【発明の詳細な説明】
本発明は新規なカルボスチリル誘導体及びその
塩に関する。
本発明のガルボスチリル誘導体は、下記一般式
(1)で表わされる。
[式中R1は水素原子を示す、R2は水素原子、
フエノキシ低級アルキル基、低級アルキル基、フ
エニル環上に置換基として低級アルキル基及びハ
ロゲン原子から選ばた基を1〜3個有することの
あるフエニル基が1個置換した低級アルキル基又
はフエニル環上に置換基としてハロゲン原子を1
〜3個有するベンゾイル基を示す。]
上記一般式(1)で表わされるカルボスチリル誘導
体は、心筋の収縮を増強する作用(陽性変力作
用)及び冠血流量増加作用を有し、例えばうつ血
性心不全、僧帽弁膜症、心房性細動、粗動、発作
性心房性頻脈等の各種心臓疾患の治療のための強
心剤として有効である。特に上記一般式(1)で表わ
される本発明誘導体は、優れた陽性変力作用及び
冠血流量増加作用を有する反面、心拍数の増加作
用は殆んど有していない点において特長付けられ
る。
上記一般式(1)中R1及びR2で定義される各基の
具体例を夫々次に例示する。
低級アルキル基…メチル、エチル、プロピル、
イソプロピル、ブチル、tert―ブチル、ペンチ
ル、ヘキシル等の炭素数1〜6のアルキル基。
フエノキシ低級アルキル基…フエノキシメチ
ル、2―フエノキシエチル、2―フエノキシプロ
ピル、3―フエノキシプロピル、1―メチル―2
―フエノキシエチル、2―フエノキシブチル、3
―フエノキシブチル、4―フエノキシブチル、
1,1―ジメチル―2―フエノキシブチル、2―
フエノキシペンチル、3―フエノキシペンチル、
4―フエノキシヘキシル等のフエノキシ基を置換
基として有する炭素数1〜6のアルキル基。
低級アルキル基及びハロゲン原子から選ばれた
基を1〜3個有することのあるフエニル基が1個
置換した低級アルキル基…ベンジル、2―フエニ
ルエチル、3―フエニルプロピル、6―フエニル
ヘキシル、4―フエニルブチル、5―フエニルペ
ンチル等の非置換フエニル基を1個有する炭素数
1〜6のアルキル並びに2―クロロベンジル、3
―ブロモベンジル、4―ヨードベンジル、2―
(4―フルオロフエニル)エチル、1―(3―ク
ロロフエニル)エチル、6―(4―ブロモフエニ
ル)ヘキシル、5―(3,4―ジクロロフエニ
ル)ペンチル、1,1―ジメチル―2―(2,5
―ジブロムフエニル)プロピル、2―もしくは3
―メチルベンジル、2―(4―メチルフエニル)
エチル、2―エチルベンジル、1―(3―エチル
フエニル)エチル、3―(4―エチルフエニル)
プロピル、6―(4―イソプロピルフエニル)ヘ
キシル、4―(4―ヘキシルフエニル)ブチル、
1,1―ジメチル―2―(3,4―ジメチルフエ
ニル)エチル、5―(3,4―ジエチルフエニ
ル)ペンチル、6―(3,5―ジメチルフエニ
ル)ヘキシル、2―メチル―3―(2,5―ジメ
チルフエニル)プロピル、3―メチル―4―クロ
ロベンジル、2―クロロ―6―メチルベンジル、

クロロベンジル等のフエニル環上に炭素数1〜6
のアルキル基及びハロゲン原子から成る群から選
ばれた置換基を1〜3個有するフエニル基を1個
有する炭素数1〜6のアルキル基。
フエニル環上に置換基としてハロゲン原子を1
〜3個有するベンゾイル基…2―,3―もしくは
4―クロロベンゾイル、2―、3―もしくは4―
フルオロベンゾイル、2―、3―もしくは4―ブ
ロムベンゾイル、2―、3―もしくは4―ヨード
ベンゾイル、3,5―ジクロロベンゾイル、2,
6―ジクロロベンゾイル、3,4―ジクロロベン
ゾイル、3,4―ジフルオロベンゾイル、3,5
―ジブロムベンゾイル、3,4,5―トリクロロ
ベンゾイル等のフエニル環上にハロゲン原子を1
〜3個有するベンゾイル基。
本発明の誘導体は、例えば下記反応行程式―1
の方法で製造され得る。
〔式中R1及びR2は前記に同じ。〕
上記反応行程式―1で示される方法は、一般式
