【発明の詳細な説明】
本発明は新規なカルボスチリル誘導体に関す
る。
本発明の化合物は新規化合物であり、一般式
〔式中Rは低級アルキル基を、Yはイオウ原子又
はスルホニル基を、nは1〜4の整数を、3、4
位の点線は飽和若しくは二重結合を夫々示す。〕
で表わされるカルボスチリル誘導体である。該化
合物は血小板凝集抑制作用及び消炎作用を有し、
血栓予防薬及び消炎剤として有用である。
一般式()で表わされる化合物において、R
で示される低級アルキル基としては炭素数1〜4
の直鎖もしくは分枝状のアルキル基を挙げること
ができ、具体的にはメチル基、エチル基、n−プ
ロピル基、イソプロピル基、n−ブチル基、tert
−ブチル基、sec−ブチル基等を例示できる。
本発明の代表的な化合物を以下に掲げる。

本発明の化合物は種々の方法により製造される
が、その好ましい1例を挙げれば下式に示す如く
公知の一般式()で表わされるヒドロキシカル
ボスチリル誘導体と一般式()で表わされる化
合物とを反応させることにより一般式()で表
わされるカルボスチリル誘導体を得、次いで一般
式()の化合物と一般式()で表わされるチ
オールのアルカリ塩とを反応させることにより一
般式()の化合物中Yがイオウ原子である化合
物(−A)が製造される。また一般式(−
A)の化合物を酸化することにより一般式()
の化合物中Yがスルホニル基である化合物(−
B)が製造される。
一般式()の化合物と一般式()の化合物
との反応は通常の脱ハロゲン化水素反応の条件下
に行なわれる。用いられる脱ハロゲン化剤として
は種々の塩基性化合物があり、たとえば、水酸化
ナトリウム、水酸化カリウム、炭酸ナトリウム、
炭酸カリウム、炭酸水素ナトリウム、炭酸水素カ
リウム、炭酸銀などの有機塩基、ナトリウム、カ
リウムなどのアルカリ金属、ナトリウムメチラー
ト、ナトリウムエチラートなどのアルコラート、
トリエチルアミン、ピリジン、N・N−ジメチル
アニリンなどの有機塩基が挙げられる。斯かる反
応は無溶媒でも、あるいは溶媒の存在下でも行な
われ、溶媒としては反応に悪影響を与えない不活
性のものがすべて用いられ、たとえば、メタノー
ル、エタノール、プロパノール、ブタノール、エ
チレングリコールなどのアルコール類、ジエチル
エーテル、テトラヒドロフラン、ジオキサン、モ
ノグライム、ジグライムなどのエーテル類、アセ
トン、メチルエチルケトンなどのケトン類、ベン
ゼン、トルエン、キシレンなどの芳香族炭化水素
類、酢酸メチルエステル、酢酸エチルエステルな
どのエステル類、N・N−ジメチルホルムアミ
ド、ジメチルスルホキサイド、ヘキサメチルリン
酸トリアミドなどの非プロトン性極性溶媒、水な
どが挙げられる。また、本反応は沃化ナトリウ
ム、沃化カリウムなどの金属沃化物の存在下に行
なうのが有利である。一般式()の化合物と一
般式()の化合物との使用割合は特に限定され
ず広範囲の中から適宜に選択されるが、通常前者
に対して後者を等モル〜5倍モル、好ましくは等
モル〜2倍モル量にて用いるのが望ましい。ま
た、その反応温度はとくに限定されないが、通
常、室温〜200℃、好ましくは50〜150℃で行なわ
れる。反応時間は通常1〜30時間、好ましくは1
〜15時間である。
一般式()の化合物と一般式()の化合物
との反応は無溶媒であるいはメタノール、エタノ
ール等のアルコール類、モノグライム、ジグライ
ム等のグライム類、エチルエーテル、テトラヒド
ロフラン、ジオキサン等のエーテル類、クロロホ
ルム、塩化メチレン等のハロゲン化炭化水素類、
ジメチルホルムアミド、ジメチルスルホキシド、
ヘキサメチルリン酸トリアミド等の溶媒中で行な
われる。一般式()の化合物の使用量は一般式
()の化合物に対して通常等モル〜5倍モル、
好ましくは等モル〜2倍モルである。この反応の
反応温度は通常0〜200℃、好ましくは室温〜100
℃であり、反応時間は0.5〜12時間である。以上
の如くして一般式(−A)の本発明化合物が製
造される。
一般式(−A)の化合物の酸化反応は慣用の
配化剤、例えば過酸化水素水、過酢酸、過安息香
酸、メタクロロ過安息香酸等の有機過酸、無水ク
