapse of time in an absorbance of a light having a given wave length. After starting to record, a light-absorption increasing factor is found at an earliest time from the absorbance which increases rectilineally to measure the antigen. In the case of B type virus surface antigen, this permits the measurement at the sensitivity which is 1,000 times as high as that of conventional types.