(2)で表わされるカルボスチリル誘導体又はそのカ
ルボキシ基の活性化された誘導体と一般式(3)で表
わされるピペラジル誘導体とを通常のアミド結合
生成反応にて反応させることにより実施される。
アミド結合生成反応は公知の各種方法例えば(イ)混
合酸無水物法即ちカルボスチリル誘導体(2)にアル
キルハロカルボン酸を反応させて混合酸無水物と
し、これにピペラジン誘導体(3)を反応させる方
法;(ロ)活性エステル法、即ちカルボスチリル誘導
体(2)をp―ニトロフエニルエステル、N―ヒドロ
キシコハク酸イミドエステル、1―ヒドロキシベ
ンゾトリアゾールエステル等の活性エステルと
し、これにピペラジン誘導体(3)を反応させる方
法;(ハ)カルボジイミド法即ちカルボスチリル誘導
体(2)にピペラジン誘導体(3)をジシクロヘキシルカ
ルボジイミド、カルボニルジイミダゾール等の活
性化剤の存在下に縮合させる方法;(ニ)その他の方
法、例えばカルボスチリル誘導体(2)を無水酢酸等
の脱水剤によりカルボン酸無水物としこれにピペ
ラジン誘導体(3)を反応させる方法、カルボスチリ
ル誘導体(2)と低級アルコールとのエステルにピペ
ラジン誘導体(3)を高圧高温下に反応させる方法、
カルボスチリル誘導体(2)の酸ハロゲン化物即ちカ
ルボン酸ハライドにピペラジン誘導体(3)を反応さ
せる方法等により実施することが出来る。またカ
ルボスチリル誘導体(2)をトリフエニルホスフイン
やジエチルクロロホスフエート等のリン化合物で
活性化し、これにピペラジン誘導体(3)を反応させ
る方法によることもできる。
上記(イ)に示す混合酸無水物法において、用いら
れる混合酸無水物は通常のシヨツテン―バウマン
反応により得られ、これを通常単離することなく
ピペラジン誘導体(3)と反応させることにより一般
式(1)の本発明化合物が製造される。シヨツテン―
バウマン反応は通常シヨツテン―バウマン反応に
慣用の塩基性化合物例えばトリエチルアミン、ト
リメチルアミン、ピリジン、ジメチルアニリン、
N―メチルモルホリン、1,5―ジアザビシクロ
〔4,3,0〕ノネン―5(DBN)、1.5―ジアザ
ビシクロ〔5,4,0〕ウンデセン―5(DBU)、
1,4―ジアザビシクロ〔2,2,2〕オクタン
(DABCO)等の有機塩基及び炭酸カリウム、炭
酸ナトリウム、炭酸水素カリウム、炭酸水素ナト
リウム等の無機灰塩基の存在下、約−20〜100℃
好ましくは0〜50℃において、約5分〜10時間好
ましくは5分〜2時間を要して行われる。得られ
た混合酸無水物とピペラジン誘導体(3)との反応
は、約−20〜150℃好ましくは10〜50℃において
約5分〜10時間好ましくは約5分〜5時間を要し
て行われる。また上記混合酸無水物法は一般にこ
の種混合酸無水物法に慣用の溶媒、具体的には塩
化メチレン、クロロホルム、ジクロロエタン等の
ハロゲン化炭化水素類、ベンゼン、トルエン、キ
シレン等の芳香族炭化水素類、ジエチルエーテ
ル、テトラヒドロフラン、ジメトキシエタン等の
エーテル類、酢酸メチル、酢酸エチル等のエステ
ル類、N,N―ジメチルホルムアミド、ジメチル
スルホキシド、ヘキサメチルリン酸トリアミド等
の非プロトン性極性溶媒などの適当な溶媒中で行
なわれる。尚上記混合酸無水物の製造において使
用されるアルキルハロカルボン酸としてはクロロ
蟻酸メチル、ブロモ蟻酸メチル、クロロ蟻酸エチ
ル、ブロモ蟻酸エチル、クロロ蟻酸イソブチル等
を例示でき、之等は通常カルボスチリル誘導体(2)
に対し少なくとも等モル量、好ましくは約1〜2
倍モル量用いられる。またピペラジン誘導体(3)の
使用割合は、通常カルボスチリル誘導体(2)に対し
て少なくとも等モル好ましくは約1〜2倍モルと
するのが好ましい。
上記(ロ)に示す活性エステル法は、例えばN―ヒ
ドロキシコハク酸イミドエステルを用いる場合を
例にとれば、反応に影響を与えない適当な溶媒中
