Description
本発明は家禽の感染に対し、更に詳しくはニユ
ウカツスル病及びアデノ様ウイルスに起因する急
激な産卵低下を特徴とする病気に対し即刻投与し
うる複合ワクチンに関する。本発明は更にこの複
合ワクチンの製造とこの産卵低下を防除するワク
チン成分の製造方法とに関する。
ニユウカツスル病〔以下にNDと略記する〕は
家禽における最も重大な呼吸病の一つであり、ウ
イルスによつてひき起こされ、全ての年令(成長
度)の家禽において高度の死亡率を惹起しうる。
ニユウカツスル病防除のための数種のワクチン
が開発されており、これらのワクチンは最高度の
安全性を与えるという理由からその不活化型で投
与されると好適であることが明らかとされてい
る。更に生ワクチンの使用は接種されたウイルス
がワクチン処理された家禽から免疫されていない
感染しやすい家禽へ時として伝播することにな
る。
比較的安全で十分に免疫力をもつ種々な生ND
ワクチンが開発されている(例えば英国特許第
1510100号明細書にみられる)が、それでもなお
不活化NDワクチンに対する明らかな要求があ
る。
家禽の呼吸病は屡々1種以上の感染源による結
果招来されるので複合ワクチンが開発された。例
えば英国特許第2798835明細書によれば感染性気
管支炎に対するワクチンとNDワクチン(両成分
とも生きたウイルスから誘導される)との組合せ
からなる。
更に、公開されたオランダ特願第7117873号明
細書(中間的に第157209号として異議の目的で公
示された)は家禽の呼吸病に対する複合ワクチン
の製造法を開示しているが、それは天然栄養培地
中でヘモフイルス ガリナリウム(Haemophilus
gallinarium)株を培養し次いで該細菌を不活化
して得られる“感染性鼻炎(coryza)に対する
死菌ワクチン”と、感染性気管支炎ウイルスから
の死菌ワクチンと、死菌NDワクチンとの組合せ
という特徴をもつ。
数種の文献〔例えばAvian Dis.7(1963)106
〜122頁、Am.J.Vet.Res.17(1956)294〜298
頁、Avian Dis.11(1967)399〜406頁及び
Virology 33(1967)598〜608頁〕は、各成分の
活性が生きたウイルスの混合によつてその相互阻
害のために減少し或は全く消失しうることを開示
している。
アデノ様ウイルス(以下ALVとして表わす)
の存在と、過去数年間で増加が認められていて特
にメデイアム ヘビイ レイ(medium heavy
lay)品種のブロイラ母鶏及び雛鶏において認め
られている急速な産卵低下及び(又は)軟殻卵及
び(又は)無殻卵の産生の増大との或る程度の関
連性については既に文献〔Avian Pathology 5
(1976)261〜272頁〕に指摘されている。この文
献はかような症状を時折“産卵低下症候群(egg
drop syndrome)”と呼んでいる。他の文献
〔Avian Pathology 6(1977)405〜413頁〕にも
127ウイルスと呼ばれる凝血ウイルス
(hemagglutinising virus)に対する抗体の産生
と上述の経済的に極めて不利な症状との間に見ら
れる関係を明らかにしている。他の報告(Avian
Pathology 7(1978)35〜47頁〕はこのアデノ様
ウイルスの存在と上述の産卵低下との間に仮定さ
れた関連性を確認している。更に問題となる型の
ウイルスの数種の特性例えば雛の赤血球の特徴的
な凝集及び既知のアデノウイルス型において抗血
清が中和されえないことを記載している。
ルクセンブルグ特許第79154号明細書は特異的
な凝血及び抗血清反応を特徴とするALVから出
発する抗“産卵低下症候群”ワクチンの製造法を
開示している。その詳細な説明及び実施例におい
てこのウイルスはEDS―76及びBC―14と呼ばれ
