Description
本発明はアシル化インドール誘導体及びその製
造法並びにそれを有効成分とする血小板凝集阻止
剤に関する。
本発明で提供される新規化合物、アシル化イン
ドール誘導体に優れた血小板凝集阻止作用を有
し、それ故、心臓血管系の硬塞の予防、術後血栓
の予防及び治療、外科手術後の血管の血栓硬塞の
予防及び治療及びアテローム性動脈硬化症、動脈
硬化症等の予防もしくは治療に、心筋層の硬塞と
卒中発作後の再発の予防又は治療等に極めて有用
な化合物である。
本発明で提供される新規化合物アシル化インド
ール誘導体は、優れた抗炎症作用、線溶活性作用
を有する化合物としても期待され、極めて有用な
化合物である。
本発明で提供される新規なアシル化インドール
誘導体は下記式〔〕
〔式中、R1は水素原子、低級アルキル基又は
アセチル基、R2は水素原子又はメチル基を示
し、R3及びR4は同一もしくは異なり、水素原
子、ハロゲン原子、低級アルキル基又は低級アル
コキシ基を示す。φはハロゲン原子又は低級アル
キル基で置換されているか、もしくは非置換のフ
エニル基、チエニル基を表わす。〕
で表わされる。
米国特許第3557142号公報には毒性の低い抗炎
症作用を有するアシル化インドール誘導体が記載
されているが、かかるアシル化インドール誘導体
はN―置換基としてカルボキシル基あるいはエス
テル基を有するものである。英国特許第959203号
公報には、抗アレルギー作用を有するアシル化イ
ンドール誘導体が記載されているが、かかるアシ
ル化インドール誘導体はN―置換基としてアミノ
アルキル基を有するものである。本願発明のアシ
ル化インドール誘導体は前述した如く、N―置換
基に、アルコール性水酸基、エーテル基、アセチ
ルオキシ基を有するものであつて、本願発明で提
供されるアシル化インドール誘導体は前記公報等
に記載されたそれとは明らかに異なるものであ
り、従来全く知られていない新規化合物であり、
本願発明のアアシル化インドール誘導体は、極め
て優れた血小板凝集阻止作用という特異な薬理作
用を有し、また抗炎症作用、線溶活性作用をも有
する化合物として期待され、それ故、医薬品とし
て極めて有用なものである。
本発明で提供される上記式〔〕で表わされる
アシル化インドール誘導体において、R1は水素
原子、低級アルキル基又はアセチル基を表わす。
かかる低級アルキル基としては、例えばメチル
基、エチル基、プロピル基、イソプロピル基、ブ
チル基等が挙げられる。これらのなかでも特に
R1が水素原子、メチル基、アセチル基のアシル
化インドール誘導体は優れた血小板凝集阻止作用
を有する。
R2は水素原子又はメチル基を表わす。
R3及びR4は同一もしくは異なり、水素原子、
ハロゲン原子、低級アルキル基又は低級アルコキ
シ基を示す。ハロゲン原子としては例えばフツ
素、塩素、臭素、ヨウ素が挙げられる。低級アル
キル基としては、例えばメチル基、エチル基、プ
ロピル基、イソプロピル基、ブチル基等が挙げら
れる。低級アルコキシ基としては、例えばメトキ
シ基、エトキシ基、プロパノキシ基、イソプロパ
ノキシ基、ブトキシ基が挙げられる。
これらのなかでも、R3及びR4は水素原子が好
ましい。
φはハロゲン原子又は低級アルキル基で置換さ
れているか、もしくは非置換のフエニル基、チエ
ニル基を表わす。ハロゲン原子としては、例えば
フツ素、塩素、臭素原子などが挙げられる。低級
アルキル基としては、例えばメチル基、エチル
基、プロピル基、ブチル基などが挙げられる。
これらのなかでは特にφがフエニル基のアシル
化インドール誘導体が優れた血小板凝集阻止作用
を示す。
本発明のアシル化インドール誘導体において、
R2がメチル基のとき、N―置換基のα―炭素原
子は不斉炭素原子であり、本発明においてはこれ
らのラセミ混合物はもちろん、光学異性体のいず
れも包含する。
このような本発明のアシル化インドール誘導体
として、具体的には例えば以下の化合物が挙げら
れる。
(1) 3―ベンゾイル―1―(2―ヒドロキシエチ
ル)―2―メチルインドール,
(2) 3―ベンゾイル―1―(1―ヒドロキシ―2
―プロピル)―2―メチルインドール,
(3) 3―ベンゾイル―1―(2―メトキシエチ
ル)―2―メチルインドール,
(4) 3―ベンゾイル―1―(1―メトキシ―2―
プロピル)―2―メチルインドール,
(5) 1―(2―アセトキシエチル)―3―ベンゾ
イル―2―メチルインドール,
(6) 1―(1―アセトキシ―2―プロピル)―3
―ベンゾイル―2―メチルインドール
などである。
しかして、本発明で提供される前記式〔〕で
表わされるアシル化インドール誘導体において、
R1が低級アルキル基又はアセチル基であるアシ
ル化インドール誘導体は、下記式〔〕
〔式中、R1′は低級アルキル基又はアセチル基
を示し、R2,R3,R4及びφは前記定義に同じ。〕
で表わされるテトラヒドロアシル化インドール誘
導体を脱水反応に付することによつて製造され
る。
また、前記式〔〕で表わされるアシル化イン
ドール誘導体において、R1が水素原子であるア
シル化インドール誘導体は、下記式〔′〕
〔式中、R1″はアセチル基を示し、R2,R3,R4
及びφは前記定義に同じ。〕