で行なわれる。該溶媒としては、具体的には塩化
メチレン、クロロホルム、ジクロロエタン等のハ
ロゲン炭化水素類、ベンゼン、トルエン、キシレ
ン等の芳香族炭化水素類、ジエチルエーテル、テ
トラヒドロフラン、ジメトキシエタン等のエーテ
ル類、酢酸メチル、酢酸エチル等のエステル類、
N,N―ジメチルホルムアミド、ジメチルスルホ
キシド、ヘキサメチルリン酸トリアミド等の非プ
ロトン性極性溶媒などが挙げられる。反応は、0
〜150℃、好ましくは10〜100℃で、5〜30時間で
終了する。ピペラジン誘導体(3)とN―ヒドロキシ
コハク酸イミドエステルとの使用割合は、後者に
対して前者を通常、少なくとも等モル、好ましく
は、等モル〜2倍モルとするのが望ましい。
上記(ニ)に示すその他の方法のうちカルボン酸ハ
ライドにピペラジン誘導体(3)を反応させる方法を
採用する場合、該反応は塩基性化合物の存在下適
当な溶媒中にて行なわれる。塩基性化合物として
は公知のものを広く使用でき、例えば上記シヨツ
デン―バウマン反応に用いられる塩基性化合物の
ほかに水酸化ナトリウム、水酸化カリウム、水素
化ナトリウム、水素化カリウム等を挙げることが
できる。溶媒としては、上記シヨツテン―バウマ
ン反応に用いられる溶媒のほかに例えばメタノー
ル、エタノール、プロパノール、ブタノール、3
―メトキシ―1―ブタノール、エチルセロソル
ブ、メチルセロソルブ等のアルコール類、ピリジ
ン、アセトン、アセトニトリル等又は上記溶媒の
二つ以上の混合溶媒等を挙げることができる。ピ
ペラジン誘導体(3)とカルボン酸ハライドとの使用
割合は特に限定がなく広い範囲内で適宜選択され
るが、通常後者に対して前者を少なくとも等モル
量度、好ましくは等モル〜2倍モル量用いるのが
よい。該反応は通常−20〜180℃程度、好ましく
は約0〜150℃にて行なわれ、一般に5分〜30時
間で反応は完結する。
またカルボスチリル誘導体(2)をトリフエニルホ
スフインやジエチルクロロホスフエート等のリン
化合物で活性化し、これにピペラジン誘導体(3)を
反応させる方法は、適当な溶媒中で行うことが出
来る。ここで溶媒としては反応に影響を与えない
ものなら何れでも使用できるが、具体的には塩化
メチレン、クロロホルム、ジクロロエタン等のハ
ロゲン化炭化水素類、ベンゼン、トルエン、キシ
レン等の芳香族炭化水素類、ジエチルエーテル、
テトラヒドロフラン、ジメトキシエタン等のエー
テル類、酢酸メチル、酢酸エチル等のエステル
類、N,N―ジメチルホルムアミド、ジメチルス
ルホキシド、ヘキサメチルリン酸トリアミド等の
非プロトン性極性溶媒などが挙げられる。上記反
応では、ピペラジン誘導体(3)自体が塩基性化合物
として働くたため、これを理論量より過剰量用い
ることにより、反応は良好に進行するが、必要に
応じて、他の塩基性化合物例えば、トリエチルア
ミン、トリメチルアミン、ピリジン、ジメチルア
ニリン、N―メチルモルホリン、DBN,DBU,
DABCO等の有機塩基及び炭酸カリウム、炭酸ナ
トリウム、炭酸水素カリウム、炭酸水素ナトリウ
ム等の無機塩基を用いることもできる。該反応は
約0〜150℃好ましくは約0〜100℃において行わ
れ、反応時間は約1〜30時間である。カルボスチ
リル誘導体(2)に対するリン化合物及びピペラジン
誘導体(3)の使用割合は、夫々通常少なくとも等モ
ル量程度好ましくは、1〜3倍モル量とされる。
また本発明の誘導体中ある群に属する化合物
(下記一般式(1―a)、(1―b)及び(1―c)
で表わされる)は、夫々以下の反応行程式―2乃
至―5に示す方法によつても製造することができ
る。
〔各式中R1は前記に同じ。R2′は低級アルカノ
イル基、低級アルコキシカルボニル基、フロイル
基又はフエニル環上に置換基として低級アルキル
基、低級アルコキシ基、ハロゲン原子、ニトロ基
及びシアノ基から選ばれた基を1〜3個もしくは
