Description
産業上の利用分野
本発明は抗微生物剤組成物、さらに詳しくは、
銅イオン触媒を用いるエンジオール化合物の酸化
反応に基づく抗微生物作用を応用する種々の殺菌
剤や、抗真菌作用、抗菌作用、抗ウイルス作用を
必要とする製剤として有用な組成物に関する。
従来の技術
アスコルビン酸等のエンジオール化合物を、水
の存在下で少量の銅化合物と接触させると、銅イ
オン触媒の作用でエンジオール化合物の酸化反応
が起こり、それにより、抗真菌作用、抗菌作用、
抗ウイルス作用のような抗微生物作用が発現する
ことが知られている(例えば、米国特許第
3065139号)。しかしながら、この反応は非常に急
速に進行し、その抗微生物作用がじきに低下して
しまい、この抗微生物作用を商業的に実用化する
ことは非常に困難である。
そこで、従来より、この抗微生物作用を商業的
に実用化するため、その安定化、持続化に関して
種々検討がなされている。例えば、米国特許第
3681492号にはアミンを用いて該酸化反応の速度
を抑制し、抗微生物作用を持続させる抗微組成物
が開示されている。また、特公昭56―5971号に
は、該抗微生物作用を応用したコンタクトレンズ
の殺菌法が開示されており、ここでは、エンジオ
ール化合物と銅化合物を一緒に、または個別に、
水を含まない剤形に処方し、使用時、水と混合す
る殺菌剤が用いられている。さらに、該殺菌剤に
は、塩化アンモニウムや米国特許第3681492号に
おけると同様な反応速度抑制のためのアミンを添
加してもよい旨記載されている。また、特公昭57
―52323号にはエンジオール化合物、銅化合物お
よび、所望により、前記のような反応速度を抑制
する物質からなる、自由水を含まないエアゾル形
として、該抗微生物作用の安定化を図る殺菌剤が
開示されている。
発明が解決しようとする問題点
しかしながら、従来の該抗微生物作用の安定
化、持続化策は必ずしも満足すべきものではな
い。すなわち、アミン等を用いて酸化反応速度を
抑制しても、抗微生物作用を商業的に実用化でき
るほどに持続させることは、ほとんど不可能であ
る。また、該反応がラジカル反応であり、少量の
水の存在でも進行するため、たとえ水を含有しな
い剤形としても、その状態を完全に保持しない限
り、抗微生物作用の安定化、持続化は困難であ
り、一方、完全に水分のない状態を保持すること
は商業的な実用化の観点からは極めて困難であ
る。さらに、エンジオール化合物と銅化合物を個
別の剤形に処方することは使用上の煩わしさや、
応用範囲が制限されるなどの問題がある。
このような事情にかんがみ、本発明は、銅イオ
ン触媒を用いるエンジオール化合物の酸化反応に
基づく抗微生物作用を商業的に充分に実用化でき
るように安定化、持続化させ、すぐれた抗微生物
作用を有する抗微生物剤組成物を提供するために
なされたものである。
問題点を解決するための手段
本発明者らは、銅イオン触媒を用いるエンジオ
ール化合物の酸化反応に基づく抗微生物作用の安
定化、持続化について種々検討した結果、その作
用機序は明らかでないが、炭素数6〜10の脂肪族
ジカルボン酸を共存させると、水の配合の有無に
かかわりなく、また、エンジオール化合物と銅化
合物を個別の剤形に処方しなくても、特異的に抗
微生物作用の安定化、持続化が図れ、すぐれた抗
微生物剤組成物が得られることを見出だした。す
なわち、本発明は、アスコルビン酸、ジヒドロキ
シマレイン酸、ジヒドロキシフマル酸、レダクチ
ン酸およびこれらの誘導体からなる群から選ばれ
るエンジオール化合物、塩化第2銅、硫酸銅、こ
れらの水和物およびオレイン酸銅からなる群から
選ばれる銅化合物および炭素数6〜10の脂肪族ジ
カルボン酸を必須成分としてなる抗微生物剤組成
物を提供するものである。
用いるエンジオール化合物は、アスコルビン
酸、ジヒドロキシマレイン酸、ジヒドロキシフマ
ル酸、レダクチン酸およびこれらの誘導体(例え
ば、塩、エーテル、エステル誘導体)からなる群
から選ばれる。エンジオール化合物の使用量は実
際に用いる化合物の種類、組成物の剤形等に応じ
て適宜選択できるが、一般に、組成物全量に対し
て0.01〜10%(重量%、以下同じ)の範囲で充分
である。
用いる銅化合物は、該抗微生物作用発現の触媒
となる銅イオンを生成する化合物であり、塩化第
二銅、硫酸銅、これらの水和物およびオレイン酸
銅からなる群から選ばれる。銅化合物の使用量も
実際に用いる化合物の種類、組成物の剤形等に応
じて適宜選択できるが、一般に、組成物全量に対
して0.0001〜0.1%の範囲で充分である。
炭素数6〜10の脂肪族ジカルボン酸、すなわ
ち、アジピン酸、ピメリン酸、スベリン酸、アゼ
ライン酸、またはセバシン酸は組成物全量に対し
て0.01〜10%の範囲で配合され、一般に、エンジ
オール化合物と同量またはそれ以下でよい。脂肪
族ジカルボン酸の量は少なすぎると効果が期待で
きず、また、多すぎると、脂肪族ジカルボン酸に
よる副作用が生じたり、また、水に溶けにくいた
めに沈澱が生じるなどの問題がでてくる。
かくして、本発明の組成物は通常の製剤化技術
により、錠剤、顆粒のような固体の剤形または水
形もしくは非水系の溶液、分散液、乳液、ローシ
ヨン、ペースト、ゲル、軟膏、エアゾルなどの剤
形にすることができ、他の成分は特に限定するも
のではない。本発明の固形の組成物は、例えば、
水に溶解することにより、抗微生物作用を発揮
し、非水系のものは水との混合または吸水、湿潤
により抗微生物作用を発揮する。水系のものは直
接適用することにより、抗微生物作用を生じる。
これらの組成物は、例えば、コンタクトレンズ洗
浄殺菌剤、含嗽剤、義歯洗浄殺菌剤、トイレやバ
ス用の洗浄殺菌剤、制汗用スプレー、フツトスプ
レー、食器等の殺菌剤、皮膚外用殺菌剤、頭髪用
剤、軟膏、クリーム、石鹸等の形態の薬用化粧料
などとして用いることができる。
実施例
つぎに実施例を挙げて本発明をさらに詳しく説
明する。
実施例 1
つぎの処方により、含嗽用顆粒を製造した。
成 分 %
アスコルビン酸 30.0
ピメリン酸 15.0
重炭酸ナトリウム 30.0
リンゴ酸 23.0
硫酸銅・5水和物 0.2
香 料 1.8
常法に従い、これらの成分をエタノールで練合
し、湿式造粒した。得られた顆粒は1gづつ分包
し、含嗽時、1包を水30〜100mlに溶解して用い
る。
実施例 2
つぎの処方により、コンタクトレンズ殺菌用錠
剤を製造した。