で表わされるテトラヒドロアシル化インドール誘
導体を脱水素反応に付し、次いで加水分解するこ
とによつて製造される。
従つて、本発明によれば次の如きアシル化イン
ドール誘導体の製造法が提供される。
すなわち、前記式〔〕で表わされるテトラヒ
ドロアシル化インドール誘導体を脱水素反応に付
することを特徴とする下記式〔′〕
〔式中、R1′は低級アルキル基又はアセチル基
を示し、R2,R3,R4及びφは前記定義に同じ。〕
で表わされるアシル化インドール誘導体の製造
法、並びに前記式〔′〕で表わされるテトラヒ
ドロアシル化インドール誘導体を脱水素反応に付
し、次いで加水分解することを特徴とする下記式
〔″〕
〔式中、R2,R3,R4及びφは前記定義に同
じ。〕
で表わされるアシル化インドール誘導体の製造法
である。
ここで使用する前記式〔〕又は〔″〕で表
わされるテトラヒドロアシル化インドール誘導体
は、いかなる方法によつて得られるものを用いて
もよく、例えば本発明者らが別途に提案した如
く、下記式〔〕
〔式中、R1,R2,R3及びR4は前記定義に同
じ。〕
で表わされるテトラヒドロインドール誘導体と、
下記式〔〕
φ―COOH ……〔〕
〔式中、φは前記定義に同じ。〕
で表わされるカルボン酸の酸無水物又は酸ハライ
ドとを酸触媒の存在下に縮合反応することによつ
て製造することができる。
前記式〔〕又は〔′〕で表わされるテトラ
ヒドロアシル化インドール誘導体の脱水素反応
は、通常使用される脱水素剤もしくは脱水素触媒
を用いて行われる。脱水素剤としては、例えば硫
黄、2,3―ジクロル―5,6―ジシアノ―p―
ベンゾキノン、2,3,5,6―テトラクロル―
p―ベンゾキノン、3,4,5,6―テトラクロ
ル―o―ベンゾキノン等が挙げられる。又、脱水
素触媒としては、例えばセレン、パラジユウム炭
素、白金、ロジユウム等が挙げられる。反応は無
溶媒下においても進行するが、場合によつては円
滑に進行させるために有機溶媒を用いてもよい。
かかる有機溶媒体としては通常用いられる不活性
溶媒を使用することができる。具体的に例示すれ
ば、テトラヒドロフラン、ジオキサン、1,2―
ジメトキシエタン、ジエチレングリコールジメチ
ルエーテル、ジエチレングリコールジエチルエー
テル、ジエチレングリコール、ジブチルエーテル
等のエーテル類、ベンゼン、トルエン、キシレ
ン、クロルベンゼン、ニトロベンゼン、ナフタリ
ン、キノリン等の芳香族炭化水素、クメン、デカ
リン等の炭化水素等が挙げられる。
本発明において用いられる脱水素触媒及び脱水
素剤は式〔〕の原料のテトラヒドロアシル化イ
ンドール誘導体に対して0.1〜20倍モル、好まし
くは0.5〜5倍モル用いられる。また、不活性有
機媒体の使用量は、反応を円滑に進行させるに十
分な量があればよく、通常は原料の1〜100倍容
量、好ましくは2〜20倍容量あればよい。
反応温度は0゜〜500゜の範囲で行われるが、
好ましくは50〜300℃の範囲である。反応の終点
はガスクロマトグラフイー、薄層クロマトグラフ
イーで反応を追跡することによつて決定される。
反応時間は反応温度や脱水素剤や脱水素触媒によ
つて異るが、通常30分〜72時間で反応は終結す
る。
反応後、目的物を単離、精製するには、カラム
クロマトグラフイー、薄層クロマトグラフイー、
蒸留、再結晶などの精製手段を用いて行うことが
できる。
このようにして、本発明で提供される前記式
〔〕で表わされるアシル化インドール誘導体の
うち、R1が低級アルキル基、アセチル基、アセ
チル基であるアシル化インドール誘導体、すなわ
ち、前記式〔′〕で表わされるものが得られ
る。
本発明で提供される前記式〔〕で表わされる
アシル化インドール誘導体のうち、R1が水素原
子であるアシル化インドール誘導体は、前記式
〔″〕で表わされるテトラヒドロアミル化インド
ール誘導体の前記の如き脱水素反応の後に、加水
分解反応に付することによつて得られる。この加
水分解反応は、それ自体公知の反応であり、一般
には苛性ソーダ、苛性カリの如きアルカリ、又は
塩酸の如き鉱酸、好ましくはアルカリの存在下に
メタノール、エタノールの如きアルコール中で、
場合によつてはテトラヒドロフラン、ジオキサン
の如きエーテル系溶媒の共存下で通常0〜80℃の
温度で処理することにより行なわれる。反応後の
目的物の単離精製は前述したと同様の方法によつ
て行なわれる。かくして本発明で提供される前記
式〔〕で表わされるアシル化インドール誘導体
のうち、R1が水素原子であるアシル化インドー
ル誘導体、すなわち前記式〔″〕で表わされる
ものが得られる。
また、本発明のアシル化インドール誘導体は下
記式〔〕
〔式中、Xはハロゲン原子、R1及びR2は前記
定義に同じ。〕
で表わされる化合物を、酸受容体の存在下で、下
記式〔〕
〔式中、φ,R2及びR3は前記定義に同じ。〕
で表わされる化合物と反応せしめることによつて
も製造することが出来る。ここで上記式〔〕に
おけるXとしては例えば臭素原子、塩素原子が挙
げられる。酸受容体としては、水素化ナトリウ
ム、水素化カリウム等の水素化アルカリ金属が好
ましいものである。
かかる反応は、ベンゼン、トルエン、キシレ
ン、ジメチルホルムアミド等の不活性な有機溶媒
