Description
도 2는 실시예 1-2의 발현벡터를 나타낸 것이다. A는 glaA 프로모터와 pdcA 종결인자를 사용하고 자체 신호서열을 포함하는 cDNA를 클로닝 할 수 있는 벡터(pASP503)를 나타내고, B는 glaA 프로모터와 pdcA 종결인자를 사용하고 An07g00440 리파제 신호서열을 포함하는 벡터(pASPW504)를 나타내며, C는 glaA 프로모터와 pdcA 종결인자를 사용하고 An07g00440 리파제 변형 신호서열을 포함하는 벡터(pASPV505)를 나타낸다.
도 3은 실시예 5의 재조합미생물 배양액 분석결과를 나타낸 것으로, A는 재조합미생물 pASPF503PMC의 배양액 분석결과, B는 재조합미생물 pASPW504PMC의 배양액 분석결과, C는 재조합미생물 pASPV505PMC의 배양액 분석결과를 나타낸다.

- 서열번호 1의 아미노산 서열로 표시되는 An07g00440 리파제 신호서열.
- 제1항에 있어서, 아스퍼질러스 나이거(Aspergillus niger) 유래인 것을 특징으로 하는 An07g00440 리파제 신호서열.
- 제1항의 An07g00440 리파제 신호서열을 코딩하는 핵산.
- 제3항에 있어서, 서열번호 3의 염기서열로 표시되는 것을 특징으로 하는 핵산.
- 제1항의 An07g00440 리파제 신호서열의 2번째 아미노산인 타이로신(tyrosine; T)이 양전하를 띠는 알지닌(arginine; R)으로 치환되고, 3번째 아미노산인 아이소루신(isoleucine; I)이 트립토판(tryptophane; T)으로 치환된 것을 특징으로 하는, 서열번호 2의 아미노산 서열로 표시되는 변형 리파제 신호서열.
- 제5항의 변형 리파제 신호서열을 코딩하는 핵산.
- 제6항에 있어서, 서열번호 4의 염기서열로 표시되는 것을 특징으로 하는 핵산.
- 제1항 또는 제5항의 리파제 신호서열을 코딩하는 핵산 및 목적단백질의 유전자를 포함하는 발현벡터.
- 제8항에 있어서, 상기 목적단백질은 카탈라아제인 것을 특징으로 하는 발현벡터.
- 제9항에 있어서, 상기 카탈라아제는 서열번호 5의 페니실륨 마네페이(Penicillium marneffei) 카탈라아제 단백질인 것을 특징으로 하는 발현벡터.
- 제8항에 있어서, 글루코아밀라아제 프로모터를 추가로 포함하는 것을 특징으로 하는 발현벡터.
- 제8항에 있어서, pASPW504PMC 또는 pASPV505PMC인 것을 특징으로 하는 발현벡터.
- 제1항 또는 제5항의 리파제 신호서열을 코딩하는 핵산과 목적단백질의 유전자가 도입되어 있는 재조합 미생물.
- 제8항의 발현벡터가 도입되어 있는 재조합미생물.
- 제14항에 있어서, 아스퍼질러스 나이거(Aspergillus niger)인 것을 특징으로 하는 재조합미생물.
- 다음 단계를 포함하는 것을 특징으로 하는 목적단백질의 제조방법:
(a) 제13항의 재조합미생물을 배양하여 목적단백질을 생성하는 단계; 및
(b) 상기 생성된 목적단백질을 회수하는 단계.
