Claims (15)
- 서열 2의 아미노산 서열을 포함하는 폴리펩타이드를 암호화하는 폴리뉴클레오타이드와의 상동성이 70% 이상인 폴리뉴클레오타이드 a),언급된 폴리펩타이드를 암호화하고 플라스미드 pEK-pckA(도 1) 또는 pEK-pckB(도 2)에 함유된 폴리뉴클레오타이드와의 상동성이 70% 이상인 폴리뉴클레오타이드 b),서열 2의 아미노산 서열과의 상동성이 70% 이상인 아미노산 서열을 포함하는 폴리펩타이드를 암호화하는 폴리뉴클레오타이드 c),위의 폴리뉴클레오타이드 a), b) 또는 c)에 상보성인 폴리뉴클레오타이드 d) 및위의 폴리뉴클레오타이드 서열 a), b), c) 및 d)의 15개 이상의 연속하는 염기를 포함하는 폴리뉴클레오타이드 e)로 이루어진 그룹으로부터 선택된 폴리뉴클레오타이드 서열을 포함하는, 코리네형 박테리아로부터 분리된 폴리뉴클레오타이드.
- 제1항에 있어서, 폴리뉴클레오타이드가 복제가능한, 바람직하게는 재조합 DNA인 폴리뉴클레오타이드.
- 제1항에 있어서, 폴리뉴클레오타이드가 RNA인 폴리뉴클레오타이드.

- 도 1에 제시된 벡터 pEK-pckA.
- 도 2에 제시된 벡터 pEK-pckB.
- 도 3에 제시되고 균주 이. 콜라이 DH5α로서 기탁번호 DSM 13047하에 기탁된 벡터 pk19mobsacBΔpck.
- 제6항 내지 제8항 중의 어느 한 항에 따른 벡터중 하나를 함유하거나 Δpck 결실체가 도입되어 있는, 숙주 세포로서 사용되는 코리네형 박테리아.
- 제1항에 따른 폴리뉴클레오타이드가 약독화되거나, 위의 폴리뉴클레오타이드를 암호화하는 폴리펩타이드의 활성이 감소된 박테리아 a),목적 생성물이 배지 또는 박테리아 세포에서 농축된 박테리아 b) 및생성물이 분리된 박테리아 c)를 사용하는, 발효에 의한 L-아미노산의 제조 방법.
- 제11항에 있어서, 코리네박테리움 글루타미쿰(Corynebacterium glutamicum) 속 박테리아가 사용되는 방법.
- 제12항에 있어서, 약독화가 도 3에 제시되고 DSM 13047로서 기탁된 플라스미드 pK19mobsacBΔpck의 보조하에 삽입 돌연변이유발에 의해 수행되는 방법.
- 제12항에 있어서, 임의로 디하이드로디피콜리네이트 신타제를 암호화하는 dapA 유전자를 동시에 과발현시키고/시키거나,S-(2-아미노에틸)시스테인 내성을 부여하는 DNA 단편을 동시에 증폭시키는 박테리아를 발효시켜 L-리신을 제조하는 방법.
- 제12항에 있어서, 임의로 호모세린 데하이드로게나제를 암호화하는 hom 유전자 및/또는 "피드백 내성" 호모세린 데하이드로게나제를 암호화하는 homdr또는 homFBR대립인자를 동시에 과발현시키는 박테리아를 발효시켜 L-트레오닌을 제조하는 방법.