中20〜150℃の温度範囲で実施することができ
る。
しかして、本発明の上記式〔〕で表わされる
アシル化インドール誘導体、なかでも特に下記式
〔〕
〔式中、R1′は水素原子、低級アルキル基又は
アセチル基、R2′は水素原子又は低級アルキル
基、R3,R4は同一もしくは異なり水素原子、ハ
ロゲン原子、低級アルキル基又は低級アルコキシ
基を示す。φ′はフエニル基又はチエニル基を示
す。〕
で表わされるアシル化インドール誘導体は、血小
板凝集阻止作用に優れ、それ故これらの化合物
は、人間を含めた哺乳動物において、血小板凝集
を抑制し、血栓形成を阻止又は予防することが望
まれる場合に投与される。
例えば、これらの化合物は心臓血管系の硬塞の
予防、術後血栓症の予防、外科手術後の血管の血
栓硬塞の予防およびアテローム性動脈硬化症、動
脈硬化症等の予防又は治療に有用である。
また、老人病患者における脳の虚血発作の予防
および心筋層の硬塞と卒中発作後の再発の予防又
は治療に用いられる。
本発明のアシル化インドール誘導体の血小板凝
集阻止作用の測定は、アラキドン酸、コラーゲン
等の種々の凝集誘起剤の凝集に対する本発明のア
シル化インドール誘導体の阻害作用を、in vitro
で定量する方法により行つた。
すなわち、in vitroの方法では、最終濃度が
0.38%になるようにクエン酸ソーダでCaイオン
をキレート化したモルモツトの血小板に富んだ血
漿(PRP)200μlに、25μlのアシル化インド
ール誘導体溶液を加えて37℃で2分間プレインキ
ユベージヨンし、しかる後に25μlのアラキドン
酸ナトリウムを加えて、血小板凝集曲線をアグリ
ゴメーターで記録し、プランク(PRP+生理食塩
水+凝集誘起剤)の最大凝集度に対する検体の最
大凝集度の百分率を100より減じた値を凝集阻害
率とすることによつて測定した。
本発明のアシル化インドール誘導体は、経口的
にあるいは直腸内、皮下、筋肉内等の非経口的に
投与されうるが、好ましくは経口投与又は直腸内
投与によるのがよい。
経口投与のためには、固形製剤あるいは液体製
剤とされる。固形製剤としては、錠剤、丸剤、散
剤あるいは顆粒剤がある。このような固形製剤に
おいては、1ツまたはそれ以上の活性物質が少く
とも1ツの不活性な希釈剤、例えば、よく用いら
れる炭酸カルシウム、バレイシヨデンプン、アル
ギン酸あるいは乳酸と混合される。
製剤は常法に従つて行なわれるが、希釈剤以外
の添加剤、例えばアテアリン酸マグネシウムの如
き潤滑剤を含有していてもよい。
経口投与のための液体製剤は、薬剤的に受容さ
れる乳濁剤、溶液剤、懸濁剤、シロツプ剤あるい
はキシル剤を含み、一般的に用いられる不活性な
希釈剤、例えば水あるいは流動パラフインを含
む。この製剤は、不活性な希釈剤以外に補助剤、
例えば湿潤剤、懸濁補助剤、甘味剤、風味剤、芳
香剤あるいは防腐剤を含む。
またこの液体製剤は、ゼラチンのような吸収さ
れる物質のカプセルとしてもよい。
直腸内投与のための固形製剤としては、1ツま
たはそれ以上の活性物質を含み、それ自体公知の
方法により製造される坐剤が含まれる。
非経口投与の製剤は、無菌の水性あるいは非水
性溶液剤、懸濁剤または乳濁剤である。非水性の
溶剤または懸濁剤としては、例えば、プロピレン
グリコール、ポリエチレングリコール、オリーブ
油の如き植物油、オレイン酸エチルのような注射
しうる有機エステルがある。このような製剤はま
た、防腐剤、湿潤剤、乳化剤、分散剤のような補
助剤を含むことができる。これらは例えばバクテ
リア保留フイルターをとおす過、殺菌剤の配合
あるいは照射によつて無菌化できる。また無菌の
固形製剤を製造し、使用直前に無菌水または無菌
の注射用溶媒に溶解して使用することができる。
本発明の活性化合物であるアシル化インドール
誘導体の投与量は、1日、体重Kgあたり0.005〜
約200mgであり、0.01〜100mgが好ましい。
これらの投与量は、患者の病状、体重、年令あ
るいは投与経路により左右される。
以上のとおり、本発明で提供する新規なアシル
化インドール誘導体は、血小板凝集阻止作用が優
れたものである。
以下、実施例をあげ、本発明を更に具体的に示
す。
実施例 1
3―ベンゾイル―1―(1―メトキシ―2―プ
ロピル)―2―メチルインドールの合成
無水安息香酸4.52g(20ミリモル)と4,5,
6,7―テトラヒドロ―1―(1―メトキシ―2
―プロピル)―2―メチルインドール2.07g(10
ミリモル)と52%沃化水素酸0.5mlを、窒素気流
下、120℃に2時間加熱撹拌した。反応後、反応
混合物を1N水酸化ナトリウム溶液50mlを加え、
室温で2時間撹拌した。次いで酢酸エチル50mlず
つ2回抽出した。酢酸エチル層を合せ、水100
ml、飽和食塩水100mlの順で洗浄した。無水硫酸
ナトリウムで酢酸エチル層を乾燥した後、溶媒を
減圧留去して得られたかつ色油状生成物をシリカ
ゲル200gのカラムクロマトグラフイーに付し
た。酢酸エチル―ベンゼン=1:20で展開し、目
的の3―ベンゾイル―4,5,6,7―テトラヒ
ドロ―1―(1―メトキシ―2―プロピル)―2
―メチルインドール1.92gを油状物として得た。
収率62%