る。このワクチンはこの新規なウイルスを組織培
養、好適には雛の胚繊維芽細胞及び未発達
(embryonal)の肝細胞又は腎細胞のような鳥の
組織培養又は胚形成された(embryonated)アヒ
ル卵中で培養して製造される。
諸文献〔Avian Pathology 5(1976)262頁、
第2節;Avian Pathology 6(1977)406頁;及
びルクセンブルグ特許第79154号明細書第1頁7
〜11行〕の記載から産卵低下はニユウカツスル
病、感染性気管支炎、鳥類脊随炎及びセフアロミ
エリチス(avian and cefalomyelitis)鳥類ホウ
ソウ(birdpox)のウイルス及びそれらの合併に
よつて生じうることが明白である。
従つて、実際的見地から家禽は屡々上述の比較
的一般に生じるウイルス病に対する2種又はそれ
以上のワクチンで同時に又は短期の間隔をおいて
ワクチン処理されるであろう。特に家禽はニユウ
カツスル病とALV起因の産卵低下とに対して同
時にワクチン処理されるであろう。かような同時
投与に対して従来一般に行われた方法は即時使用
可能な別々のワクチンの標準量を混ぜ、スポツト
上に構成された1回分のワクチン量を投与するこ
とからなり、従つてその1回分の量は別々に投与
されるワクチンに比べて実質的に2培の容量から
なり各ウイルス量は希釈されている。更に、正確
な混合は保証されえないし、又一方において外来
性感染物質による問題を生じうる。
従つて本発明の目的はニユウカツスル病に対し
ても、又ALVに起因する産卵低下に対しても有
効で現在の多くの関連諸国の獣医達の厳しい要件
に合致していて即座に使用しうる複合のワクチン
の発見である。
驚くべきことに、複合の即時投与可能なワクチ
ンが不活化NDウイルス液と不活化ALV液との混
合によつて製造されうることが見出された。本明
細書中の用語“即座に投与しうる”、“即刻投与し
うる”、“即座に使用し得る”、及び“即時投与可
能な”“複合不活化ワクチン”はすべて次のこと
を意味する。即ち本発明の複合ワクチンは完成さ
れた形状のワクチンであること、即ち対象動物へ
の投与の際に混合されたり、相互に添加されたり
する必要のない複合ワクチンであることを意味す
る。これは従来技術のワクチンと対照的であつ
て、従前においては2種のワクチンは個別に注射
されるか又は使用直前に、標準容積を満足する状
態となるまで、少くとも混合されねばならなかつ
たのである。
本発明はまた少くとも1種のNDV液と少くと
も1種の不活化ALV液とから誘導され、また上
述のウイルス型の2種以上から随意に誘導される
不活化複合ワクチンにも関していることが理解さ
れよう。
本発明による方法において不活化NDV含有液
は原理的にはそれ自体既知の方法で、例えば所望
のウイルスを受精鶏卵中で培養し、培養されたウ
イルスをこの種のワクチンに普通な力価をもつ液
体中に集め、それを不活化し、そのウイルスを適
正な緩衝溶液中に懸濁し、それを均質に(ホモジ
ナイズ化)し、これをALVから誘導された不活
化ウイルス液と混合して製造されるが、この
ALVから誘導された不活化ウイルス液はそのウ
イルスをアヒル受精卵又はアヒル細胞の細胞培養
中で培養し、生じたウイルスをこの種のワクチン
に普通な力価の液中に集め、そのウイルス液を、
それ自体既知の方法によつて不活化することによ
つて製造される。
このNDV含有液の製造には種々なほとんど無
毒又は実質無毒〔遅現系又は速現系(lentogenic
or velogenic)〕で秀れた免疫性をもつNDウイル
ス、例えばラソタ(LaSota)又はヒチネル
(Hitchner)ウイルス(lentogenic)又はハーツ
(Herts)33/56(velogenic)ウイルスを使用し
うる。
利用しうるウイルス株の特定例は下記の通りで
