Abstract

PURPOSE:To obtain a dry type immunoassay element having high sensitivity by incorporating the high-polymer antigen which is the conjugate of ligand or the deriv. thereof and high-polymer compd., water soluble high-polymer material, and the conjugate of an antigen reacting with the ligand and the enzyme acting on the water soluble high polymer into a water permeable layer. CONSTITUTION:Water-soluble dextrin is used as the high-polymer compd. bonded to the ligand (e.g., urine) or the deriv. thereof in the uniform water permeable layer formed by using gelatin, etc., as a binder. Conjugation of the ligand or the deriv. thereof (introduced with an amino group, etc.) and the high-polymer compd. is executed by taking the functional groups of both such as amino groups into consideration to prepare the high-polymer antigen. Starch, etc., are used for the water soluble high-polymer material and the conjugate using amylase is incorporated in the enzyme. The water soluble high-polymer material is used as the substrate of the enzyme-antibody conjugate, by which theophylline is quantitatively analyzed easily and rapidly with high sensitivity.