メチルインドール1.55g(5ミリモル)と2,3
―ジクロル―5,6―ジシアノ―p―ベンゾキノ
ン2.72g(12ミリモル)とをベンゼン20mlに溶か
し、24時間加熱還流した。室温に放冷後、析出し
た結晶をロ去し、ロ液を減圧濃縮した。得られた
黒かつ色の油状物をシリカゲル100gのカラムク
ロマトグラフイーに付し、酢酸エチル―ベンゼン
=1:25で展開し、目的の3―ベンゾイル―1―
(1―メトキシ―2―プロピル)―2―メチルイ
ンドール650mgの油状物を得た。収率42%

実施例 2
1―(1―アセトキシ―2―プロピル)―3―
ベンゾイル―2―メチルインドールの合成
1―(1―アセトキシ―2―プロピル)―4,
5,6,7―テトラヒドロ―2―メチルインドー
ル23.2g(0.099モル)と無水安息香酸44.8g
(0.198モル)と52%沃化水素酸1mlを窒素気流下
145℃に2時間加熱撹拌した。室温に放冷後、酢
酸エチル300mlに反応混合物を溶かし、1N苛性ソ
ーダ溶液100ml、水100ml、飽和食塩水100mlの順
で洗浄した。有機層を芒硝乾燥後、溶媒を減圧留
去して得られた黒かつ色油状物をシリカゲル850
gのカラムクロマトグラフイーに付し、ベンゼ
ン:酢酸エチル=19:1で展開し、目的の1―
(1―アセトキシ―2―プロピル)―3―ベンゾ
イル―4,5,6,7―テトラヒドロ―2―メチ
ルインドールを油状物として29.6g得た。収率88
%

