【発明の詳細な説明】
多孔質ヒドロキシル燐灰石材料
本発明は多孔質ヒドロキシル燐灰石材料、および医薬、歯科技工、並びに家畜治
療薬に於けるその応用に関するものである。
米国特許第3.929.971号により、特定な微細構造を持ち、生命物質(b
i oma t e r i a 1) 、例えば人工器官物質、骨移植など
として用いられる合成多孔質ヒドロキシル燐灰石材料が知られている。この以前
から知られたヒドロキシル燐灰石材料は海中動物、特に珊瑚およびヒトデの炭酸
塩骨格から造られる。
その製造に用いられる原料物質のために、特定な多孔性を有し、特定な形を持つ
新規な多孔質ヒドロキシル燐灰石材料を提供するのが本発明の一つの目的である
。
この目的のために、本発明によれば、石灰で外皮を形成した藻のカルシウム富化
基礎骨格から造られる多孔質ヒドロキシル燐灰石材料が提供される。
本発明の好ましい実施態様によれば、多孔質ヒドロキシル燐灰石材料の孔は骨髄
のような再吸収可能なまたは骨を誘発する物質で満たされている。
本発明による多孔質ヒドロキシル燐灰石材料は特に人工骨、人工歯根、安定化物
質、人工中空体、補助物質、裏打ち物質、または欠陥充填物質として用いられる
。
ヒドロキシル燐灰石祠料の製造のために、本発明によれば、全ての藻は、大洋の
沿海地方および亜沿岸帯からの藻で、ベンツイン(benthish)外皮生成
藻、例えばリトサムニオン(Li thothamnion)属及びリトフィラ
ム(Li thophyl lum)属を含む、特にザンゴモ科(Corall
inaceae)属、例えばコラリナオフィシナリス
(Corallina officinalis)、コラリウムルーベンス(C
oralliumrubens)、リトサムニオン力ル力ラム(Lithoth
amnion calcarum)およびミル科(Codiaceae)属、例
えばハリメダ属(Halimeda 5pec、)が用いらち、これ等は、一般
に少量ではあるが、石灰を表面に堆積する。
藻の石灰骨格を得るため、藻の有機成分を最初に取除かなくてはならない。これ
は、いわゆる[浸してふやけさせるJ (maceration)ことで実行す
ることができる。この目的のため生きているか、または乾燥した藻の材料を銅錯
塩、例えばシュヴアイツァー試薬または塩化リチウムまたはシュウ化物溶液で処
理するか、熱分解する。この様な浸してふやけさせる方法、即ち有機材料を除去
するための方法は前述の米国特許第3.929.971号に記載されていると同
様で、例えば稀薄次亜塩素酸ソーダ溶液中に於ける浸漬である。
シ二ヴアイツァー試薬または塩化リチウム溶液により浸してふやけさせるか、ま
たは熱分解後に粒状で存在する、有機物質のない残留炭酸カルシウム基質の材料
は蒸溜水で完全に洗浄される。材料の粒度は0.1乃至2ミリメートルである。
藻から得られるカルシウム富化基礎骨格は次に既知の方法でヒドロキシル燐灰石
に転化される。このヒドロキシル燐灰石への転化は、通常、熱水法または燐酸塩
浴を用いて行はれる。この目的のため、アルカリ金属燐酸塩、例えばオルト燐酸
ナトリウム、オルト燐酸カリウムとオルト燐酸アムモニウム、酸性燐酸塩および
燐酸塩混合物を用いてもよい。同様に、オルト燐酸もこの目的に用いてもよい。
炭酸カルシウムをヒドロキシル燐灰石に転化するこの熱水処理は上述の米国特許
第3.929.971号に同様に開示されている。この熱水処理は高圧高温で通
常の方法で行はれる。
この様な処理の例は石灰で外皮を形成された藻の孔の多い、ふやけた粒子を、燐
酸水素ナトリウムおよび二重に蒸溜した水と、密閉容器中で2乃至3日の間、以
下の条件で、1:1:4の割合で維持することを含む。
(イ) 300℃および500バール、あるいは(ロ) 600℃および100
0バール。
用いた物理的条件に応じて骨手術における材料の再吸収性に影響を及ぼすことが
できる。 熱水処理後、ヒドロキシル燐灰石材料は、通常、粒状を示す。この粒
状から、ち密な断片が、凝集、例えば通常の焼結法、もし必要なら圧力の適用下
に、通常の焼結促進物質の添加により製造できる。
この粒状物質からち密な断片への凝集のため、この粒状物質は、最初、成形体へ
圧縮され約4時間の予熱段階の後、これ等は1000乃至1400℃で4乃至5
時間焼成され、引続き、例えば4乃至5時間冷却される。焼結後のヒドロキシル
燐灰石本体の孔の大きさを慎重に定めるため、焼結は実験的に測定されたH2O
(二重蒸溜)の添加で行われる。
本発明によるヒドロキシル燐灰石材料の孔は再吸収可能な材料で満たされること
も出来る。この方法で医薬、歯科技工、並びに家畜治療薬に於ける、その応用に
対してのヒドロキシル燐灰石材料の適合性が改善され得る。
そして特に孔の充填によりヒドロキシル燐灰石材料で起こる感染率は低下され、
および/または骨の誘発が増加され得る。再吸収可能な材料として、凍結乾燥さ
れた異種構造のフラーゲンフィラメント、相同に凍結乾燥されたフィブリン、石
膏、骨髄やポリ乳酸調製物のような蛋白質、例えばポリジオキサノンが用いられ
る。
本発明によるヒドロキシル燐灰石材料の実施態様の利点は、米国特許第3.92
9.971号により珊瑚材料から製造されたヒドロキシル燐灰石材料と比べ、あ
る種の可能な用途に対して利益を与える粒状の形態に存する。

- (1)ヒドロキシル燐灰石に転化された石灰で外皮を形成された藻のカルシウム 富化基礎骨格から造られたた多孔質ヒドロキシル燐灰石材料。
- (2)前記材料の孔が再吸収可能な材料で満たされている請求の範囲第1項に記 載の多孔質ヒドロキシル燐灰石材料。