低級アルキレンジオキシ基を有することのあるベ
ンゾイル基を示す。X1は水酸基を示す。〕
即ち上記一般式(1―a)で表わされる本発明
誘導体は、一般式(4)の化合物に一般式(5)の化合物
又はそのカルボキシ基の活性化された化合物を反
応させることにより製造される。上記反応は、前
記反応行程式―1に示す一般式(2)で表わされるカ
ルボスチリル誘導体と、一般式(3)で表わされるピ
ペラジン誘導体との反応と同様の操作及び条件下
に行なうことができる。
〔各式中R1は前記に同じ。R2″は低級アルケニ
ル基、低級アルキニル基、低級アルカンスルホニ
ル基、フエノキシ低級アルキル基、置換基として
シアノ基又はカルバモイル基を有することのある
低級アルキル基、フエニル環上に置換基として低
級アルキル基、低級アルコキシ基、ハロゲン原
子、ニトロ基、アミノ基、低級アルカノイルアミ
ノ基及び低級アルキルチオ基から選ばれた基を1
〜3個有することのあるフエニル低級アルキル
基、又はフエニル環上に置換基として低級アルキ
ル基を有することのあるフエニルスルホニル基を
示す。X2はハロゲン原子、低級アルカンスルホ
ニルオキシ基、アリールスルホニルオキシ基又は
アラルキルスルホニルオキシ基を示す〕。
即ち一般式(1―b)で表わされる本発明誘導
体は、一般式(4)で表わされる化合物と一般式(6)で
表わされる化合物とを反応させることにより製造
される。この反応は前記したカルボン酸ハライド
にピペラジン誘導体(3)を反応させる方法と同様の
操作及び条件下に実施することができる。
尚上記一般式(6)で表わされる化合物において、
X2で定義されるハロゲン原子は具体的には塩素、
弗素、臭素及び沃素原子であり、低級アルカンス
ルホニルオキシ基としては具体的にはメタンスル
ホニルオキシ、エタンスルホニルオキシ、イソプ
ロパンスルホニルオキシ、プロパンスルホニルオ
キシ、ブタンスルホニルオキシ、tert―ブタンス
ルホニルオキシ、ペンタンスルホニルオキシ、ヘ
キサンスルホニルオキシ基等を例示でき、またア
リールスルホニルオキシ基としては具体的にはフ
エニルスルホニルオキシ、4―メチルフエニルス
ルホニルオキシ、2―メチルフエニルスルホニル
オキシ、4―ニトロフエニルスルホニルオキシ、
4―メトキシフエニルスルホニルオキシ、3―ク
ロルフエニルスルホニルオキシ、α―ナフチルス
ルホニルオキシ基等の置換又は未置換のアリール
スルホニルオキシ基を例示でき、またアラルキル
スルホニルオキシ基としては具体的にはベンジル
スルホニルオキシ、2―フエニルエチルスルホニ
ルオキシ、4―フエニルブチルスルホニルオキ
シ、4―メチルベンジルスルホニルオキシ、2―
メチルベンジルスルホニルオキシ、4―ニトロベ
ンジルスルホニルオキシ、4―メトキシベンジル
スルホニルオキシ、3―クロルベンジルスルホニ
ルオキシ、α―ナフチルメチルスルホニルオキシ
基等の置換又は未置換のアラルキルスルホニルオ
キシ基を例示できる。
〔各式中R1は前記に同じ。R2は低級アルケ
ニル基、低級アルキニル基、フエノキシ低級アル
キル基、置換基としてシアノ基又はカルバモイル
基を有することのある低級アルキル基又はフエニ
ル環上に置換基として低級アルキル基、低級アル
コキシ基、ハロゲン原子、ニトロ基、アミノ基、
低級アルカノイルアミノ基及び低級アルキルチオ
基から選ばれた基を1〜3個有することのあるフ
エニル低級アルキル基を示す。Xは上記X1及び
X2基と同一基を示す。〕
即ち一般式(1―c)で表わされる本発明誘導
体は、一般式(2)の化合物と一般式(7)の化合物を反
応させ、かくして得られる一般式(8)の化合物に一
般式(9)の化合物を反応させることにより製造でき
る。上記第1の工程における反応は、前記一般式
(2)の化合物と一般式(3)の化合物との反応と同様の
操作及び条件下に行なわれる。
