Claims (18)
- 특정 염기를 가진 뉴클레오티드 배열이 3'측에 존재하는 수식대상의 뉴클레오티드쇄에, 상기 특정 염기를 포함한 뉴클레오티드에 특이적인 분해효소를 작용시켜서 상기 수식대상의 뉴클레오티드쇄의 3'말단에 소정 수식물질에 대해서 결합능을 가지는 관능기를 부여하고, 상기 관능기를 가진 뉴클레오티드쇄의 3'말단에 상기 수식물질을 결합시키며, 상기 특정 염기로서 히포크산틴을 사용하고, 상기 분해효소로서 3-메틸아데닌 DNA 글리코시다아제를 사용하고, 또한 상기 관능기를 알데히드기로 하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 1항에 있어서, 특정 염기가, 주쇄가 되는 뉴클레오티드쇄의 3'측에 위치하고, 상기 특정 염기가 상기 주쇄에 존재하지 않는 염기인 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 1항에 있어서, 특정 염기를 가진 뉴클레오티드 배열을, 상기 뉴클레오티드쇄의 3'측에 부가하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 3항에 있어서, 특정 염기를 가진 뉴클레오티드 배열을 테일링법에 의해 부가하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 3항에 있어서, 수식대상으로 하는 1개쇄 뉴클레오티드에, 상기 뉴클레오 티드쇄의 3'말단영역에 상보적인 배열을 3'말단영역에 가지는 뉴클레오티드쇄를 어닐하고, 상기 수식대상의 뉴클레오티드쇄의 3'측에 특정 염기를 포함한 상보적 뉴클레오티드 배열을 부가하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 3항에 있어서, 한쪽 또는 양쪽의 뉴클레오티드쇄를 수식대상으로 하는 2개쇄 뉴클레오티드를, 수식대상의 뉴클레오티드쇄의 3'말단영역의 염기배열을 특이적으로 인식하는 제한효소로 절단하고, 절단한 수식대상의 뉴클레오티드쇄의 3'측에 특정 염기를 포함한 상보적 뉴클레오티드 배열을 부가하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 3항에 있어서, 2개쇄 뉴클레오티드의 한쪽 또는 양쪽인 수식대상의 뉴클레오티드쇄의 3'말단측에, 특정 염기를 포함한 뉴클레오티드 배열을 5'말단영역에 가진 2개쇄 뉴클레오티드를 DNA리가아제로 접합하고, 접합한 뉴클레오티드쇄의 상기 특정 염기보다 3'말단측의 염기배열을 특이적으로 인식하는 제한효소로 절단함으로써, 상기 수식대상의 뉴클레오티드쇄의 3'측에 특정 염기를 포함한 뉴클레오티드 배열을 부가하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 3항 또는 제 6항에 있어서, 뉴클레오티드 배열의 부가는 DNA 폴리머라제를 이용해서 뉴클레오티드를 주입함으로써 행하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 1항에 있어서, 특정 염기를 가진 뉴클레오티드 배열이 존재하는 뉴클레오티드쇄가, 화학합성된 뉴클레오티드쇄인 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
- 제 1항에 있어서, 분해효소를 작용시키기에 앞서 특정 염기와 상보적인 염기배열을 가지는 올리고뉴클레오티드를 첨가하는 것을 특징으로 하는 뉴클레오티드쇄 수식방법.

- 제 1항 또는 제 14항에 있어서, 수식물질이, 아미노알칸티올 또는 아미노실란커플링 화합물을 포함하는 유전자해석을 위한 기판으로의 결합능을 가진 물질인 것을 특징으로 하는 뉴클레오티드쇄 수식방법.
