Claims (11)
- 성장인자를 포함하는 줄기세포 배양액을 준비하는 단계와;
상기 줄기세포 배양액에 성장인자 보호제를 혼합하여 혼합액을 형성하는 단계와;
상기 혼합액을 동결건조하여 동결건조 조성물을 얻는 단계를 포함하는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제 1항에 있어서,
상기 줄기세포 배양액을 준비하는 단계는,
배지에 상기 성장인자가 배출되도록 줄기세포를 배양하고, 상기 줄기세포를 상기 배지로부터 여과(filter)하여 얻는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제 1항에 있어서,
상기 성장인자는,
혈소판유래성장인자(platelet-derived growth factor, PDGF), 섬유아세포성장인자(fibroblast growth factor, FGF), 형질전환성장인자(trans-forming growth factor, TGF), 각질세포성장인자(keratinocyte growth factor, KGF), 간세포성장인자(hepatocyte growth factor, HGF), 혈관내피성장인자(vascular endothelial growth factor, VEGF), 콜라겐(collagen), 파이브로넥틴(fibronectin), 항산화효소(superoxide dismutase, SOD) 및 이의 혼합으로 이루어진 군으로부터 선택되는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제 1항에 있어서,
상기 성장인자는 상기 줄기세포 배양액 내에 150 내지 2,000pg/mL 농도로 포함된 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제 1항에 있어서,
상기 성장인자 보호제는 이당류, 글루코스 및 사이클로덱스트린을 포함하는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제 5항에 있어서,
상기 이당류는,
맥아당(malt sugar), 셀로비오스(cellobiose), 겐티오비오스(gentiobiose), 멜리비오스(melibiose), 락토스(lactose), 투라노오스(turanose), 소포로오스(sophorose), 수크로오스(sucrose) 및 이의 혼합으로 이루어진 군으로부터 선택되는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제5항에 있어서,
상기 혼합액 전체 100중량부 중 상기 이당류는 0.01 내지 0.3중량부, 상기 글루코스는 0.05 내지 0.5중량부, 상기 사이클로덱스트린은 0.02 내지 0.6중량부를 포함하는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 제 1항에 있어서,
상기 동결건조는,
-20 내지 -30℃에서 1차동결 및 -70 내지 -100℃에서 2차동결한 후, 진공 상태에서 20 내지 25℃에서 1차건조 및 30 내지 40℃에서 2차건조하여 온도가 점진적으로 조절되는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물 제조방법. - 성장인자를 포함하는 줄기세포 배양액에 성장인자 보호제를 혼합하여 혼합액을 형성한 후, 상기 혼합액을 동결건조하여 얻어지는 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물.
- 제 9항에 있어서,
상기 동결건조 조성물은,
상기 혼합액 상태의 성장인자 농도로부터 동결건조 직후 성장인자의 농도 감소율이 5 내지 20%인 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물. - 제 9항에 있어서,
상기 동결건조 조성물은,
상기 동결건조 직후 성장인자의 농도로부터 실온에서 보관되는 상태의 분말형태 동결건조 조성물 성장인자의 농도 감소율이 5 내지 20%인 것을 특징으로 하는 줄기세포 성장인자를 포함하는 동결건조 조성물.