また上記第1工程に引き続く第2工程は、一般
式(8)の化合物におけるXの種類に応じて、下記の
如くして行なわれる。即ち一般式(8)の化合物とし
てXがハロゲン原子、低級アルカンスルホニルオ
キシ基、アリールスルホニルオキシ基又はアラル
キルスルホニルオキシ基を示す化合物を用いる場
合、該一般式(8)の化合物と一般式(9)の化合物との
反応は、一般に適当な不活性溶媒中塩基性縮合剤
の存在下又は不存在下に行なわれる。不活性溶媒
としては例えばベンゼン、トルエン、キシレン等
の芳香族炭化水素類、メタノール、エタノール、
イソプロパノール、ブタノール等の低級アルコー
ル類、酢酸、酢酸エチル、ジメチルスルホキシ
ド、ジメチルホルムアミド、ヘキサメチルリン酸
トリアミド等を挙げることができる。また塩基性
縮合剤としては例えば炭酸ナトリウム、炭酸カリ
ウム、炭酸水素ナトリウム、炭酸水素カリウム等
の炭酸塩、水酸化ナトリウム、水酸化カリウム等
の金属水酸化物、ナトリウムメチレート、ナトリ
ウムエチレート等の金属アルコラート、ピリジ
ン、トリエチルアミン等の第3級アミン等を挙げ
ることができる。一般式(8)の化合物と一般式(9)の
化合物との使用割合は特に限定がなく広い範囲内
で適宜選択でき、通常前者に対して後者を少なく
とも等モル量程度、好ましくは等モル〜5倍モル
量用いるのがよい。該反応は通常40〜120℃程度、
好ましくは50〜100℃にて行なわれ、一般に5〜
30時間程度で反応は終了する。
また一般式(8)の化合物としてXが水酸基を示す
化合物を用いる場合、該一般式(8)の化合物と一般
式(9)の化合物との反応は、脱水縮合剤の存在下無
溶媒下又は適当な溶媒中にて行なわれる。ここで
脱水縮合剤としては例えばポリリン酸等の縮合リ
ン酸類、正リン酸、焦リン酸、メタリン酸等のリ
ン酸類、正亜リン酸等の亜リン酸類、五酸化リン
等の無水リン酸類、塩酸、硫酸、ホウ酸等の酸
類、リン酸ナトリウム、ボロンホスフエート、リ
ン酸第二鉄、リン酸アルミニウム等の金属リン酸
塩類、活性アルミナ、重硫酸ナトリウム、ラネー
ニツケル等を挙げることができる。また溶媒とし
ては例えばジメチルホルムアミド、テトラリン等
の高沸点溶媒を挙げることができる。一般式(8)の
化合物と一般式(9)の化合物との使用割合は特に限
定がなく広い範囲内で適宜選択でき、通常後者に
対して前者を0.8モル量程度以上、好ましくは0.8
モル〜2倍モル量用いるのがよいい。脱水縮合剤
の使用量は特に限定されず広範囲から適宜選択し
得、一般式(8)の化合物に対して通常触媒量以上、
好ましくは0.5〜5倍モル量程度用いるのがよい。
上記反応は有利には酸化反応を防止するため不活
性ガス例えばCO2又はN2気流中で行ない得る。
また反応は通常常圧下、約100〜350℃好ましくは
125〜255℃にて約3〜10時間で行なわれる。
〔各式中R1及びR2は前記に同じ。〕
一般式(1―c)で表わされる本発明誘導体
は、また上記のように一般式(2)の化合物にモルホ
リン(10)を反応させモルホリン誘導体(11)としたの
ち、これに一般式(9)の化合物を反応させることに
よつても製造できる。
上記において一般式(2)の化合物とモルホリン(10)
との反応は、前記した一般式(2)の化合物と一般式
(3)の化合物との反応と同様の操作及び条件下に行
ない得る。またかくして得られる一般式(11)の化合
物と一般式(9)の化合物との反応は、無溶媒下又は
適当な溶媒中酸の存在下にて行なわれる。溶媒と
しては例えばテトラリン、ジメチルホルムアミ
ド、ジメチルスルホキシド、ヘキサメチルリン酸
トリアミド等の高沸点溶媒を使用できる。酸とし
ては例えば塩酸、硫酸、臭化水素酸等を使用でき
る。一般式(11)の化合物と一般式(9)の化合物との使
用割合は、特に限定されず広い範囲内で適宜選択
でき、通常前者に対して後者を少なくとも等モル
量程度、好ましくは等モル〜2倍量モル量用いい
