Abstract
Description
도 2는 본 발명자들이 직접 실험한 완도호랑가시나무(Ilex x wandoensis) 암그루와 수그루, 호랑가시나무(Ilex cornuta) 암그루와 수그루, 감탕나무(Ilex integra) 암그루와 수그루, 먼나무(Ilex rotuda) 암그루와 수그루 각 종에 대해 본 발명의 4 종의 프라이머에 의해 검출된 마커 염기서열 및 NCBI GenBank에서 발췌한 7종(Ilex latifolia, Ilex sp., Ilex delavayi, Ilex szechwanensis, Ilex polyneura, Ilex pubescens, Ilex wilsonii)의 동일 지역의 염기서열을 병합해서 유전적 거리를 측정하고, 유전목(또는 계통수)(genetic tree, Phylogenetic tree)(UPGMA; Unweighted Pair Group Method with Arithmetic Mean)을 도출한 결과를 보여준다.
도 3 내지 6은 상기 각 종에 대해 기존 유니버셜 마커 psbA-trnH(도 3), trnL-trnF IGS(도 4), trnL intron(도 5) 및 엽록체 rbcL(도 6)를 적용하여 유전적 거리를 측정하고, 유전목(genetic tree, UPGMA)을 도출한 결과를 보여준다.

- 제1항의 프라이머 세트를 포함하는 완도호랑가시나무(Ilex x wandoensis)의 모계(Maternal origin) 식별용 조성물.
- 제1항의 프라이머 세트를 포함하는 완도호랑가시나무(Ilex x wandoensis)의 모계(Maternal origin) 식별용 키트.
- 다음 단계를 포함하는 완도호랑가시나무(Ilex x wandoensis)의 모계(Maternal origin) 식별 방법:
(a) 시료에서 DNA를 분리하는 단계;
(b) 상기 분리된 DNA를 주형으로 하고, 제1항의 프라이머 세트를 이용하여 PCR(Polymerase Chain Reaction)을 실시하여 표적 서열을 증폭하는 단계; 및
(c) 상기 증폭 산물을 검출하는 단계. - 제5항에 있어서, 상기 (b) 단계에서 PCR의 프라이머 어닐링 온도(annealing temperature)는 45℃ 내지 55℃인 것을 특징으로 하는, 방법.
