【発明の詳細な説明】 本発明は優れた抗菌性を示す新規マクロライド
抗生物質に関する。 さらに詳しくは、新規マクロライド抗生物質産
生放線菌であるミクロモノスポラ・エス・ピー・
A11725菌株(Micromonospora sp.A11725)を
培地に培養し、その培養物から新規マクロライド
抗生物質A11725およびA11725を抽出分離し
た新規マクロライド抗生物質A11725および
A11725のエポキシを有機溶媒中にて塩化第一
クロムにて処理して、二重結合となした新規マク
ロライド抗生物質A11725aおよび対照として
のA11725aに関するものである。 本発明の新規マクロライド抗生物質A11725
a(以下、A11725aという)、および対照とし
てのA11725a(以下、A11725aという)は
夫々下記の物理化学的性質を有する塩基性含窒素

は第1表の通りである。 【表】 【表】 上記諸性質より本物質は、塩基性の16員環マク
ロライド系物質で、デソサミンおよび6−デオキ
シ−2.3−ジ−O−メチル−ヘキソースの各糖を
1ケ有するもので、かつ構造として式 で表わされる化学構造(ただし、R1は水素原子
または水酸基を示す)を有し、分子中にアルデヒ
ド基を有しておらず、さらにその出発物質たる新
規マクロライド抗生物質A11725およびA11725
(以下、各々A11725、A11725という)の
分子内のエポキシを二重結合に変換せしめたもの
で、A11725aはそのR1が水素原子、A11725
aはそのR1が水酸基で示される各物質であつて、
これらのことより本物質は、既知の物質に該当す
るものはなく、新規物質であると判断される。 なお本発明物質は通常医薬的に許容される鉱酸
や有機酸等との塩類の形で使用出来、例えば酒石
酸塩、クエン酸塩、コハク酸塩等の塩類の形で使
用出来る。 本発明の抗生物質は錠剤および粉末として経口
的に服用することもでき、また静脈注射により適
用することもできる。 その使用量は成人1日当り、400〜2000mg程度
でよく、グラム陽性菌例えばブドウ球菌による呼
吸器感染症に対して有効に使用される。さらに本
発明の抗生物質は飼料添加用抗生物質や動物治療
用抗生物質として利用される。 また、本発明におけるA11725およびA11725
は、その物理化学的性質等より塩基性マクロラ
イド群に属する抗生物質で既知の物質に該当する
ものはなく、新規物質と判断されるものであつ
て、A11725、A11725は例えばミクロモノス
ポラ属に属するミクロモノスポラ・エス・ピー・
A11725菌株の培養物より得られる。本菌、新規
マクロライド抗生物質産生放線菌は、富山県下新
川郡宇奈月町のじやがいも畑の土壌より分離した
放線菌であつて、ミクロモノスポラ属に属するミ
クロモノスポラ・エス・ピー・A11725と称する
もので、本菌は工業技術院微生物工業技術研究所
に微生物受託番号「微工研条寄第705号、FERM
−BP 705」〔本菌株はミクロモノスポラ・グリゼ
オルビダA11725(Micromonospora griseorubia
A11725)として菌株名が変更されている〕とし
て受託されているものである。 さらに本菌の菌学的諸性状は以下の通りであ
る。 〔〕 形態的性状 基生菌糸は長く伸長し、波状で、単純分枝をな
し、直径0.6〜0.8μで菌糸の分断はみられない。
胞子は基生菌糸より生じた短かい胞子柄の先端に
1個づつ形成され、球形、ないし卵形で大きさは
1.0〜1.5μであり、その表面はとげ状の突起を有
しており、一見コンペイトウ様である。又寒天培
地上では培地組成により未発育の気菌糸を形成し
たり、コロニー表面に黒色の胞子層を形成したり
する。 〔〕 各種培地における生育状態 各種培地上で30℃、20日培養し、観察した結果
は第2表に示した。色の表示はカラーハーモニー
マニエル(Color Harmony Manual)第4版

陰性 上記のように、本菌A11725株は分枝をもつた
基生菌糸より生じた胞子柄の先端に胞子を1個づ
つ着生し、真性の気菌糸を形成せず、中温菌であ
ることより、ミクロモノスポラ
(Micromonospora)属に属する菌株である。 以上のことより、本菌A11725株をミクロモノ
スポラ・エス・ピー・A11725(Micromonospora
sp.A11725)と称することにした。 本発明の親規抗生物質A11725およびは下
記の如くして得られる。即ち例えば上記のミクロ
モノスポラ・エス・ピー・A11725菌株を通常の
微生物の培養に使用する培地成分を含む培地にて
好気的に培養する。培地は固型培地または液状培
地が用いられるが、特に大量生産のためには液状