るのがよい。該反応は通常50〜250℃程度、好ま
しくは150〜200℃にて行なわれ、一般に1〜24時
間程度で反応は終了する。
更に一般式(1)で表わされる本発明誘導体中R1
が水素原子以外の基である化合物(一般式(1―
e)の化合物)は、R1が水素原子である化合物
(一般式(1―d)の化合物)より以下の反応行
程式―6に示す方法によつても製造することがで
きる。
〔各式中R2及びX2は上記に同じ。R1′は水素原
子以外のR1基を示す。但しR2は水素原子でない
ものとする。〕
上記において一般式(1―d)の化合物と一般
式(12)の化合物との反応は、例えば塩基性化合物の
存在下適当な溶媒中にて行なうのがよい。ここで
塩基性化合物としては例えば水素化ナトリウム、
カリウム、ナトリウム、ナトリウムアミド、カリ
ウムアミド等を挙げることができる。また溶媒と
しては例えばジオキサン、ジエチレングリコー
ル、ジメチルエーテル等のエーテル類、トルエ
ン、キシレン等の芳香族炭化水素類、ジメチルホ
ルムアミド、ジメチルスルホキシド、ヘキサメチ
ルリン酸トリアミド等を挙げることができる。一
般式(1―d)の化合物と一般式(12)の化合物の使
用割合は特に限定がなく広く範囲内で適宜選択で
き、通常前者に対して後者を少なくとも等モル程
度以上、好ましくは等モル〜2倍モル程度用いる
のがよい。該反応は通常0〜70℃程度、好ましく
は0℃〜室温付近にて行なわれ、一般に0.5〜12
時間程度で反応は終了する。
また一般式(1―d)で表わされる本発明の化
合物のうちR2がフエニル環上に置換基としてア
ミノ基を有する基である化合物は、該フエニル環
上の置換基がニトロ基である対応する化合物を還
元することによつても容易に製造され得る。この
還元は芳香族ニトロ基を芳香族アミノ基に還元す
る通常の条件下に実施できる。より具体的には亜
硫酸ソーダ、亜硫酸ガス等の還元剤を用いる方
法、パラジウム―炭素等の還元触媒を用いる接触
還元法等によればよい。
以上詳述した反応行程式―1、―4及び―5に
おいて、本発明誘導体の製造に利用される一般式
(2)の化合物は、一部新規化合物を包含しており、
該化合物は例えば下記反応行程式―7に示す方法
により製造することができる。
〔式中R1及びXは上記に同じ。R3は芳香族ア
ミン残基を、R3―Hは芳香族アミン類を、また
X2′はハロゲン原子を示す。〕
上記において一般式(15)の化合物を得る反応
は適当な溶媒中又は無溶媒下に一般式(13)の化
合物に芳香族アミン類(14)を反応させることに
より行うことが出来る。溶媒としては、反応に影
響を与えないものであればいずれも使用すること
が出来、例えば塩化メチレン、クロロホルム、ジ
クロロメタン等のハロゲン化炭化水素類、ジエチ
ルエーテル、テトラヒドロフラン、ジメトキシエ
タン等のエーテル類、酢酸メチル、酢酸エチル等
のエステル類、N,N―ジメチルホルムアミド、
ジメチルスルホキシド、ヘキサメチルリン酸トリ
アミド等の非プロトン性極性溶媒などが挙げられ
る。芳香族アミン類としては、ピリジン、キノリ
ン等を例示できる。該芳香族アミンの使用量は、
一般式(13)の化合物に対して、少なくとも等モ
ル、好ましくは大過剰量使用するのがよい。反応
温度は、50〜200℃好ましくは70〜150℃であり、
反応は3〜10時間で終了する。また上記により得
られる一般式(15)の化合物の加水分解反応は、
水又はメタノール、エタノール等のアルコール
中、水酸化ナトリウム又は水酸化カリウムのよう
な無機塩基類もしくは塩酸、臭化水素酸の酸を用
いて室温〜150℃下に1〜10時間程度を要して行
なわれる。また反応行程式―1において他方の原
料とする一般式(3)で表わされるピペラジン誘導体
もその一部に新規の化合物を包含しており、該化
合物は前記一般式(4)の化合物と一般式(5)又は一般