アスコルビン酸 30.0
スベリン酸 10.0
重炭酸ナトリウム 14.0
硫酸銅・5水和物 0.2
塩化ナトリウム 0.6
滑沢剤、中和剤 適 量
乳 糖 残 部
常法に従い、直接打錠法で1個100mgの錠剤を
製造した。この1錠を10mlの水に溶解して、コン
タクトレンズの殺菌に用いる。
実施例 3
つぎの処方により、制汗用エアゾルを製造し
た。

アゾルを製造した。
実施例 4
つぎの処方により、ゲル剤の剤形の皮膚外用殺
菌剤を製造した。

常法に従い、A成分およびB成分を各々、混
合、溶解し、両溶液を合し、撹拌しながら硫酸銅
を添加、混合してゲル剤を得た。
発明の効果
各種の脂肪族ジカルボン酸の抗微生物作用安定
化効果を試験するために、第1表に示した種々の
脂肪族ジカルボン酸およびアスコルビン酸を、
各々、濃度1%となるように、また、硫酸銅・5
水和物を濃度0.01%となるように20%アルコール
水溶液に溶解し、PHを6.0に調整し、密栓して40
℃で14日間放置後、その抗菌性を測定した。対照
として、脂肪族ジカルボン酸無添加の溶液、ジカ
ルボン酸の代わりに、エンジオール化合物の酸化
反応速度を抑制することが知られている塩化アン
モニウムを添加した溶液およびアスコルビン酸無
添加の溶液についても、同様に抗菌性を測定し
た。
抗菌性の測定は、スタフイロコツカス・アウレ
ウス(Staphylococcus aureus)の菌体を最終濃
度1×107個/mlとなるように各溶液に加え、添
加直後および15分間放置後に寒天平板に塗布し、
37℃で18時間インキユベーシヨン後、菌の生育の
有無をつぎの基準で判定することにより、行なつ
た。

That is, the present invention provides enediol compounds selected from the group consisting of ascorbic acid, dihydroxymaleic acid, dihydroxyfumaric acid, reductinic acid, and derivatives thereof, cupric chloride, copper sulfate, hydrates thereof, and copper oleate.