培地、特に水性培地が適当である。 培地成分として、炭素源にはグルコース、澱
粉、グリセリン、蔗糖、モラツセ、デキストリン
などが使用するに適する。 窒素源にはペプトン、肉エキス、大豆粉、カゼ
イン水解物などが適するが、綿実粕、コーンスチ
ープリーカー、硝酸塩、アンモニア塩も利用でき
る。 その他無機物質としてナトリウム、カリウム、
マグネシウム、カルシウム、コバルト、マンガ
ン、鉄などの陽イオンを含有する物質、および
(または)塩素、硫酸リン酸、酢酸などの陰イオ
ンを含有する物質が使用できる。更に菌の発育促
進因子として乾燥酵母、酵母エキスが使用でき
る。又培地のPHを調節するため炭酸カルシウムを
培地に加えることもできる。 その他培養中の発泡をおさえるためシリコン樹
脂、動植物油などの適当量を培地に加えることが
できる。 本発明方法を実施するに特に適する培地は培地
成分としてグルコース、デキストリン脱脂大豆
粉、炭酸カルシウム、塩化コバルトを含む培地で
ある。 培養条件は公知の抗生物質生産に用いられる公
知の培養条件が使用できる。培養温度は20℃ない
し37℃の範囲で、特に26℃ないし30℃の温度が適
する。培養日数は培養条件によつてことなるが、
通常4〜5日である。 培養方法は公知の培養方法がいずれも使用でき
るが特に醗酵タンク中における通気撹拌培養法が
大量生産に適する。本発明の抗生物質A11725
およびを培養物から分離、採取するためには、
先づ菌体およびその他の固型物を過または遠心
分離法によつて除去し、液より有機溶媒による
抽出法により抽出するのが最も適する方法であ
る。抽出に用いる有機溶媒としては、クロロホル
ム、ジクロルエチレン、トリクロルエチレンなど
の塩素化炭化水素、酢酸エチル、酢酸ブチル、酢
酸アミルなどの脂肪酸エステルなどが用いられる
が、その他A11725およびをよく溶解し、水
と混合しにくい有機溶媒であればいづれも使用で
きる。 A11725およびを含む有機溶媒抽出液は減
圧下で有機溶媒を留去することにより1/100〜1/2
00容に濃縮し、次いでこの濃縮液を塩酸、硫酸、
酢酸の酸でPH1.0〜3.0とした後、水層を分離し、
苛性ソーダ、苛性カリまたはアンモニアなどのア
ルカリ液でPH7.8〜9.0に調整し、再び有機溶媒で
抽出する。この抽出液を減圧下で溶媒を留去し乾
固することによりA11725およびを含有する
粗物質を得る。この粗物質をシリカゲルなどを用
いたカラムクロマト法、向流分配法などの手段を
用いて分画し、各分画についてシリカゲル薄層ク
ロマトグラフイーに付してその成分を確認し、
A11725およびを純粋に含有する分画を集め
てこれを減圧下で溶媒を留去、乾固して夫々の白
色粉末を得る。 このようにして得られたA11725およびは、
下記の物理化学的性質を有する塩基性含窒素クロ
ライド化合物である。 A11725 物質の色;白色粉末 分子式;C37H61NO12 元素分析; (実測値)C=62.34 H=9.25 N=1.96 分子量;711(マススペクトルより) 融 点;103〜107℃ 〔α〕25 D;−40.0゜ (C=1.0、メタノール) 紫外部吸収スペクトル;第7図(メタノール
中、0.025mg/ml)の通りであり、 λmax217nm(E1% 1cm=340) シヨルダーλmax240nm(E1% 1cm=180) を示す。尚第7図の波長の単位はnmを示す。 赤外部吸収スペクトル;第8図(KBr法)
の通りである。尚第8図の波長の単位はcm-1で
ある。 核磁気共鳴スペクトル;第9図(CDCl3,
100MHzTMS)の通りである。尚第9図の数値

トルを例示すれば第3表の通りである。 【表】 【表】 これらの諸性質より、A11725およびA11725
は各々16員環マクロライド系物質で、デソサミ
ンおよび6−デオキシ−2,3−ジ−O−メチル
−ヘキソースの各糖を1ケ有するもので、かつ構
造として式 で表わされる化学構造(ただし、R1は水素原子
または水酸基を示す)を有し、また分子中にアル
デヒド基を有していないもので、A11725はそ
のR1が水素原子、A11725はそのR1が水酸基で
示される各物質であつて、これらのことより、本
発明の出発物質たるA11725およびA11725は
新規物質と認められるものである。 次いで、上記の新規マクロライド抗生物質たる
A11725より本発明の目的物たるA11725a、
また対照としてのA11725より本発明の目的物
たるA11725aを得るに当つては、例えば、
A11725、A11725を氷酢酸、プロピオン酸な
どの低級脂肪酸の溶媒に溶解し、次いで冷却下に