ロム酸と空気等を用いて行なわれる。斯かる酸化
剤の使用量は一般式(−A)の化合物に対して
通常2倍〜大過剰量、好ましくは2〜4倍モルで
ある。該酸化反応は通常の不活性溶媒、例えば酢
酸、プロピオン酸、クロロホルム、塩化メチレ
ン、ジメチルホルムアミド等の溶媒中で行なわれ
る。反応温度は通常−20〜100℃、好ましくは0
〜50℃であり、反応時間は通常1〜24時間であ
る。以上の如くして一般式(−B)の本発明化
合物が製造される。
斯くして得られる本発明化合物は反応終了後常
法に従つて反応混合物から単離される。例えば反
応混合物の溶剤を留去あるいは稀釈することによ
り得られる。得られた化合物を必要に応じ分別再
結晶法、カラムクロマトグラフイー、薄層クロマ
トグラフイー等の通常の方法により更に精製する
ことができる。
更に一般式()における(−CH2)−oが有枝の
アルキレン基に置き変わつた化合物、例えば6−
(3−エチルチオ−2−メチルプロポキシ)−3・
4−ジヒドロカルボスチリル、6−(3−エチル
スルホニル−2−メチルプロポキシ)カルボスチ
リルも本発明化合物と同様の薬理活性を有する。
以下に実施例を掲げて本発明を更に明らかにす
る。
実施例 1
エタノール200mlに6−ヒドロキシ−3・4−
ジヒドロカルボスチリル32.5g、ナトリウムエチ
ラート16.5g及び沃化カリウム0.5gを加えて還
流させ溶解させる。この溶液に1−ブロモ−3−
クロロプロパン35gを加えて還流撹拌を3時間行
う。冷後、反応液を飽和食塩水に注ぎ析出晶を
取する。希水酸化ナトリウム水溶液で十分洗浄し
たのち水洗する。得られた結晶をエタノールから
再結晶して無色針状晶の6−γ−クロロプロポキ
シ−3・4−ジヒドロカルボスチリル28gを得
る。融点131〜133℃
N・N−ジメチルホルムアミド50mlに沃化カリ
ウム0.2g及びエチルメルカプタン2mlを加えて
室温撹拌下水素化ナトリウム0.25gを添加する。
水素の発生終了後、6−γ−クロロプロポキシ−
3・4−ジヒドロカルボスチリル1.5gを添加し
80〜90℃にて3時間撹拌する。冷後反応液を水約
350mlに注ぎ析出晶を取する。得られた結晶を
乾燥後リグロインから再結晶して無色針状晶の6
−(3−エチルチオプロポキシ)−3・4−ジヒド
ロカルボスチリル1.3gを得る。
融点87〜89℃
上記と同様にして下記化合物を得る。
Γ6−(3−エチルチオプロポキシ)−カルボス
チリル 融点126〜128℃
Γ6−(2−ブチルチオエトキシ)−3・4−ジ
ヒドロカルボスチリル 融点86.0〜88.5℃
実施例 2
6−(3−エチルチオプロポキシ)−3・4−ジ
ヒドロカルボスチリル1.0gを酢酸30mlに溶解さ
せ30%H2O2水2mlを加えて室温下、撹拌を一昼
夜行う。反応後反応液を飽和食塩水約400mlに注
ぎ析出晶を取し、水洗後乾燥する。得られた結
晶をクロロホルムから再結晶して無色針状晶の6
−(3−エチルスルホニルプロポキシ)−3・4−
ジヒドロカルボスチリル0.88gを得る。融点184
〜185.5℃
上記と同様にして下記化合物を得る。
Γ6−(3−エチルスルホニル)プロポキシ−
カルボスチリル 融点221〜223℃
実施例 3
エタノール200mlに6−ヒドロキシ−3・4−
ジヒドロカルボスチリル32.5g、ナトリウムエチ
ラート16.5g及び沃化カリウム0.5gを加えて還
流させ溶解させる。この溶液に1−ブロモ−2−
クロロエタン31.9gを加えて還流撹拌を3時間行
う。冷後、反応液を飽和食塩水に注ぎ折出晶を
取する。希水酸化ナトリウム水溶液で十分洗浄し
たのち水洗する。得られた結晶をエタノールから
再結晶して6−β−クロロエトキシ−3・4−ジ
ヒドロカルボスチリル18gを得る。
融点150−152℃。
N・N−ジメチルホルムアミド50mlに沃化カリ
ウム0.2g及びエチルメルカプタン2mlを加えて
室温撹拌下水素化ナトリウム0.25gを添加する。
水素の発生終了後、6−β−クロロエトキシ−
3・4−ジヒドロカルボスチリル1.4gを添加し
80〜90℃にて3時間撹拌する。冷後反応液を水約
