【発明の詳細な説明】[Detailed description of the invention]

<産業上の利用分野> この発明は、新規な美白化粧料、特にキノコの
一種「ケロウジ」から抽出されたチロシナーゼ活
性の抑制効果を有する物質を有効成分として含有
するもので刺激性が極めて少ない美白化粧料に関
する。 <従来の技術および発明の背景> 肌の黒化には色素のメラニンが深く関与してい
るものと考えられている。すなわち、メラニンが
紫外線等の外的刺激を受けて肌の皮膚組織で増殖
され、そのために肌の黒化が促進されシミ・ソバ
カス・色黒等の症状が引き起こされるものと考え
られている。 肌の美白の作用機序としていくつか挙げられる
が、メラニンの生成に関与するチロシナーゼ(酵
素)の活性化を抑制することがその一つとして提
唱されている。 従来、チロシナーゼの活性を抑制し得る公知物
質としてグルタチオンが有効であるとされてい
る。しかし、このグルタチオンが配合された化粧
料が湿性である場合には、該グルタチオンは酸化
され易く、かつ変色・変臭を誘発し易いため特に
化粧料については致命的な問題があつた。また、
桑白皮、芍薬、当帰、黄精、川キユウ、大ソウ、
生キヨウ、黄ゴン等の各生薬(天然物)中にもチ
ロシナーゼ活性抑制効果を有する物質の存在が知
られている。また、生薬(天然物)中、特に菌類
には茯苓(Poria cocos)および木耳
(Auricularia auricula)中にもチロシナーゼ活
性抑制効果を有する物質の存在が報告されている
(フレグランスジヤーナル通巻第13、p26〜29
<1975>)。その他に、前記生薬のほか、或種の
キノコ類、つまりツクリタケ(Agaricus
bisporus)の抽出物(特公昭59−48804号)や、
シイタケ(Lentinus edodes)の抽出物〔主婦の
友生活シリーズ「健康食品と健康法」p29、(昭
50年)〕等のそれぞれに美白効果を有する物質が
含まれていることが報告されている。これら天然
物中に存在する公知のチロシナーゼ活性抑制物質
はいずれも刺激性が少ないという利点があるが、
反面そのチロシナーゼ活性抑制率が小さいという
問題がある。それ故、天然物中に存在し、チロシ
ナーゼ活性の抑制率の高い物質で、しかも化粧料
に適した化学的安定性・保存性のある物質の発見
および開発が望まれている。 <発明が解決しようとする問題点> この発明は、上記問題点を解消し且つ前記要望
に応えるために、刺激性が少なく且つチロシナー
ゼ活性抑制効果を有する物質を検索・開発するた
めに、各種天然物中のチロシナーゼ活性抑制物質
を検索およびその抑制率の測定をしたところ、前
記文献・報告等に認められる各種菌類とは分類学
上全く異なる「属(genus)」に属する菌であつ
て、担子菌類の一種のイボタケ科コウタケ属のキ
ノコのケロウジ〔Sarcodon scabrosus(Fr.)
Karst.〕から有機溶剤、グリコールまたは/およ
び水の単独または複数混合物により抽出される抽
出物中に、優れたチロシナーゼ活性抑制効果を有
する新規物質を発見したことに基づき完成され
た。この発明は、安定性が高く、また刺激性が少
なく、しかも優れたチロシナーゼ活性抑制効果を
有する美白化粧料を提供することを目的とする。 <問題点を解決するための手段> 上記の目的を達成するために、この発明では、
ケロウジ〔Sarcodon scabrosus(Fr.)Karst.〕
の子実体(キノコ)から有機溶剤、グリコールま
たは/および水により抽出された抽出物のうちか
ら選ばれた1種または2種以上の抽出物を有効成
分として配合することとした。 ケロウジからの有効成分の調製方法をさらに詳
細に説明すると、 原料は、ケロウジ(Sarcodon scabrosus(Fr.)
