Abstract

PURPOSE:To prepare a sample for endotoxin assay with high sensibility, by gel filtration of a plasma sample using a gel of allyldextran cross-linked with N,N'- methylenebisacrylamide. CONSTITUTION:A sample of plasma or blood product is subjected to gel filtration using a gel of cross-linked allyldextran with N,N'-methylenebisacrylamide to prepare a sample for endotoxin assay, followed by recovery of a fraction of the highest molecular weight. As a kit for sample preparation above mentioned, a set is usable, that is composed of a column for sample filtration, a solution contg. a high molecular colored materials for addition to the sample and a buffer solution for sample elution. In this case, the column for sample filtration is made up in such a way that a column of 4-9mm. diameter is filled up with 1.5-2.5ml of gel of cross-linked allyldextran with N,N'-methylenebisacrylamide which is allowed to be in equilibrium with a buffer solution of 7.0-7.4pH and sealed.