ある:Institut fuer Medizinische
Mikrobiologie,Infections und Seuchenmedizin
der Ludwig Maximilians Univorsitaet,
Muenchenから得られるP/76/5,P/76/
4,及びP/76/3株;American Type
Culture Collection,Rockville,Mayland,USA
から得られるVR―108,VR―107,VR―109,
VR―699,VR―623株;Centr.Vet.Labs.,
Weybridge,Great Brittainから得られる
Queensland V4株又はCollection d′ Institut
Pasteur,ParisにI.091として寄託されたL/
Z258Pウイルス。
この不活化ALV液成分の製造にはCollection
d′ Institut Pasteur,ParisにNo..090として寄
託された継代ウイルス127株が好適に使用されう
るが、その他の、形態よりはむしろ抗原性につい
て、産卵低下をおこすALVウイルス型も十分に
利用されうる。このウイルス株I.090は好適には
アヒル卵中で培養される。
この不活化ALV液の製造は好適には上述の
Avian Patholagy文献に示されるように継代ウイ
ルス株127から誘導されたウイルスから出発され
るが、他の産卵低下病をひき起こすALV型も十
分に利用されうる。
この複合ワクチンの製造には150〜250mlの緩衝
化されたNDV含有液を不活化し、随意にホモジ
ナイズし、100〜200mlの予め不活化され随意にホ
モジナイズされた緩衝化ALV含有液と共に600〜
700mlの油相に添加する。この油相の必須成分は
軽パラフイン系鉱油(FDA品質)、植物油、及び
ナフテン系鉱油からなる群から選ばれる少くとも
一種の成分と、一種又はそれ以上の適正な乳化
剤、例えば酸化アルキレン付加物及び(又は)六
価アルコール及び(又は)高級天然脂肪酸(C10
〜C20)から誘導されるエステル及びエステル―エ
ーテルのような非イオン性界面活性化合物とであ
る。それらの例としてはマンニド モノオレエー
ト〔スパン(Span)80,アラセル(Arlacel)
80,アラセルA〕及びポリオキシエチレン
(20)ソルビタンモノオレエート〔例えばトウイ
ン(Tween)80〕がある。
この緩衝化されたNDV含有液と緩衝化された
ALV含有液との容積比は3:4〜5:1に変動
しうるが好適には約3:2である。
両方のウイルス液からなる水相と油相との容積
比は1:1〜3:7で変動しうるが、好適には
7:13である。
この水相は所望の粘性の少い安定な液状の完成
エマルジヨンを得るために激しく撹拌しながら、
及び(又は)ホモジナイズしながら油相に添加さ
れねばならないことが見出された。
この油相は2〜20重量%(油相重量に対し)の
乳化剤、好適には2〜15重量%のアラセルA又
はアラセル80又はスパン80と0.2〜4%のト
ウイン80とを含む。この油相の各成分は好適に
は別々にオートクレーブ中で少くとも110℃に加
熱されるか又は混合物として滅菌過される。
両方の出発ウイルス液成分の不活化は通常の不
活化剤を用いて、例えばβ―プロピオラクトンに
より行われうるが随意に安定化剤又はホルムアル
デヒドと組合わされる。好適にはβ―プロピオラ
クトンは緩衝化されたND―ウイルス含有液に対
し0.05〜0.25重量%(水相重量に対し)の濃度で
添加され、その液を約37℃に1〜2時間、好適に
は90分間恒温保持し、一方緩衝化されたALV液
はホルムアルデヒドにより好適には約20℃におい
て約20時間0.02〜0.5重量%の濃度で不活化され
る。次に必要に応じてそのウイルス液を通常の方
法でホモジナイズする。緩衝溶液中のチオメルサ
