Abstract
본 발명의 인간 HO-1과 TNFR1-Fc 융합 유전자를 동시 발현하며 GGTA1 유전자가 넉아웃된 형질전환 돼지는 HO-1 유전자의 항산화 작용 및 세포 보호 기능 등에 의해 장기 분리 및 체외 배양 시에 산화적 스트레스를 감소시킬 수 있고, TNFR1-Fc 발현에 의해 이식 초기에 일어나는 TNF-α 매개의 염증 반응도 감소시킬 수 있으며, 또한 수지상 세포의 성숙을 억제하고 T 세포의 증식과 활성화를 조절하여 급성 혈관계 거부 반응을 감소시켜 이식된 장기의 초기 생착을 촉진할 수 있다. 나아가, GGTA1에 의한 초급성 면역 거부반응을 억제함으로써 이식된 장기의 생존율을 증가시킬 수 있다. 이에, 상기 형질전환 돼지를 이용하여 이종 이식 시 면역 거부 반응이 억제된 장기를 생산할 수 있다.

Claims (11)
- a) 돼지로부터 체세포를 분리하는 단계;
b) 상기 체세포에 인간 HO-1 (Heme oxygenase-1) 유전자 및 인간 TNFR1-Fc (Tumor necrosis factor receptor 1-Fc) 융합 유전자를 도입시키고, GGTA1 (α-1,3-galactosyltransferase) 유전자를 넉아웃 (knock-out)시키는 단계;
c) 인간 HO-1 유전자 및 인간 TNFR1-Fc 융합 유전자가 도입되고 GGTA1 유전자가 넉아웃된 체세포를 선별한 후 배양하는 단계; 및
d) 돼지 난자의 핵을 제거하고 상기 선별된 체세포와 융합하여 체세포 핵이 이식된 수정란을 제조하는 단계를 포함하는, 이종 장기 이식 시 면역 거부 반응이 억제된 형질전환 돼지의 제조 방법.
- 제1항에 있어서, 상기 b) 단계의 인간 HO-1 유전자는 서열번호 1의 염기서열을 포함하는 것인, 방법.
- 제1항에 있어서, 상기 b) 단계의 인간 TNFR1-Fc 융합 유전자는 서열번호 8의 염기서열을 포함하는 것인, 방법.
- 제1항에 있어서, 상기 b) 단계는 인간 HO-1 유전자를 포함하는 단일 벡터 및 인간 TNFR1-Fc 융합 유전자를 포함하는 단일 벡터를 별도로 또는 동시에 체세포에 도입시키는 것인, 방법.
- 제1항에 있어서, 상기 유전자 넉아웃은 TALEN (transcription activator-like effector nuclease)을 이용하여 수행되는 것인, 방법.

- 인간 HO-1 (Heme oxygenase-1) 유전자 및 인간 TNFR1-Fc (Tumor necrosis factor receptor 1-Fc) 융합 유전자가 도입되고, GGTA1 (α-1,3-galactosyltransferase) 유전자가 넉아웃 (knock-out)된, 이종 장기 이식 시 면역 거부 반응이 억제된 장기 이식용 형질전환 돼지.
- 제8항의 형질전환 돼지를 제조하기 위한 체세포 공여 세포주.
- 제1항의 방법에 의해 형질전환 돼지를 제조하는 단계; 및
상기 형질전환 돼지로부터 이식용 장기를 분리하는 단계를 포함하는, 이종 장기 이식 시 면역 거부 반응이 억제된 장기를 제조하는 방법.
- 제10항에 있어서, 상기 장기는 췌도, 췌장, 심장, 신장, 간장, 폐장 및 각막으로 이루어진 군에서 선택되는 1 이상인 것인, 방법.
