Abstract
PURPOSE: An agarose derivative is used as a partitioning agent to treat cytosine deaminase with column chromatography, thus effecting simple and efficient purification.
CONSTITUTION: Cells in the suspension of cytosine deaminase-producing microorganism are crushed and extracted to give a solution containing cytosine deaminase. The resultant solution or its partially purified solution is subjected to the column chromatography filled with a partitioning agent of ω-aminoalkylated agarose, agarose with ω-aminoalkylated thiol-substituted pyrimidine immobilized, ω- aminoalkylagarose with carboxyl-substituted pyrimidine immobilized, ω-carboxylalkylagarose with amino-substituted pyrimidine immobilized or ω-carboxyalkanoyl- ε-iminoalkyl-agarose with amino-substituted pyrimidine immobilized.
