Description

【発明の詳細な説明】 （産業上の利用分野） 本発明はさそうの治療に有用な安定化された透明な過酸
化ベンゾイル組成物に間する。貯蔵中退酸化ベンゾイル
が分解さるのを防止する組成物に関する。
（従来技術） 例えばリチャード　エム　コックス（Ｒｉｃｈａｒｄ　
ＭＣｏｘ）とレオナード　アール　シアフオ（Ｌｅｏｎ
ａｒｄ　ＲＣｉａｆｏ）に対して１９８０年ｌθ月２０
日公布されスタイフェル　ラボラトリーズ、インコーホ
レイチット（Ｓｔｅｉｆｅｌ　Ｌａｂｏｒａｔｏｒｉｅ
ｓ、　Ｉｎｃ、）に譲渡されたアメリカ合衆国特許３，
５３５，４２２号中で開示されている様に過酸化ベンゾ
イルを局所的に適用することが尋常性じそう（ａｃｎｅ
　ｖｕｌｇａｒｉｓ）の治療に有効なことがわかった。
主な嫌気性さそうを起こすバクテリアであるプロピオノ
バクテリウムアクネ（Ｐｒｏｐｉｏｎｏｂａｃｔｅｒｉ
ｕｍＡｃｎｅ）対する過酸化ベンゾイルの抗微生物効果
のために過酸化ベンゾイル組成物は主な抗さそう組成物
になった。一般に過酸化ベンゾイルは約２．５から約Ｉ
Ｏ重量／重量％までの過酸化ベンゾイルを含んでいる懸
濁液として局所的に適用される。
しかしながらざそうの治療に於ける過酸化ベンゾイルの
有効性は冒された区域に適用された過酸化ベンゾイルと
過酸化ベンゾイル組成物の安定性の関数であることがわ
かった。しかしながら過酸化ベンゾイルは溶液中で極め
て不安定で急速に劣化して不活性の安息香酸になる。以
前の過酸化べンゾイル組成物は貯蔵中に急速に分解し従
って程良い貯蔵寿命を有していない。そうであっても望
ましい抗さそう効果を与えるものは分解に際し生ずる遊
離基の酸化効果であるから、皮膚に接触したとき過酸化
ベンゾイルの分解が起こることが極めて望ましい。
（発明が解決しようとする問題点） 従って貯蔵中安定でありながら皮膚に接触したとき容易
に分解する過酸化ベンゾイル組成物を得ることが極めて
望ましい。その上過酸化ベンゾイル自体は無色無臭の結
晶質の固体であるからそのような過酸化ベンゾイル組成
物が透明であることは更に望ましいことであろう。
（問題を解決する為の手段） 過酸化ベンソイルが溶解されているＣＢ乃至Ｃ１０のト
リグリセリドを使用することによって安定な透明な過酸
化ベンゾイル組成物を得ることが出来ることが見つけら
れた。
本発明の組成物は一般式 ％式％） の短い鎖のトリグリセリドのｌＯＯχにするのに充分な
量中の約０．１乃至約３．８重ｌｌ／重量％の過酸化ベ
ンゾイルの安定な透明な無水組成物からなる。
式中、Ｒ，Ｒ，とＲ２は６乃至１０個の炭素原子を含ん
でいる直鎖又は分枝鎖の飽和脂肪族炭化水素基である。
Ｒ，Ｒ，と　Ｒ２は同か又は異なった脂肪族炭化水素基
である。基のアシル部分は一般にカプロン酸、エナンチ
ル酸、カプリル酸、ペラルゴン酸又はカプリン酸の様な
脂肪酸から誘導される。
本発明に従って、トリカブロイン、トリカブリリン、ト
リカプリン、又はそれらの混合物の様な単一のトリグセ
ライト又はトリグリセリドの混合物が使用されうる。
所望により本発明の組成物は一般に約８重量／重量％迄
の量のゲル化剤を存在すしめうる。好ましいゲル化剤は
シリカである。他の任意付加的な成分、例えは約２重１
１／重量％迄の少量の滑剤も又存在しうる。滑剤の例と
して例えばシクロメチコーン（ｃｙｃｌｏｍｅｔｈｉｃ
ｏｎｅ）などの様な低分子量のシリコーン油が挙げられ
る。
本発明の典型的組成物は次の過酸化ヘンジイルゲルから
成る。