ール(thiomersal)又はホルマリンのような防腐
剤をそこで随意に水相に添加しうる。
投与後に所望の好ましい力価を与える複合ワク
チンを得るために109.8〜1010.5のEID50/ml好適
には1010EID50/mlの力価をもつNDV出発溶液
を使用せねばならず、また少くとも102HA単位、
好適には103HA単位の活性をもつALV液を使用
せねばならない。
本発明によつて提供される複合ウイルスワクチ
ンの好ましい性質は当業界の熟練者によつて全く
予知されえないことは明白である。その理由は当
業界の熟練者によつて確実に排除されえないいく
つかの好ましからぬ反応があるからである。最終
ワクチン中の一方又は両方のウイルス成分の活性
はそのウイルス液成分間の好ましからぬ相互作用
により、或はこれらの液成分と利用された補助化
成品の一種との相互作用によつて好ましからぬ水
準にすでに減少しうる。
例えば当業熟練者は、添加されたβ―プロピオ
ラクトンが長すぎる期間作用することによつて一
方のウイルス成分を過度に不活化してしまうこ
と、しかもかような長期間の作用はもう一方の成
分の不活化には必要なものであるということを確
かに排除しないであろう。
本発明による複合ワクチンの利用はスポツト上
で構成ワクチンを混合して製造される先行技術の
ワクチンと較べて投与のためにより少量の容積
(例えば1cm3の代りに0.5cm3)で済み、従つて局部
刺激を起し難いという点で有利である。更に投与
に必要な身体には異物である化成品の量がより少
くてよい。その上、混合と異物感染物質の導入に
よつて生じる上述の問題も回避される。
こうして製造されたワクチンは通常産卵前に10
〜20週令の鶏に投与される。
本発明は更に適正なALV含有の原料液の製造
法に関するがそれはアデノ様ウイルスをアヒル卵
中又はアヒル胚繊維芽細胞の培養、例えば単一層
ローリングボトル培養又は固体不活性担体に付着
した細胞培養上で培養することからなる。
本発明の好適な態様によればアヒル胚繊維芽細
胞の単一層ローリングボトル培養は原料細胞を少
くとも70〜90部のイーグル培地(Eagle′s
medium)、随意に5〜15部の仔牛血清、2〜4
%の炭酸水素ナトリウム溶液及び1〜5部の1種
又はそれ以上の抗生物質例えばペニシリンG、ス
トレプトマイシン及びナタマイシンの溶液又はサ
スペンシヨンからなる培養培地中に懸濁して形成
され、その培養液は細胞培養がほとんど終了した
ときに分離される。その細胞培養をウイルスサス
ペンシヨンで感染させ、37℃で0.5〜2時間恒温
保持してウイルスを付着させ、続いて2〜10%の
仔牛血清、2〜10部のトリプトースホスフエート
ブイヨン(30g/)、10〜20部の牛の羊水
(BAF)及び上述の添加剤を含むイーグル培地か
らなる維持培地を添加し、CPE出現後0.5〜2日
でそのウイルス含有培地及び細胞を収穫する。
この細胞培養に30×106TCID50/ローリング
培養ボトルの活性をもつALVサスペンシヨンを
接種する。これに関連して付言すると細胞サスペ
ンシヨン培養はこれまでに論じられた製法に適正
でないことが見出されている。
このワクチン製造にはCollection d′ Institut
Pasteur,Parisにno.I.0.90で寄託されている継代
ウイルス株127が好適に使用され、この株は好適
にはアヒル卵中で培養される。
ルクセンブルグ特許第79154号明細書が“産卵
低下症候群”に対するワクチン製造を一般的に開
示していることは真実であるが本発明による許容
されうる高力価のこれらのワクチン液に対する商
業的規模での好適な製造法は該明細書に全く記載
されておらず、また示唆すらされていない。
この点に関して該ルクセンブルグ特許明細書第
2頁、28〜30行に明瞭にそのウイルス(i,a.)
