Abstract

NEW MATERIAL:A compound shown by formula I (R1 is H or acyl; R3 is H or hydroxyl group). EXAMPLE:1-(3-Hydroxy-4-hydroxymethyl-4-cyclopenten-1-yl)cytosine. USE:An antitumor agent and antiviral agent showing the same pharmaceutical actions as those of an optically active substance. PREPARATION:First, a 1-aminocyclopentene compound shown by formula II (R3 and R4 are bonded to form hydroxyl-protecting group; R5 is hydroxyl protecting group) is reacted with a 3-lower alkoxy-2-propenoyl isocyanate to give a compound shown by formula III (R6 is lower alkyl), which is subjected to ring closure. Then hydroxyl groups at 2'-, 3'- and 6'-positions of the formed substance are protected with acyl group to give a compound shown by formula IV (R3'-R5' are acyl), which is successively thiolated, alkylated and aminated to give a compound shown by formula V. Finally the compound shown by formula V is deprotected to give the aimed compound wherein R1 is H and R2 is hydroxyl group.