1N,苛性ソーダ溶液100ml、水200ml、飽和食塩
水で洗い、無水硫酸マグネシユウムで乾燥した。
溶媒を減圧留去して得られた黒かつ色油状物をシ
リカゲル150gのカラムクロマトグラフイーに付
し、ベンゼン:酢酸=20:1で展開し、目的の1
―(1―アセトキシ―2―プロピル)―3―ベン
ゾイル―2―メチルインドールを油状物として
1.44g得た。収率25%

実施例 3

プロピル)―2―メチルインドールの合成
1―(1―アセトキシ―2―プロピル)―3―
ベンゾイル―2―メチルインドール597mg(1.78
ミリモル)をメタノール10mlと1N、苛性ソーダ
溶液5mlに溶かし、室温で3時間撹拌した。メタ
ノールを減圧留去後、残査を酢酸エチル50mlに溶
かし、水50ml、飽和食塩水50mlで洗浄した。芒硝
乾燥後、溶媒を減圧留去し、目的の3―ベンゾイ
ル―1―(1―ヒドロキシ―2―プロピル)―2
―メチルインドールを油状物として520mg得た。
収率100%

実施例 4
3―ベンゾイル―1―(2―メトキシエチル)
―2―メチルインドールの合成
4,5,6,7―テトラヒドロ―1―(2―メ
トキシエチル)―2―メチルインドール1.2g
(6.2ミリモル)と無水安息香酸2.8g(12.4ミリモ
ル)と52%沃化水素酸0.15mlとを窒素気流下、
130℃に3.5時間加熱撹拌した。反応混合物を室温
に放冷後、酢酸エチル80mlに溶かし、飽和重水
100ml、水100ml、飽和食塩水100mlの順で洗浄
し、芒硝乾燥した。溶媒を減圧留去して得られた
黒かつ色油状物をシリカゲル50gのカラムクロマ
トグラフイーに付し、ベンゼン:酢酸エチル=
20:1で展開し、油状の3―ベンゾイル―4,
5,6,7―テトラヒドロ―1―(2―メトキシ
エチル)―2―メチルインドールを1.05g得た。
収率57%

し、氷冷下撹拌しながら水素化ナトリウム90mg

え、撹拌を一夜続けた。ジメトキシエタンを減圧
留去後酢酸エチル50mlと反応物を溶かし水50ml飽
和食塩水50mlの順で洗浄し、芒硝乾燥した。溶媒
を減圧留去して得られた油状生成物をシリカゲル
15gのカラムクロマトグラフイーに付し、ベンゼ
ン:酢酸エチル:20:1で展開し、各フラクシヨ
ンを10mlずつ分取し、フラクシヨン7〜14に目的
のエチルエーテル化合物を油状物として270ml得
た。収率56%このものの物性データは以下のとお

