Abstract

PURPOSE: To isolate the pellucid zone of mammals which can be used as a contraceptive, in high purity, by centrifuging the ovary of a mammal in a plural layers of solvents having different specific gravities from each other, and centrifuging the separated egg cells in the similar manner.
CONSTITUTION: The ovary of a mammal is extracted, homogenized with an arbitrary homogenizing means, and centrifuged in plural layers (usually two layers) of solvent having different gravities from each other (e.g. a buffer solution of 0% sucrose concentration and a 40W60% sucrose solution) to isolate the egg cells from the ovary. The centrifugal separation is carried out usually at an acceleration of about 1,000W4,000g. The egg cells are accumulated to the interface of the layers having different specific gravities, and the most impurities are precipitated to the bottom and removed. The isolated egg cells are disintegrated and the pellucid zones are released from the cells by homogenizing the cells. After the treatment, the product is again centrifuged using plural solvents having different specific gravities to separate the protoplasm membrane and the contents of the cells from the product. The pellucid zone having high specific gravity is precipitated to the bottom, and collected in high purity.