Karst.)の子実体(キノコ)の生体またはその子
実体(キノコ)の乾燥体を使用する。前記ケロウ
ジは、ヒダナシタケ目イボタケ科に属する担子菌
の一種で、別称イヌシシタケ、イヌススタケ、イ
ヌタケ、カラジシ、カラシシタケとも称せられ、
苦み成分を含んだ無毒の子実体(キノコ)を形成
する。ケロウジは前記苦み成分を含むために食用
とはされない。また、マツタケとその生息地域を
同じくするうえ一旦定着すると容易に消えない性
質があるところからマツタケの強敵とされてい
る。したがつて、この発明は従来利用度の低かつ
たケロウジの有効利用の途を拓くものでもある。 ケロウジからチロシナーゼ活性抑制効果を有す
る新規な有効成分の抽出用溶剤等としては、各種
有機溶剤、グリコール、水の単独液または/およ
びこれらの混合液が利用できる。 ケロウジからチロシナーゼ活性抑制効果を有す
る新規物質の抽出に利用される前記有機溶剤の例
としては、メタノール、エタノール、プロパノー
ル、イソプロピルアルコール、n−ブタノール、
ラウリルアルコール、セタノール、ステアリルア
ルコール、オレイルアルコール、ベヘニルアルコ
ールなどの炭素数1〜22の低級または/および高
級の直鎖または/および側鎖1価アルコールが利
用できるほか、アセトン、エチルメチルケトン、
イソプロピルメチルケトンなどの各種ケトン類、
酢酸エチルや脂肪酸のエチルエステルなどの各種
エステル類、ジクロルメタン、クロロホルム、ジ
クロルエタン等のハロゲン化アルカン(主として
塩化物)、ジエチルエーテル、エチルセロソルブ
等の各種エーテル類、その他ベンゼン、トルエン
等の芳香族炭化水素等々、各種極性有機溶媒また
は/および各種非極性有機溶媒が利用できる。ま
た、上記有機溶剤の単一で利用することもできる
し、適当な配合比に混合した混合液を利用するこ
ともできる。また必要に応じて加温溶融したもの
や2種以上の溶剤等の混合溶融したものを適宜利
用することができるのは勿論である。 上記有機溶剤のうち、前記アルコール類ではメ
タノール、エタノール、イソプロパノール、n−
ブタノールなどの炭素数1〜5の低級アルコール
類、エーテル類ではジエチルエーテルなど、ケト
ン類ではアセトン、エチルメチルケトン、イソプ
ロピルメチルケトンなど、エステル類では酢酸エ
チルなど、ハロゲン化アルカン類ではジクロルメ
タン、クロロホルム、ジクロルエタンなど、芳香
族炭化水素類ではベンゼン等々、極性の大きい有
機溶剤ないし中程度の極性を有する有機溶剤が特
に好適である。 また、ケロウジからチロシナーゼ活性抑制効果
を有する新規物質の抽出に利用される前記グリコ
ールの例としては、ポリエチレングリコール、プ
ロピレングリコール、1,3−ブチレングリコー
ル(1,3−BG)等の各種グリコール類が利用
できる。その他に、グリセリン、ジグリセリン、
トリグリセリン等の多価アルコール類が利用でき
る。前記グリコール類では、なかんずく1,3−
ブチレングリコール特に好適である。 さらにまた、ケロウジに含まれるチロシナーゼ
活性抑制効果を有する新規物質は、水により抽出
することができる。 また、ケロウジからチロシナーゼ活性抑制効果
を有する物質は、前記各種有機溶剤、グリコー
ル、水の各種単独でも抽出することができるが、
これら各種有機溶剤・グリコール類のそれぞれ2
種類以上の混合液〔たとえばベンゼン/酢酸エチ
ル(4:1混合液)等〕でも利用することができ
るし、さらにまた、1種又は2種以上の前記有機
溶剤と1種または2種以上の前記グリコールとの
混合液等(たとえば30%1,3−ブチレングリコ
ール/水混液)、またはこれら有機溶剤/グリコ
ール/水との混合液により抽出することができる
のは勿論である。 乾燥(風乾を含む)したケロウジまたは生のケ
ロウジ(1重量部または1容量部)にたとえばジ
クロルメタン(CH2Cl2)等の有機溶剤などを
(1〜20重量部または1〜20容量部)加え、室温
で約5〜20日間/回、1回ないし2回抽出した
後、固液分離して抽出部(前記例ではジクロルメ
タン抽出液)を得る。この抽出物(部)は、必要
に応じて溶媒等を留去して保存し、必要ならば適
切な濃度に希釈したのち以下のチロシナーゼ活性
抑制率の測定の試料とする。 その他に、所望の有機溶剤、グリコール、水等
を用いて同様に各ケロウジ抽出部を得る。このよ
うにして得たケロウジ各抽出部は、チロシナーゼ
活性抑制率の測定の試料とする。 このようにして調製されたケロウジの前記各抽
出部(たとえばメタノール抽出部、n−ブタノー
ル抽出部、ジクロルメタン抽出部、酢酸エチル抽
出部、30%1,3−ブチレングリコール(1,3
−BG)水溶液抽出部、蒸留水抽出部等々)のそ
れぞれは、チロシナーゼ活性抑制率の測定に供す
る。その結果、前記抽出物のいずれにも顕著なチ
ロシナーゼ活性抑制効果が存在するのが認められ
る。 前記ケロウジ各抽出区分の前記チロシナーゼ活
性の抑制率の測定は、次の方法により測定する。 前記ケロウジの各抽出部をたとえば30%1,3
−ブチレングリコール水溶液等の適当な溶剤によ