は雛の胚繊維芽細胞上で培養されうると記載され
ていることが指摘される。しかしながらかような
方法は所望の経済的に要求される結果を確かに全
くもたらさないことが判明している。第6頁29〜
35頁には未発達の雛肝細胞の培養を使用すること
を開示しているが、商業的規模での未発達肝細胞
の培養はまだ極めて困難であつて従つて高価な製
造法であると考えられているので所望の経済的に
要求される成績を決してもたらさない。
本発明は以下の諸例によつて例示される。
例 1
a アデノ様ウイルス(継代株127)の培養
ベルコ(Bellco)社製のローリング培養ボトル
中で0.22重量%のNaHCO3及び2容量%のペニシ
リンG、ストレプトマイシン及びナタマイシン含
有の抗生物質溶液を含むイーグル10%仔牛血清培
地中、DEF(Duck Egg Fibroblasts)(アヒル
卵繊維芽細胞)上で、12〜13日間抱卵されたアヒ
ル卵からの細胞培養物を製造した。
細胞培養がほとんど終了してからこの培地を吸
引する。そこでその培養に継代ウイルス127株の
サスペンシヨンを接種し、37℃で1時間恒温保持
し、5%の仔牛血清、10容量%のトリプトース
ホスフエート ブイヨン(30g/)、10容量%
の牛の羊水(BAF)、0.22重量%のNaHCO3及び
2容量%の上述の抗生物質溶液を含むイーグル維
持培地で満たす。
CPEが出現し、細胞がほとんどガラス壁の所
まで来るようになつたらその翌日にこのALV含
有培地及び細胞を収穫する。このウイルス サス
ペンシヨンを−20℃で凍結し、ワクチン製造用に
貯える。このサスペンシヨンのHA力価及び
TCID50を測定する。
b1 NDVの培養
抱卵11日間の後のSPF卵にL/Z258Pウイルス
を接種した。この培養を37℃で72時間恒温保持し
そのAAF(Amnion Allantonic Fluid)を収穫し
た。AAFはその時点で約1010EID50/mlを含有す
る。このウイルス サスペンシヨンを−20℃又は
それ以下で凍結し、ワクチン製造用に貯える。
b2 11日間抱卵されたSPF卵にハーツ(Herts)
33/56種ウイルスを接種した。37℃で24〜48時間
恒温保持した後にAAFを収穫するとこれはその
時点で約1010EID50/mlを含有する。そのウイル
ス サスペンシヨンを−20℃又はそれ以下で凍結
し、ワクチン製造用として貯える。
c ウイルス サスペンシヨンの処理
この凍結されたウイルス サスペンシヨンを解
凍し、それを0.05〜0.25%のβ―プロピオラクト
ンを用いて37℃の水浴中で90分間不活化する。そ
のサスペンシヨンを+4℃で一夜貯える。この不
活化はDEF(アヒル卵線維芽細胞)又はCEF
(Chicken Egg Fibroblasts)(鶏卵線維芽細胞)
に関するCPE′ Sの形成により観察され、次に血
液吸収試験(hemabsorption test)される。更に
予備抱卵された鶏卵に不活化ウイルス液を接種し
て恒温保持し、対照として用いた。
このウイルス サスペンシヨンをウルトラ テ
ユラツクス(Ultra Turrax)社製のホモジナイ
ザでホモジナイズする。所望によつてはこのサス
ペンシヨンをアデノ様ウイルス サスペンシヨン
のAAF及びHA力価のEID50に応じてPBS+0.3%
のホルマリンで希釈する。その後にこのNDV―

された:

まゝ残る;
2 37℃に4週間放置しても水相が分離しない。
こうして製造されたエマルジヨンの最終濃度
は、21%NDV含有の緩衝化された液と14%
ALV127含有の緩衝化された液とである。
得られたワクチンは動物一頭当り0.5mlの用量
で胸筋又は脚筋中に筋肉内投与、或は頚に皮下投
