Description

【発明の詳細な説明】 (イ)産業上の利用分野 この発明はバニルマンデル酸およびホモバニリン酸の同
時分析法に関する。さらに詳しくは、高速液体クロマト
グラフィーと電気化学検出器を用いるバニルマンデル酸
(以下VMA)およびホモバニリン酸(以下HVA)の
同時分析法に関する。
(ロ)従来技術 小児ガンの一種である神経芽細胞腫の早期発見法として
、尿中のVMAとHVAの二成分を測定する方法が有用
であることが知られており、これは、通常高速液体りO
マドグラフィーを用いて行なわれている。
高速液体クロマトグラフィーにより分離し、蛍光検出器
により分析する従来技術によるVMAおよびHVA同時
分析法においては、移動相としてpH2〜3.5の酸性
緩衝液とアセトニトリルの混合溶媒が用いられている。
pH2〜3.5と酸性にするのはVMAと尿中夾雑物質
(蛍光物質)との分離を効率良く達成するためである。
(ハ)発明が解決しようとする問題点 上記に示したように、蛍光検出器を用いる場合には、p
H2〜3.5の酸性条件下で行なう必要があった。しか
しながら酸性度を強めるとVMA%HMA共に検出感度
が著しく低下する傾向がある(第4図参照)。従って、
分離が良好に行なわれても、分析の精度が低下するとい
う問題点があった。
そこで、蛍光検出器よりも高感度なアンベロメトリやク
ーロメトリを用いた、いわゆる電気化学検出器を用いる
方法が考えられる。
しかしながら蛍光検出器を用いる従来技術の移動相と同
じもの、すなわち−2〜3.5の酸性!1衝液とアセト
ニトリルの混合溶媒からなる移動相と電気化学検出器と
組合せた場合には、他の夾雑物のピークことに尿中の夾
雑物ピークによってVMAのピークが妨害され定量困難
となる不都合が生じる(第2図参照)。
このために各ピーク間隔を広げる目的でアはトニトリル
の添加量を減少させることも考えられるが、この場合に
はVMAのピークが明瞭に得られても、HMAの溶出時
間が長<(40分以上)実用には到庇適さない(第3図
参照)。
この点に関し、発明者らは鋭意研究を行なった結果、移
動相の−を4〜8滅で上昇させることにより、分析時間
の増大を1& <ことなく HV AおよびVMAと尿
中夾雑物(電気化学的陽性物質)の分離が達成され、さ
らにVMAの感度自体も茗しく向上することができると
いう事実を見出した。
(ニ)問題点を解決ザるための手段および作用かくして
この発明によればバニルマンデル酸およびホモバニリン
酸を含み得る試料を高速液体クロマトグラフィーに付す
ことからなり、該クロマトグラフィーにおける移動相と
して、水と混合しつる有機溶媒を任意に含むpH4〜8
のリン酸緩衝液を用いかつ検出器として電気化学検出器
を用いて分離分析を行なうことを特徴とするバニルマン
デル酸およびホモバニリン酸の同時分析法が提供される
この発明に使用する移動相は、−4〜8に調製したリン
酸緩衝液または有機溶媒−リン酸緩衝液の混合溶媒が使
用でき、リンF11M’に液としては例えば、リン酸ナ
トリウム緩衝液、リン酸カリウム緩衝液などが使用可能
である。
リン酸緩衝液に任意に含ませてよい有機溶媒は水と混合
しうるちのであればよく、例えばアセトニトリル、メタ
ノール等である。
有機溶媒のリン酸緩衝液に対する割合は、0〜awt%
であることが適切である。awt%を越えると−をこの
範囲で低めてもVMAが迅速に溶出されすぎて好ましく
ない。すなわち有機溶媒の岱は、VMAが少なくとも明
瞭に保持され、溶媒のピークやインジェクションショッ
クのピークに極端に近接しないように規定するのが好ま
しい。
電気化学検出器は当該分野に用いられるものはいずれも
使用できて、例えばアンベロメトリ、クーロメトリ等が
使用できる。電解電圧は+0.2〜+ 0.5 mVv
sPb極に設定するのが適している。
かかる電気化学検出器はクーロケム5100A <米国
ESA社)等の名称で入手可能である。
カラムとしては、通常の逆相液体クロマトグラフィー用
のカラムが使用できて、例えばいわゆるODSカラム、
C8カラムまたはTMSカラムが使用可能であり、OD
Sカラムが好ましい。
(ホ、)実施例 以下この発明を実施例によって説明するが、この発明を
限定するものではない。
下記の条件下でVMAおよびHVAの同時分析を行なっ
た。
(ω 分析条件 カラム:S旧m −pack  CLC−ODS(6,
OM [、D、 X15alL、 。
島津製作所製) カラム温度:40℃ 流Δ:1.01/分 検出器:クーロケム5100A 移動相:PH5,0の10 mMリン酸ナナトリウム緩
衝液975容量%とアセトニトリル 2.5容量%の混合溶媒 +b)  サンプル 生後約3カ月児の尿を採尿し、この11ノに酢酸エチル
および塩酸を加え、Ij!拌し、有機層を分取し、蒸発
乾固し、これを200dの移動相に溶解したものを注入
用試料とした。
上記この発明の方法において得られたクロマトダラムを
第1図に示した。図中(1)はVMA (以下同じ)、
(211,1HMA(以下同じ)である。このようにこ
の発明によれば、VMA付近の夾雑物とVMAの分離も
良く、さらにVMA、HVAの高感度で適正なピークを
短時間で得られていることがわかる。
第2図および第3図は移動相として蛍光検出器を用いる
従来技術の移動相(第2図・・・・・・PH3,0のリ
ン1%9101Mナトリウム緩衝液96.5容量%とア
セトニトリル3.5容量%の混合溶媒、第3図・・・・
・・−3,0のリン1%101Mナトリウム緩衝液97
.5容量%とアセトニトリル2.5容量%の混合溶媒)
を用いた比較例である。
第2図は、溶出に要する時間は第1図に比較して長いに
もかかわらず、VMA付近の夾雑物とVMAの分離が充
分ではない。
第3図は、VMA付近の夾雑物とVMAの分離は良くな
っているがHMAの溶出時間が非常に長い。
このように比較例はともに実用性のないものである。
(へ)発明の効果 この発明の方法によれば、VMAおよびHVAの分析に
おいてシャープなピークを得ることができて、従来にな
い定量性の蟲い分析を行なうことができる。またこれに
あわせて溶出時間の短縮化の効果も得られる。

  1. 【特許請求の範囲】 1、バニルマンデル酸およびホモバニリン酸を含み得る
    試料を高速液体クロマトグラフィーに付すことからなり
    、 該クロマトグラフィーにおける移動相として、水と混合
    しうる有機溶媒を任意に含むpH4〜8のリン酸緩衝液
    を用いかつ検出器として電気化学検出器を用いて分離分
    析を行なうことを特徴とするバニルマンデル酸およびホ
    モバニリン酸の同時分析法。