り適当な濃度(たとえば約1〜5%等の濃度であ
つて、475nmにおける吸光度が適当な値を示す
範囲の濃度)に希釈したものを試験溶液として調
整する。 試験管にそれぞれL−チロシン溶液(0.3mg/
ml)1mlと、マツキルベイン緩衝液(McIlvain´s
Buffer Solution)(PH6.8)1mlとを入れておき、
これらの各試験管にケロウジ抽出液の前記希釈液
およびブランクテスト用の30%1,3−ブチレン
グリコール水溶液をそれぞれ0.9ml加え、これを
37℃の恒温水槽中で10分間インキユベートする。 前記インキユベートしたものにチロシナーゼ溶
液(1mg/ml・マツキルベイン緩衝液)を0.1ml
加えて、よく撹拌し直ちに各反応液を分光光度計
にセツトし475nmにおける吸光度を経時的に測
定する。(各時での吸光度値を、チロシナーゼ溶
液添加直後に対しては添字oを、添加後x分イン
キユベート経過後に対しては添字xをそれぞれ付
して示す)各吸光度値を次式に代入してチロシナ
ーゼ活性抑制率を算出する。なお、この発明のチ
ロシナーゼ活性抑制率の算出には、反応液投入後
15分後の吸光度値を使用する。 チロシナーゼ活性抑制率=(Bx−Bo)−(Ax−Ao
)/(Bx−Bo)×100………式 Bo:ブランク溶液の0分後における吸光度値 Bx:ブランク溶液のx分後における吸光度値 Ao:試験溶液の0分後における吸光度値 Ax:試験溶液のx分後における吸光度値 なお、前記吸光度値はドーバクロム(メラニン
の前駆物質)の生成量により測定されるものであ
る。 ケロウジから抽出された前記各抽出物は、その
抽出過程からも明らかなように、各種有機溶剤
(極性溶剤、非極性溶剤)、グリコール類、水等に
対する相溶性に優れている。特に化粧料に使用さ
れる各種基剤および各種有機溶媒等に対する溶解
性、混和性、相溶性などに優れているという特性
を有する。つまり化粧料によく利用される有機溶
媒等たとえば、エタノール、イソプロピルアルコ
ール、ラウリルアルコール、セタノール、ステア
リルアルコール、オレイルアルコール、ベヘニル
アルコール、プロピレングリコール、1,3−ブ
チレングリコール、ジプロピレングリコール、ミ
リスチン酸イソプロピル、パルミチン酸イソプロ
ピル、その他各種動物性オイルおよび植物性オイ
ル等に対する溶解性等が優れている。 そして、ケロウジから各種有機溶剤、グリコー
ル類、水等により抽出された前記各抽出物は、公
知の各種化粧量基剤等を配合して、クリーム、乳
液、化粧水、パツク、洗顔料などの各種基礎化粧
料、フアンデーシヨン、ほほ紅、口紅、白粉など
の各種メーキヤツプ料、石鹸、シヤンプー、リン
ス、香水、オーデコロンその他の化粧料に対して
広範囲に適用できる。 また、前記各種化粧料の形状は、溶液、エマル
ジヨン、軟膏、オイル、ワツクス、ゲル、ゾル、
粉末(パウダー)、スプレー(エアゾール)等の
各種形状で適用することが出来る。 ケロウジの各種有機溶剤、グリコール類また
は/および水による前記各抽出物は、いずれも皮
膚に対する毒性および刺激性がなく(累積刺激試
験およびパツチテスト法等による)、熱、光に対
する安定性が高く、さらには各種化粧料基剤、各
種化粧料添加剤(たとえば、各種界面活性剤、溶
剤、色素、香料、防腐剤、抗酸化剤、保湿剤、ビ
タミン、動植物抽出物その他の各種添加剤)に対
する安定性も高いという卓越した特性を有してお
り、これらの各種化粧料基剤および化粧料添加剤
等とも併用することができる。 ケロウジの各有機溶剤による抽出部、グリコー
ル類による抽出部、水による抽出物それぞれの各
種化粧料中に対する配合量は、その使用形態によ
り変化させることができるので特に限定されな
い、原則的には、有効量存在すればよいことにな
るが、一般的には化粧量組成物中(総重量)に対
して0.01〜20重量%、好ましくは0.5〜10重量%
配合するのがよい。 <作用> この発明にかかる美白化粧料は、ケロウジから
の有機溶剤、グリコールまたは/および水により
抽出される抽出物が有するチロシナーゼ活性抑制
作用により、優れた美白効果を奏する。 <実施例> (1) ケロウジ抽出物の調製。 ケロウジの生体(1Kg)を室温で約24時間風
乾し、乾燥ケロウジ(約90〜100g)を得る。 前記乾燥ケロウジ(50g)にジクロルメタン
(CH2Cl2)約500mlを加え、室温で7日間時々
撹拌抽出する。このジクロルメタンの抽出を2
回繰り返し、いずれも固液分離してその液層を
収集しジクロルメタン抽出部(約950ml)を得
た後、溶剤を留去して保存する。 以下同様に、有機溶剤には、メタノール、n
−ブタノール、酢酸エチルを、グリコールには
30%1,3−ブチレングリコール/水混合液
を、その他に蒸留水を用いてそれぞれケロウジ
から各抽出物を得る。 風乾のケロウジから抽出された各ケロウジ抽
出物として得たものは、次のとおりである。
(なお、蒸留水抽出物(粗抽出物)にはケロウ
ジの50〜55%が抽出され、他の抽出部と比較し