実施例12 in vitro血小板凝集阻止作用
本発明のアシル化インドール誘導体のin vitro
における血小板凝集阻止作用をモルモツトPRPを
用いて測定した。
結果は血小板凝集の50%阻止濃度(IC50)で示
した。凝集剤はアラキドン酸ナトリウムをを用い
た。
〔PRP,薬物,凝集剤の調製〕
(1) PRP(富血小板血漿)の調製
体重350〜600gに雄性ハートレー系モルモツ
トより心臓穿刺法によつてクエン酸血(3.8%
クエン酸ソーダ1容と血液9容の割合)を採取
した。
得られたクエン酸血を1000rpmで10分間室温
で遠心し、上清(PRP)を分離した。
得られたPRPは室温に保存し、なるべく早く
使用するようにし、調整後4時間を経過したも
のは使用しなかつた。
(2) 薬物の調整
被検薬物は一般的には10mg/mlとなるように
ジメチルスルフオキシドに溶解し、生理食塩水
で稀釈して2500μg/ml,1000μg/ml,750
μg/ml,500μg/ml,250μg/ml,100μ
g/mlの溶液を各1mlづつ調整した。
但し、フリーのカルボキシル基を有する化合
物は0.1MNaHCO3にて溶解して10mg/mlのナ
トリウム塩溶液とし、同様に生理食塩水にて稀
釈して被検薬物溶液を調整した。
血小板凝集阻止試験の結果、100μg/ml
(終濃度に換算して10μg/ml)でも薬物が血
小板の凝集を完全に抑制している場合には、さ
らにその薬物溶液を生理食塩水にて稀釈して行
き(75μg/ml,50μg/ml,25μg/ml,10
μg/ml)それぞれの稀釈液についてさらに血
小板阻止試験を行つた。
(3) 凝集剤の調整
シグマ社製アラキドン酸(99% pure)を
0.1MNaHCO3にて溶解して3.3mMアラキドン
酸ナトリウム塩溶液を調整する。これを生理食
塩水にて稀釈して1mMとして実験に使用し
た。3.3mM液は保存母液として冷蔵庫中に保
存し、1mMは使用のつどこの母液から新規調
整した。
〔血小板凝集阻止試験〕
(1) ブランクの血小板凝集度
アグリゴメーターの37℃のキユベツト中であ
らかじめ温めておいた200μのPRPに25μ
の生理食塩水と25μの凝集剤溶液を加えて血
小板を凝集させ、その凝集曲線をブライストン
社製アグリゴメーターにて3分間記録した。こ
の血小板凝集における最大凝集度をブランクの
最大凝集度とした。
(2) アラキドン酸ナトリウム塩の凝集強度の調整
200μのPRPに25μのイブプロフエン溶
液(250μg/ml或いは100μg/ml)を加え、
上記(1)と同様にして2分間スターラーで撹拌し
ながらアグリゴメーターキユベツト中でプレイ
ンキユベーシヨンし、しかる後に25μの
1mMアラキドン酸ナトリウム液を加えて上記
(1)と同様にして血小板凝集曲線を記録し、曲線
から、最大凝集度を算出し凝集阻害率を下記式
にて算出した。

−イブプロフエン添加系の最大凝集度/プランクの最
大凝集度×100
この時250μg/mlのイブプロフエンを添加
した系では阻害率が50%以上、100μg/mlの
イブプロフエン添加系では阻害率が50%以下に
なつている事を確認し、なつていない場合はア
ラキドン酸ナトリウム塩液が適当濃度でないの
で濃度の調整をやりなおし、再試験を行ない条
件を合わすようにした。
(3) 血小板凝集阻止試験
200μのPRPに25μの被検薬物溶液を加
え、上記(2)と同様にして37℃2分間プレインキ
ユペーシヨンした後、上記(2)で濃度調整したア
ラキドン酸ナトリウム液25μを加えて凝集曲
線を3分間記録し、その時間内における血小板
の最大凝集度を測定して阻害率を下記式にて算
出した。阻害率が50%を越す薬物の最底濃度を
IC50値として示した。

結果は第1表に示した通りである。
なお、被検薬物の経口投与による急性毒性試験
をマウスを用いて実施した。LD50値は3000〜
1000mg/Kg以上であつた。亜急性毒性試験として
ラツトに7日間20,100,500mg/Kgを連続経口投
与した結果、100mg/Kgまで異常が認められなか
つた。