０．０χにするのに充分な量 本発明の好ましい組成物は次の組成物の過酸化ベンゾイ
ルゲルからなる。
謄勿　　　　　　　　　ｌ工１１１Ｊ＝ＪＪ）無水過酸
化ベンゾイル　　　２．５ｚ Ｃｏ　−Ｃｏｏ　）リグリセリト ６０／４０混合物　８７．５χ シリカ　　　　　　　　　８．０χ シクロメチコーン （ダウコーニング３４４流体）２．Ｏγ過酸化ベンゾイ
ル透明溶液を先ず！・リグクセリド中でつくり、次いで
他の任意付加的成分、例えばゲル化剤、滑剤などを加え
、透明なゲル生成物をつくる。
（効果） 好ましい組成物は室温で６か月の貯蔵の後９１％過酸化
ベンゾイルゲルのままで残っている透明で安定なもので
ある。その上上記の組成物は３人の志願者に対して行な
われた１０日の生体内の抗細菌研究で試験された時、皮
膚微生物　を生じるプロビオノバクテリウムアクネを１
．５乃至２．０対数丈減少させるのに有効な事がわかっ
た。志願者には組成物が顔半分の区域に一日二回適用さ
れた。皮膚の微生物　は２，３．５及び１０日に得られ
、嫌気性条件下で適当な培養の後皮膚の微生物　を測定
した。細菌の減少は顔の治療された側と治療されない側
の微生物の数を比較する事によって決められた。
さそうは本発明の透明で安定な組成物の抗ざそうに有効
な量を患者に局所的に適用することで患者に於いて処置
された。

Claims (1)

  1. 【特許請求の範囲】 １、Ｃ＿６乃至Ｃ＿１＿０のトリグリセリド中の約０．
    １から約３．８重量／重量％迄の無水過酸化ベンゾイル
    からなる透明安定な過酸化ベンゾイル組成物。 ２、約２．５重量／重量％の量で組成物中に過酸化ベン
    ゾイルが存在する請求範囲１の組成物。 ３、ゲル化剤を含んでいる特許請求の範囲第１の組成物
    。 ４、ゲル化剤を含んでいる特許請求の範囲第２の組成物
    。 ５、約０．１乃至約３．８重量／重量％の無水過酸化ベ
    ンゾイル、 約８．０重量／重量％迄のゲル化剤、 約２．０重量／重量％迄の滑剤、 １００．０重量／重量％にするに充分なトリグセライド
    、 からなる特許請求の範囲第３の組成物。 ６、約２．５重量／重量％の無水過酸化ベンゾイル、約
    ８７．５重量／重量％のＣ＿８−Ｃ＿１＿０のトリグリ
    セリド混合物、 約８．０重量／重量％のシリカ、 約２．０重量／重量％のシクロメチコーン、からなる特
    許請求の範囲第３の組成物。 ７、Ｃ＿６乃至Ｃ＿１＿０のトリグリセリド中の約０．
    １乃至約３．８重量／重量％迄の無水過酸化ベンゾイル
    からなる透明安定な過酸化ベンゾイル組成物の抗ざそう
    に有効な量をざそうの治療の必要な患者に局所適用する
    ための治療薬。 ８、約２．５重量／重量％の量で組成物中に過酸化ベン
    ゾイルが存在する特許請求の範囲第７の治療薬。 ９、ゲル化剤を含んでいる特許請求の範囲第７の治療薬
    。 １０、ゲル化剤を含んでいる特許請求の範囲第８の治療
    薬。 １１、約０．１乃至約３．８重量／重量％の無水過酸化
    ベンゾイル、 約８．０重量／重量％迄のゲル化剤、 約２．０重量／重量％の滑剤、 １００重量／重量％にするに充分なトリグリセリド、 からなる請求範囲７の治療薬。 １２、約２．５重量／重量％の無水過酸化ベンゾイル、
    約８７．５重量／重量％のＣ＿８〜Ｃ＿１＿０のトリグ
    リセリド混合物、 約８．０重量／重量％のシリカ、 約２．０重量／重量％のシクロメチコーン、からなる特
    許請求の範囲第７の治療薬。
