Abstract

PURPOSE:To accurately determine trace amounts of immunosuppressive acid protein (IAP) and to apply this method in cancer diagnosis, by method wherein an anti-IAP antibody, having a given concentration, is reacted with a specimen, especially a stepwise diluted solution of urine, and an IAP-sensitized red blood cell is then added to the reaction product to investigate the occurrence of agglomeration. CONSTITUTION:IAP taken from human body fluid is injected to a rabbit, and the obtained immune serum is incorporated into normal human serum to prepare anti- IAP antibody, which is the diluted to a given concentration to prepare a diluted solution of said antibody. Meanwhile, an IAP-sensitized sheep red blood cell is prepared. Said anti-IAP antibody diluted solution is added to a specimen (serum, abdominal dropsy, urine, etc.), especially that obtd. by the addn. of a stepwise diluted solution of urine and incubated at 37 deg.C for about 30min is prepd., and then IAP- sensitized red blood cell PBS-BS liquid is added thereto. After the solution is allowed to stand at a room temperature for 2hr, the occurrence of agglomeration of the red blood cell is checked. The occurrence of agglomeration shows the presence of IAP. This enables the sensitive and accurate detection of IAP which appears especially distinctly in a cancer patient.
