Description
도 2는 독소물질인 마이크로시스틴-LR의 구조식이다.
도 3은 본 발명의 제1 실시예인 조류로부터 색소물질과 독소물질을 동시에 분리 정제하는 방법을 나타내는 흐름도이다.
도 4는 색소물질인 피코시아닌의 검출결과이다(615nm).
도 5는 독소물질인 마이크로시스틴의 검출결과이다(238nm).
도 6은 마이크로시스틴-RR, YR 및 LR의 구조식이다
도 7은 분취형 액체크로마토그래피로 분리한 마이크로시스틴-RR, YR 및 LR 독소의 검출결과이다.

Claims (19)
- 수계에 발생한 조류를 수거하는 S-1 단계;
수거한 조류에 함유된 수분과 조류를 분리하는 제1 원심분리 및 여과하는 S-2 단계;
수분이 제거된 조류를 동결 건조하는 S-3 단계;
인산염 완충액을 첨가한 후 제2 원심분리하여 제2 상등액과 제2 침전물로 분리하는 S-4 단계;
상기 제2 상등액으로부터 색소물질인 피코시아닌을 분리하고, 상기 제2 침전물로부터 독소물질인 마이크로시스틴을 분리하는 S-5 단계; 및
분리된 피코시아닌과 마이크로시스틴을 각각 농축하는 S-6 단계를 포함하되,
상기 S-4 단계에서는 건조된 조류 1g에 대하여 1M 인산용액을 50㎖ 내지 100㎖ 첨가하며, 제2 원심분리단계는 3,500 내지 4,500 rpm에서 15분 내지 25분간 실시하고,
상기 S-5 단계에서의 피코시아닌은 크기배제 크로마토그래피를 이용하여 615nm 파장에서 검출되고, 마이크로시스틴은 상기 제2 침전물에 4 부피% 내지 6 부피%에 해당되도록 초산용액을 첨가하는 단계, 초음파를 조사하는 단계, 제3 원심분리 단계 및 제3 상등액을 수득하는 단계를 포함하되,
상기 초음파를 조사하는 단계에서는 조사강도 18 KHz 내지 22 KHz 및 4~15℃에서 1 내지 2분 조사와 1 내지 2분 정지를 반복하면서 25분 내지 35분 실시하는 것을 특징으로 하는 조류로부터 색소물질과 독소물질을 분리 정제하는 방법.

분리된 색소물질과 독소물질을 농축하는 S-6 단계는 40~50℃에서 감압농축하는 것을 특징으로 하는 조류로부터 색소물질과 독소물질을 분리 정제하는 방법.
- 제5항에 있어서,
상기 감압농축하는 S-6 단계 이후에 색소물질과 독소물질의 순도를 높이는 S-7 단계를 더 포함하는 것을 특징으로 하는 조류로부터 색소물질과 독소물질을 분리 정제하는 방법.
