くはこの閉環ラクトン体はいずれもコレステロー
ル合成阻害作用を有するが、特にM−4のコレス
テロール合成阻害作用は顕著であり、ML−236B
ラクトンの10倍以上の阻害作用を示す。しかしな
がら、M−4はML−236Bラクトンを投与した動
物の代識産物としてのみ得られていたものであ
り、量産性に乏しく、従つて本物質の経済的な生
産方法を検討していたところ、微生物の作用によ
り、ML−236B化合物を式()を有するML−
236B誘導体もしくはこの閉環ラクトン体に変換
せしめ得ることを見出し、本発明を完成した。 ML−236BラクトンおよびML−236Bカルボン
酸自体は既知物質であり、青カビの一種ペニシリ
ウム・チトリヌムの代識産物より分離、精製され
た物質で、次式に示される化学構造を有してお
り、実験動物から分離した酵素系や培養細胞系に
おいてコレステロールの生合成をその律速酵素の
3−ヒドロキシ−3−メチルグルタリル・コエン
ザイムAリダクターゼと競合することにより阻害
し、動物の個体レベルにおいても強力な血清コレ
ステロールの低下作用を示すことが知られている
(特開昭50−155690号、ジヤーナル・オブ・アン
チビオテイクス 29巻 1346〜1348頁 1976

せしめることによつても達成される。 この場合、微生物(生菌)培養では培養条件に
よつてカルボン酸型、ラクトン型、アルカリ金属
塩または低級アルキルエステル型として生成す
る。また休止菌体系および酵素抽出液(無細胞抽
出液)ではアルカリ金属塩として得られる。 あるいは、ML−236B化合物から微生物変換に
よつて得られるカルボン酸型をそのまま、または
ラクトン型に変換した後、所望により化学的常
法、例えばジアゾアルカンまたは塩を形成する物
質と処理することにより、これらを低級アルキル
エステル型もしくはアルカリ金属塩として得るこ
ともできる。また、同様に例えばアルカリ金属塩
を所望により化学的常法によつて処理することに
より、低級アルキルエステル型に変換することも
できる。 次に実施例を示す。 実施例 1 下記組成の培地100mlを含有する500ml容坂口フ
ラスコ20本にアブシテイア・コエルレア
IFO4423を植菌し、26℃、120s.p.m.で振盪培養
し、2日後、ML−236B Na塩を最終濃度で0.05
%になるように添加して更に5日間26℃、120s.

で洗浄し、硫酸ナトリウムで脱水後、減圧乾固し
た。残渣をローバー・カラム(メルク社製RP−
8サイズA)を用い、25%アセトニトリルで溶出

イソM−4′とM−4は双方共薄層クロマトグラフ
イー(TLC)(プレート;メルク社製シリカゲル
Art5715溶媒;ベンゼン:アセトン:酢酸=
50:50:3)によりRf値0.45を示す。上記抽出
液を飽和食塩溶液で洗浄し、ジアゾメタンのエー
テル溶液を加え、30分放置後、減圧乾固した。残
渣をローバー・カラム(メルク社製Si60、サイズ
A)にかけ、ベンゼン:酢酸エチル=1:1の系
で精製するとイソM−4′メチルエステルを含む区
分(活性区分)とM−4メチルエステルを含む
区分(活性区分)に分けられる。活性区分は
200mgが得られた。活性区分をローバー・カラ
ム(メルク社製RP−8サイズA)を用い、35%
アセトニトリルで溶出し、イソM−4′メチルエス
テルの精製標品78mgが無色油状体として得られ
た。なお本操作におけるジアゾメタンに代えて、
適当なジアゾアルカンを使用すると該当するイソ
M−4′のアルキルエステルが得られる。イソM−
4′メチルエステルは次の特性を有する。 (1) NMRスペクトル 重クロロホルム中、内部標準にTMSを使用
して、100MHzで測定したNMRスペクトルを第
1図に示す。 (2) マススペクトル N・O−ビス(トリメチルシリル)トリフル
オロアセトアミドでシリル化した後、日本電子
製D−300型を用いて測定した。 M/e:654(M+)、552、462、372、272、

185.3mgが得られた。活性区分をローバー・カ
ラム(メルク社製RP−8、サイズA)を用い、
35%アセトニトリルで溶出し、M−4メチルエス
テルの精製標品20mgが無色油状体として得られ
た。なお本操作におけるジアゾメタンに代えて、
適当なジアゾアルカンを使用すると該当するM−
4のアルキルエステルが得られる。 M−4メチルエステルは次の特性を有する。 (1) NMRスペクトル 重クロロホルム中内部基準にTMSを使用し

変換反応液1.9を得た。変換反応液を過し、
液をトリフルオロ酢酸でPH3.0に調整した。次
いで、1の酢酸エチルで3回抽出するとM−3
とイソM−4′とM−4を含む区分が得られる。上
記抽出液を飽和食塩水で洗浄し、硫酸ナトリウム
で脱水後、触媒量のトリフルオロ酢酸を添加して
ラクトン化した。次いで上記抽出液を5%炭酸水
素ナトリウム水溶液で洗浄し、硫酸ナトリウムで
脱水後、減圧乾固した。残渣をローバー・カラム
(メルク社製Si60サイズA)にかけ酢酸エチルで
溶出しイソM−4′ラクトン区分とM−4ラクトン
区分を分離採取した。イソM−4′ラクトン区分を
ベンゼン:酢酸エチル(1:1)の系で精製し、
イソM−4′ラクトン198mgが得られた。更にこれ
をローバー・カラム(メルク社製RP−8サイズ
A)を用い、35%アセトニトリルで溶出し、イソ
M−4′ラクトンの精製標品82mgが得られた。 イソM−4′ラクトンは次の特性を有する。 (1) NMRスペクトル 重クロロホルム中、内部標準にTMSを使用
して100MHzで測定したNMRスペクトルを第3
図に示す。 (2) 紫外部吸収スペクトル(メタノール溶液) λnax(nm):229、234.8、244.5 (3) 赤外部吸収スペクトル(薄膜法) 第4図に示す。 実施例 14 カニンガメラ・エチヌラータ IFO 4445を用
い、実施例13と同様に操作してイソM−4′ラクト
ンを得た。 実施例 15 シンセフアラストラム・ラセモーサム IFO
4814を用い、実施例13と同様に操作してイソM−
4′ラクトンを得た。 実施例 16 シンセフアラストラム・ラセモーサム IFO
4828を用い、実施例13と同様に操作してイソM−
4′ラクトンを得た。 実施例 17 ストレプトマイセス・ロゼオクロモゲナス
NRRL 1223を用い、実施例13の方法に従つてイ
ソM−4′ラクトンを得た。 実施例 18 実施例13と同様に操作して、ローバー・カラム
で分離したM−4区分を同様に精製してM−4ラ

精製し、再度ローバー・カラム(メルク社製RP
−8サイズA)にかけ、35%アセトニトリルで溶
出し、M−3メチルエステルの精製標品を得た。
なお本操作におけるジアゾメタンに代えて適当な
ジアゾアルカンを使用すると該当するM−3のア
ルキルエステルが得られる。 M−3メチルエステルの物性値は次の通りであ

後、50%アセトンでM−4ナトリウム塩、イソM
−4′ナトリウム塩およびM−3ナトリウム塩を含
有する区分を溶出し、凍結乾燥品830mgを得た。