式(6)の化合物との反応において、一般式(4)の化合
物に替えピペラジンを用いることにより、容易に
収得できる。
本発明の一般式(1)で表わされるカルボスチリル
誘導体のうち、塩基性基を有する化合物は、医薬
的に許容される酸を作用させることにより容易に
酸付加塩とすることができる。該酸としては例え
ば、塩酸、硫酸、リン酸、臭化水素酸等の無機
酸、シユウ酸、マレイン酸、フマール酸、リンゴ
酸、酒石酸、クエン酸、安息香酸等の有機酸を挙
げることができる。
斯くして得られる各々の行程での目的化合物
は、通常の分離手段により容易に単離精製するこ
とができる。該分離手段としては、例えば溶媒抽
出法、稀釈法、再結晶法、カラムクロマトグラフ
イー、プレパラテイブ薄層クロマトグラフイー等
を例示できる。
尚本発明は光学異性体も当然に包含するもので
ある。
一般式(1)の化合物は通常、一般的な医薬製剤の
形態で用いられる。製剤は通常使用される充填
剤、増量剤、結合剤、付湿剤、崩壊剤、表面活性
剤、滑沢剤などの稀釈剤あるいは賦形剤を用いて
調製される。この医薬製剤としては各種の形態が
治療目的に応じて選択でき、その代表的なものと
して錠剤、丸剤、散剤、液剤、懸濁剤、乳剤、顆
粒剤、カプセル剤、坐剤、注射剤(液剤、懸濁剤
等)などが挙げられる。錠剤の形態に成形するに
際しては、担体としてこの分野で従来公知のもの
を広く使用でき、例えば乳糖、白糖、塩化ナトリ
ウム、ブドウ糖、尿素、デンプン、炭酸カルシウ
ム、カオリン、結晶セルロース、ケイ酸などの賦
形剤、水、エタノール、プロパノール、単シロツ
プ、ブドウ糖液、デンプン液、ゼラチン溶液、カ
ルボキシメチルセルロース、セラツク、メチルセ
ルロース、リン酸カリウム、ポリビニルピロリド
ンなどの結合剤、乾燥デンプン、アルギン酸ナト
リウム、カンテン末、ラミナラン末、炭酸水素ナ
トリウム、炭酸カルシウム、ポリオキシエチレン
ソルビタン脂肪酸エステル類、ラウリル硫酸ナト
リウム、ステアリン酸モノグリセリド、デンプ
ン、乳糖などの崩壊剤、白糖、ステアリン、カカ
オバター、水素添加油などの崩壊抑制剤、第四級
アンモニウム塩基、ラウリル硫酸ナトリウムなど
の吸収促進剤、グリセリン、デンプンなどの保湿
剤、デンプン、乳糖、カオリン、ベントナイト、
コロイド状ケイ酸などの吸着剤、精製タルク、ス
テアリン酸塩、ホウ酸末、ポリエチレングリコー
ルなどの滑沢剤などが例示できる。さらに錠剤は
必要に応じ通常の剤皮を施した錠剤、例えば糖衣
錠、ゼラチン被包錠、腸溶被錠、フイルムコーテ
イング錠あるいは二重錠、多層錠とすることがで
きる。丸剤の形態に成形するに際しては、担体と
してこの分野で従来公知のものを広く使用でき、
例えばブドウ糖、乳糖、デンプン、カカオ脂、硬
化植物油、カオリン、タルクなどの賦形剤、アラ
ビアゴム末、トラガント末、ゼラチン、エタノー
ルなどの結合剤、ラミナラン、カンテンなどの崩
壊剤などが例示できる。坐剤の形態に成形するに
際しては、担体として従来公知のものを広く使用
でき、例えばポリエチレングリコール、カカオ
脂、高級アルコール、高級アルコールのエステル
類、ゼラチン、半合成グリセライドなどを挙げる
ことができる。注射剤として調製される場合に
は、液剤および懸濁剤は殺菌され、かつ血液と等
張であるのが好ましく、これら液剤、乳剤および
懸濁剤の形態に成形するのに際しては、稀釈剤と
してこの分野において慣用されているものをすべ
て使用でき、例えば水、エチルアルコール、プロ
ピレングリコール、エトキシ化イソステアリルア
ルコール、ポリオキシ化イソステアリルアルコー
ル、ポリオキシエチレンソルビタン脂肪酸エステ
ル類などを挙げることができる。なお、この場合
等張性の溶液を調製するに充分な量の食塩、ブド
ウ糖あるいはグリセリンを製剤中に包有せしめて
もよく、また通常の溶解補助剤、緩衝剤、無痛化
剤などを添加してもよい。更に必要に応じて着色
