Abstract

PURPOSE: To improve the durability of an immobilized enzyme, by using a boric acid buffer solution containing citric acid in a column packed with the immobilized enzyme having the optimal pH in an acidic region, e.g. glucose oxidase, cholesterol oxidase, peroxidase, urease, etc.
CONSTITUTION: An enzyme, e.g. glucose oxidase, cholesterol oxidase, peroxidase or urease, having the optimal pH in an acidic region is immobilized by the well- known method. The resultant immobilized enzyme is then packed in a column, and the enzymic analysis is carried out with a phosphoric acid buffer solution containing an acid, e.g. citric acid or succinic acid.
