Claims (9)
- 세균 내에서 트레할로스 합성 능력이 감소되거나 결실된, L-글루탐산 생산 능력을 갖는 코리네형 세균.
- 제1항에 있어서, 트레할로스 합성 능력이 트레할로스 합성 경로의 효소를 암호화하는 염색체 유전자 속으로 돌연변이를 도입하거나 당해 유전자를 파괴함으로써 감소되거나 결실되는 코리네형 세균.
- 제2항에 있어서, 트레할로스 생합성 경로의 효소를 암화화하는 유전자가 트레할로스-6-포스페이트 신타제를 암호화하는 유전자, 말토올리고실트레할로스 신타제를 암호화하는 유전자 또는 이들 유전자 둘 다로 이루어지는 코리네형 세균.
- 제3항에 있어서, 트레할로스-6-포스페이트 신타제를 암호화하는 유전자가 서열 30의 아미노산 서열을 암호화하고, 말토올리고실트레할로스 신타제를 암호화하는 유전자가 서열 32의 아미노산 서열을 암호화하는 코리네형 세균.
- 제1항 내지 제4항 중의 어느 한 항에 따르는 코리네형 세균을 배지에서 배양하여 배지에 L-글루탐산을 생산, 축적하는 단계 및당해 배지로부터 L-글루탐산을 수집하는 단계를 포함하는, L-글루탐산의 생산방법.
- 다음의 (A) 또는 (B)에서 정의되는 단백질을 암호화하는 DNA:(A) 서열 30의 아미노산 서열을 갖는 단백질,(B) 하나 이상의 아미노산 잔기의 치환, 결실, 삽입 또는 첨가를 포함하고 트레할로스-6-포스페이트 신타제 활성을 갖는, 서열 30의 아미노산 서열을 갖는 단백질.
- 제6항에 있어서, 다음의 (a) 또는 (b)에서 정의되는 DNA:(a) 서열 29의 뉴클레오타이드 서열의 484번 내지 1938번 뉴클레오타이드 잔기를 적어도 포함하는 뉴클레오타이드 서열을 함유하는 DNA,(b) 엄격한 조건(stringent condition)하에서 서열 29의 뉴클레오타이드 서열의 484번 내지 1938번 뉴클레오타이드 잔기를 적어도 포함하는 뉴클레오타이드 서열과 하이브리드화 가능하고, 위의 뉴클레오타이드 서열과 55% 이상의 상동성을 나타내며, 트레할로스-6-포스페이트 신타제 활성을 갖는 단백질을 암호화하는 DNA.
- 다음의 (A) 또는 (B)에서 정의되는 단백질을 암호화하는 DNA:(A) 서열 32의 아미노산 서열을 갖는 단백질,(B) 하나 이상의 아미노산 잔기의 치환, 결실, 삽입 또는 첨가를 포함하고 말토올리고실트레할로스 신타제 활성을 갖는 서열 32의 아미노산 서열을 갖는 단백질.
- 제8항에 있어서, 다음의 (a) 또는 (b)에서 정의되는 DNA:(a) 서열 31의 뉴클레오타이드 서열의 82번 내지 2514번 뉴클레오타이드 잔기를 적어도 포함하는 뉴클레오타이드 서열을 함유하는 DNA,(b) 엄격한 조건하에서 서열 31의 뉴클레오타이드 서열의 82번 내지 2514번 뉴클레오타이드 잔기를 적어도 포함하는 뉴클레오타이드 서열과 하이브리드화 가능하고, 위의 뉴클레오타이드 서열과 60% 이상의 상동성을 나타내며, 말토올리고실트레할로스 신타제 활성을 갖는 단백질을 암호화하는 DNA.