剤、保存剤、香料、風味剤、甘味剤などや他の医
薬品を該製剤中に含有せしめてもよい。
かくして調製される医薬製剤(強心剤)中に含
有されるべき一般式(1)の化合物の量はとくに限定
されず広範囲に選択されるが、通常全製剤組成物
中1〜70重量%、好ましくは1〜30重量%とする
のがよい。
また上記強心剤の投与方法にはとくに制限はな
く、各種製剤形態、患者の年令、性別その他の条
件、疾患の程度などに応じた方法で投与される。
例えば錠剤、丸剤、液剤、懸濁剤、乳剤、顆粒剤
およびカプセル剤の場合には経口投与される。ま
た注射剤の場合には単独であるいはブドウ糖、ア
ミノ酸などの通常の補液と混合して静脈内投与さ
れ、さらには必要に応じて単独で筋肉内、皮内、
皮下もしくは腹腔内投与される。坐剤の場合には
直腸内投与される。
上記強心剤の投与量は用法、患者の年令、性別
その他の条件、疾患の程度などにより適宜選択さ
れるが、通常有効成分である一般式(1)の化合物の
量は1日当り体重1Kg当り約0.01〜10mgとするの
がよい。また、投与単位形態中に有効成分を0.1
〜200mg含有せしめるのがよい。
以下に参考例及び実施例を挙げる。
参考例 1
6―(α―ピリジニウムアセチル)―3,4―
ジヒドロカルボスチリルクロライド50g及び水酸
化ナトリウム50gを水1に懸濁し、90〜100℃
で3時間撹拌する。反応終了後濃塩酸を加えてPH
約2とし析出晶を取し、ジメチルホルムアミド
より再結晶して、6―カルボキシ―3,4―ジヒ
ドロカルボスチリル19.1gを得る。

20:1)で単離精製したのち、エタノールから再
結晶して、6―(4―ベンジル―1―ピペラジニ
ルカルボニル)―3,4―ジヒドロカルボスチリ
ル325mgを得る。

ゾイル)―1―ピペラジニルカルボニル〕―3,
4―ジヒドロカルボスチリル1.5gを得る。

上記パラベン類、メタ重亜硫酸ナトリウムおよ
び塩化ナトリウムを撹拌しながら80℃で蒸留水に
溶解する。得られた溶液を40℃で冷却し、これに
本発明化合物、ポリエチレングリコールおよびポ
リオキシエチレンソルビタンモノオレエートを順
次溶解させ、次にその溶液に注射用蒸留水を加え
て最終の容量に調製し、適当なフイルターペーパ
ーを用いて滅菌過して、1mlずつアンプルに分
注し、注射剤を調製する。
<薬理試験>
体重8〜13Kgの雌雄雑種成犬に、ペントバルビ
タールのナトリウム塩を30mg/Kgの割合で静脈内
投与して麻酔させる。ヘパリンのナトリウム塩を
1000U/Kgの割合で静脈内投与後脱血致死させ、
心臓をロツク液中に摘出する。右冠状動脈より洞
結節動脈に向つてカニユーレを挿入し、右心房を
カニユーレと共に摘出する。次いで予めペントバ
ルビタールのナトリウム塩(30mg/Kg、静脈内投
与)により麻酔させ、ヘパリン処理(1000U/
Kg、静脈内投与)した体重18〜27Kgの雌雄雑種成
犬につき、その頚動脈から血液をペリスタリツク
ポンプを介して右冠状動脈に挿入したカニユーレ
に導き、右心房を潅流する。潅流圧は100mmHgの
定圧とする。右心房の運動は静止張力2g下で、
力変位変換器を介して心房筋の収縮力を測定す
る。冠動脈血流量は電磁流量計を用いて測定す
る。全ての記録はインク書き記録計上に記録させ
る。尚この方法の詳細は千葉らにより報告されて
いる〔Japan,J.Pharmacol,25,433〜439
(1975)、Naunyn―Schmiedberg′s Arch.
Pharmacol,289,315〜325(1975)〕。
供試化合物は、右冠状動脈に挿入したカニユー
レに近接して接続したゴムチユーブを介して動脈
内に10〜30μの容量で注射する。供試化合物の
陽性変力作用は、化合物投与前の発生張力に対す
る%変化として表わし、また冠血流量の変化は、
投与前からの絶対値(ml/分)として表わす。結
果を下記第1表に示す。