与で使用された。接種後8日目に凝血阻止拮抗化
合物(hemagglutination inhibiting
anticompounds)が認められた。
なお凝血阻止は免疫能の指標をなすものである
が、上記の試験における予防接種の効果は、該接
種後8日目に関連する抗体(即ち凝結阻止拮抗化
合物)の存在が観察され得たことによつて認めら
れたのである。
例 2
例1の方法を繰返すが但し次の変更を行つた:
a 例1のa項によるALV製造の代りに予め9
日間抱卵されたアヒル卵に継代127ウイルス株
を接種してALV含有液を製造する。37℃で7
日間恒温保持した後にAAF(Amnion
allantoic fluid)を収穫する。その時点でその
AAFは約1:1024のHA力価を持つ。このウイ
ルス サスペンシヨンを引続き−20℃で凍結し
ワクチン製造用に貯える。

例 4
ALV含有液とNDV含有液とをそれぞれ例1の
対応する工程に従つて製造した。このウイルス
サスペンシヨンを例1のc項のように処理した後
にこのNDV―AAFとALV含有液とを4:3の割
合で混合し、例1のd項に記載の方法でエマルジ
ヨンを製造した。しかしながらここではその油相
と混合ウイルス含有液とを6:4の割合で混合し
たが、そこで用いられる油相は例1のd項で使用
されたものである。
例 5
例4に従つて製造された混合ワクチンを次のよ
うに試験した。
予めワクチン処理されていない8週令のSPF雛
をこの混合ワクチンで免疫した。この場合に雛一
羽当り0.5mlの混合ワクチンを雛に対し筋肉(も
も)内注射して接種した。注射は胸の筋肉に対し
或は皮下に対しても行われ得る。この免疫された
動物に由来する血清中のHAI(凝血阻止)単位数
で示されるHAI力価は同一条件下で比較される組
成物の対応する単一ワクチンで免疫された動物に
見られる抗体水準から著しくは偏倚していなかつ
た。
見出されたHAI単位とHAI水準(レベル)とを
時間に対して対数スケールで2×の形でそれぞれ
表わしてプロツトして得られた累進曲線を添付図
に示した。
このNDV―HAI単位に関して、生NDVワクチ
ンで処理(通常圃場で行われるように)される以
前に予め免疫された動物については著しく高い価
が認められた。
この動物では例えば約1016.0〜1017.0NDV―HAI
単位が複合NDV―ALVワクチンで免疫後4〜8
週間で検出される。
比較例
本発明の複合不活化ワクチンがアデノ様ウイル
スに起因する産卵低下に対し有効であるとされる
技術的根拠を示すために以下に比較例について記
述する。
複合NCD/ALVワクチンを用いオンドリに対
してこれを接種した後、及びNCDワクチン単独
並びにALVワクチン単独を用い別の群のオンド
リに対して接種した後に夫々の被検群における抗
体産生を比較した。

ALV(アデノ様ウイルス)含有液と共に油相に
添加し、但し該ALV含有液はアヒル卵中又はア
ヒル胚繊維芽細胞の細胞培養物中の継代ウイルス
株127をアヒル卵中において又はアヒル胚繊維芽
細胞中において培養して得られ予め不活化され随
意にホモジナイズされたものであり、該ニユウカ
ツスル病ウイルス含有液と該アデノ様ウイルス含
有液との容積比は3:4〜5:1に変動し得るも
のであり、上記の油相は軽パラフイン系鉱油、植
物油、ナフテン系鉱油からなる群から必須成分と
して選ばれた少くとも1種の成分及び酸化アルキ
レン付加物及び(又は)六価アルコールと脂肪酸
(C10〜C20)とから誘導されるエステル及びエステ
ル・エーテルからなる群から選ばれた少くとも1
種の乳化剤を含有するものであり、“両方のウイ
ルス液から形成される水相”と油相との容積比が
