Abstract
NEW MATERIAL:Human tumor cytotoxic factor hTCF having 30,000+ or -5,000mol. wt. and pH 6.0+ or -0.5 isoelectric point. USE:An antitumor agent. PREPARATION:For example, peripheral blood of healthy people is collected, a mononuclear cell is isolated by centrifugal separation, inoculated into a medium containing bovine fetal serum and lipopolysaccharide, cultivated at 37 deg.C for 16hr, the mononuclear cell is removed by centrifugal separation and the culture mixture is filtered by a membrane filter and sterilized to give crude hTCF-containing solution. The solution is concentrated by an ultrafilter, passed through a column packed with crosslinked dextran gel, eluted to collect a fraction having cytocidal effects. The fraction is concentrated and purified by high- performance liquid gel filtration chromatography to give hTCF.