塩化第一クロムを加え、さらに室温下3〜20時間
反応せしめればよい。また使用する塩化第一クロ
ムとしては、A11725,に対し2モル倍量以
上使用すればよい。次いでこの反応物を水または
氷水に注ぎ、その溶液を塩基性化合物、例えば炭
酸ナトリウムなどの水溶液にてPH8.5程度の弱塩
基性となした後酢酸エチルまたはベンゼンなどの
溶媒にて抽出し、その抽出液を水洗、乾燥して、
その溶媒を減圧除去して粗粉末を得、さらにこれ
を、例えばクロロホルム:メタノール:28%アン
モニア水(400:10:1)の展開溶媒からなるシ
リカゲルクロマトグラフイーにて精製することに
よつて、本発明の目的物たるA11725a、
A11725aを得る。 このようにして得られた本発明のA11725a
はA11725の分子内のエポキシを結合基に変換
せしめた二重結合を形成したもので、また
A11725aはA11725の分子内のエポキシを結
合基に変換せしめ二重結合を形成したものであつ
て、抗菌性を有する親規マクロライド抗生物質で
ある。 次に実施例および参考例を挙げて具体的に述べ
るが、本発明はこれによつて何んら限定されるも
のではない。 実施例 1 後述参考例1と同様にして得られたA11725
/gを、30mlの氷酢酸に溶解し、次いで冷却下
1.0gの塩化第一クロムを加え、さらに室温下16
時間撹拌反応せしめた。次いでその反応液を、
700mlの氷水に注ぎ、さらにその溶液を炭酸ナト
リウム水溶液にてPH8.5に調整した後、酢酸エチ
ル400mlにて3回抽出し、その酢酸エチル層を回
収、併合し、これを水洗した後無水硫酸ナトリウ
ムを加えて乾燥し、その後これを減圧乾固して
A11725a含有粗粉末約800mgを得た。次いでこ
の粗粉末を、シリカゲルを充填したカラム(24×
55cm)を用いて、クロロホルム:メタノール:28
%アンモニア水(400:10:1)の展開溶媒にて
溶出せしめて1フラクシヨン15mlづつ分画して、
そのフラクシヨンNo.130〜210を回収し、これを併
合後減圧乾固して精製されたA11725a605mgを
得た。 実施例 2 実施例1のA11725の代りに、後述参考例1
と同様にして得られたA11725を用いて、以下
実施例1と同様に行ない、そのシリカゲルカラム
にてフラクシヨンNo.150〜220を回収して、精製さ
れたA11725a612mgを得た。 参考例 1 〔A〕 デキストリン1%、ブドウ糖1%、カゼ
イン水解物0.5%、酵母エキス0.5%、炭酸カル
シウム0.1%を含有する培地(PH7.0)100mlを
内容500ml容の三角フラスコに分取し、120℃、
20分間加熱殺菌した。本培地10本にミクロモノ
スポラ・エス・ピー・A11725株の斜面培養物
よりの一白金耳を接種し、30℃、120時間振盪
培養した。次いでこれを上記と同一組成の加熱
殺菌した培地20を含有する30容ジヤーフア
ーメンターに移植し、30℃、72時間300r.p.m.
毎分20の無菌空気の条件下で通気撹拌培養し
た。次いでデキストリン5%、ブドウ糖0.5%、
脱脂大豆粉3%、炭酸カルシウム0.2%を含有
する加熱殺菌した培地(PH7.2)200を含有す
る250容タンクへ上記の培養物10を移植し、
30℃、120時間、250r.p.m.毎分100の無菌空
気の条件下通気撹拌培養し、培養物190を得
た。 〔B〕 上記培養物190を過し、菌体および
その他の固型物を別した後液160を得た。
この液を同量の酢酸エチルで抽出し、目的物
を含有する酢酸エチル溶液160を得た。 これを減圧下50に濃縮し、次いでPH2.5の
塩酸水溶液20と混合し目的物を水層に転溶し
た。さらに塩酸水溶液のPHを濃アンモニアを用
いてPH8.5に調整し、20のクロロホルムによ
り抽出し、クロロホルム層を濃縮乾固し粗製品
8.5gを得た。 〔C〕 上記粗製品8.5gをクロロホルム50mlに
溶解しあらかじめクロロホルムで充填したシリ
カゲルカラム(3cm×55cm)上に吸着させた。
次いでクロロホルム−メタノール−28%アンモ
ニア(20:1:0.1)よりなる溶媒で展開し、
15mlづゝ分画した。各フラクシヨンについてバ
チルスズブチルスを用いた抗菌力及びクロロホ
ルム−メタノール−7%アンモニア水(40:
12:20:下層)を展開溶媒とした薄層クロマト
グラフイーにより目的物を確認しその含有画分
を集めた。 第61画分より第78画分はA11725−と同定
される物質のみを含有し、この画分を濃縮乾燥
してA11725−1.2gを得た。第126画分より
第160画分はA11725と同定される物質のみ含
有し、この画分を濃縮乾燥してA117251.7g
を得た。