350mlに注ぎ折出晶を取する。白色結晶の6−
(2−エチルチオエトキシ)−3・4−ジヒドロカ
ルボスチリル1.1gを得る。融点96−99℃。

以下本発明化合物につき行なつた薬理試験例を
あげる。
〈薬理試験〉
ネイチヤー第927〜929頁(1962年)に記載の方
法に準じて血小板凝集阻止作用を調べた。即ち血
小板凝集阻止作用をAG−型の凝集計
(aggregometer)〔プライスマン・マニユフアク
チユアリング・コンパニー(Bryston
Manufacturing Co.)製〕を用いて測定した。兎
から採取した血液試料はクエン酸ナトリウムと全
血液の混合物でその混合比率は1:9(容量比)
である。該試料を1000rpmで10分間遠心分離し
て、血小板濃度の高い血漿(platelet rich
plasma)(以下「PRP−1」という)を得る。得
られたPRP−1を分離し、残りの血液試料を
3000rpmで15分間さらに遠心分離して血小板濃度
の低い血漿(platelet poor plasma)(以下
「PPP」という)を得る。
前記PRP−1中に含まれている血小板の数をブ
レツチヤー・クロンカイト法(Brecher−
Clonkite Method)で測定し、PRP−1をPPPで
希釈してアデノシン・ジホスフエート(ADP)−
誘発凝集試験に供するため300000/〓の血小板を
含む試料(以下「PRP−2」という)を調製し、
またコラーゲン−誘発凝集試験に供するため
450000/〓の血小板を含む試料(以下「PRP−
3」という)を調製した。
(1) ADP−誘発凝集抑制試験
試験すべき化合物を予め定めた濃度で含有す
る溶液0.01mlに上記で調製したPRP−2を0.6
ml加え、混合物を温度37℃の恒温槽に1分間入
れた。次に該混合物にADP溶液を0.07ml加え
た。この混合物の透過度を測定し、透過度の変
化を撹拌器の回転速度1100rpmにて凝集計を用
いて測定した。この試験において用いられる
ADP溶液は、オーレン・ベロナール緩衝液を
用い、濃度が7.5×10-5Mとなるように調製し
たものである。血小板の凝集が最大となつた時
点(光の透過度が最大となつた時点)の凝集率
を下記の式より算出した。
凝集率=c1−a1/b1−a1×100
ここでa1:PRP−2の透過度
b1:PPPの透過度
c1:試験化合物及びADPを混合したPRP−2
の透過度
上式で算出された凝集率をB1とする。また
試験化合物を使用しない以外は上記と同様にし
て血小板を凝集させて凝集率を求め、この凝集
率をコントロールの凝集率A1とする。
試験化合物の血小板凝集阻止作用は、コント
ロールの凝集率に対して阻止率(%)として求
めた。
阻止率(%)=A1−B1/A1×100
(2) コラーゲン−誘発凝集抑制試験
試験すべき化合物を予め定めた濃度で含有す
る溶液0.01mlに上記で調製したPRP−3を0.6
ml加え、混合物を温度37℃の恒温槽に1分間入
れた。次に該混合物にコラーゲン溶液を0.07ml
加えた。この混合物の透過度を測定し、透過度
の変化を撹拌器の回転速度1100rpmにて凝集計
を用いて測定した。この試験においてコラーゲ
ンは、100mgのコラーゲンにオーレン・ベロナ
ール緩衝液(PH7.35)5mlを加えてすりつぶ
し、その上澄液を使用した。血小板の凝集が最
大となつた時点(光の透過度が最大となつた時
点)の凝集率を下記の式より算出した。
凝集率=c2−a2/b2−a2×100
ここでa2:PRP−3の透過度
b2:PPPの透過度
c2:試験化合物及びコラーゲンを混合した
PRP−3の透過度
上式で算出された凝集率B2とする。また試
験化合物を使用しない以外は上記と同様にして
血小板を凝集させて凝集率を求め、この凝集率
をコントロールの凝集率A2とする。
試験化合物の血小板凝集阻止作用は、コント
ロールの凝集率に対して阻止率(%)として求
めた。
阻止率(%)=A2−B2/A2×100
後記する各実施例で得た本発明化合物につき、
上記で求めた阻止率(%)が50%となる各試験化
合物の使用濃度(IC50)を求めた結果を下記第1
表に示す。
