Claims (21)
- 중합 가능한 분리기 물질(여기서, 중합 가능한 분리기 물질은 올리고머, 광개시제, 및 안정화제를 포함하고, 전혈과 유동 가능하고, 제1 평균 분자량을 가지며, PRP 분획의 평균 밀도와 비-PRP 분획의 평균 밀도 사이의 밀도를 갖는다)을 포함하는 수집관에서 전혈의 샘플로부터 PRP 분획을 분리하는 방법으로서,
원심분리 프로토콜을 사용하여, 중합 가능한 분리기 물질을 포함하는 수집관에서 전혈의 샘플을 원심분리하는 단계;
(여기서, 원심분리 프로토콜을 사용한 원심분리는 중합 가능한 분리기 물질이 PRP 분획에서 혈소판의 상당한 활성화 없이 PRP 분획과 비-PRP 분획 사이를 유동할 수 있게 하고, 상당한 활성화는 PRP 분획 중의 혈소판이 리스토세틴 및 콜라겐 중의 적어도 하나에 응집하는 능력에 기초하여 측정 가능하다);
상기 분리기 물질을, UV 에너지에의 노출을 통해, 혈소판의 상당한 활성화 없이 경화시켜 고체 차단막을 형성하는 단계로서, 고체 차단막의 중합 가능한 분리기 물질은 제1 평균 분자량보다 큰 제2 평균 분자량을 갖는, 단계; 및
(여기서, 고체 차단막은, UV 에너지에의 노출 후, 수집관에 대해 정지되며 수집관으로부터 PRP 분획의 적어도 95%를 제거하도록 PRP 분획 아래 및 비-PRP 분획 위의 중간 위치에 있다)
를 포함하는, 방법. - 청구항 1에 있어서, 상기 PRP 분획이 전혈의 샘플로부터 10% 이하의 백혈구를 포함하고, 상기 PRP 분획 중의 백혈구의 25% 이하가 과립구인, 방법.
- 청구항 1에 있어서, 상기 경화 단계 이후 적어도 1일 후에 상기 수집관에서 상기 PRP 분획을 균질화시키는 단계를 추가로 포함하고, 여기서, 상기 PRP 분획의 균질화 단계가, 이로부터 취한 상이한 분취액이 사용된 세포 계수법의 1 변동 계수 (one coefficient of variation)내에 있는 혈소판 수를 갖도록 실질적으로 균일한 PRP 분획을 야기하는, 방법.
- 청구항 1에 있어서, 상기 PRP 분획이 상기 전혈의 샘플의 혈소판 농도의 적어도 200%인 혈소판 농도를 갖는, 방법.
- 청구항 1에 있어서, 상기 PRP 분획이 상기 전혈의 샘플의 혈소판 농도의 180%(포함) 내지 260%(포함)인 혈소판 농도를 갖는, 방법.
- 청구항 5에 있어서, 상기 PRP 분획이 상기 전혈의 샘플의 혈소판 농도의 200%(포함) 내지 250%(포함)인 혈소판 농도를 갖는, 방법.
- 청구항 1에 있어서, 상기 분리기 물질이 올리고머, 광개시제 및 안정화제를 포함하고, 상기 광개시제가 5% 미만의 농도도 존재하며, 상기 안정화제가 0.5 wt% 미만의 농도로 존재하는, 방법.
- 청구항 1에 있어서, 상기 수집관이 튜브 캡(tube cap)을 포함하고,
상기 캡의 적어도 일부를 뚫고 상기 수집관에서 PRP 분획과 접촉하도록 하는 크기 및 치수의 니들을 포함하는 어댑터를 제공하는 단계를 추가로 포함하며;
여기서, 상기 니들이 유체 분배기에 유동적으로 연결되고;
상기 니들이 상기 수집관에 적어도 부분적으로 있는 경우 상기 유체 분배기가 수집관의 바깥에 있으며;
상기 유체 분배기가 점안기로서 배열되고 PRP 분획의 멸균되고, 조절된 점적식 분배를 가능하게 하는 메카니즘을 포함하는, 방법. - 청구항 8에 있어서, 상기 유체 분배기가 작은 튜브 및 진공 벌브를 포함하고, 한번에 단일 방울(single drop)로 제1 말단으로부터 PRP 분획을 채취하고 제2 말단으로부터 PRP 분획을 방출하도록 구성되는, 방법.
- 청구항 8에 있어서, 상기 메카니즘이 적어도 제1 일방향 밸브를 포함하는, 방법.
- 청구항 1에 있어서, 상기 수집관이 튜브 캡을 포함하고, PRP 분획을 이동시키기 위해 어댑터를 사용하는 단계를 추가로 포함하며, 상기 어댑터가
제1 내부 구획, 제2 내부 구획, 및 제1 및 제2 개방 말단 사이에 배치된 기저 구조를 갖는 하우징;
(여기서, 기저 구조는 하우징의 내부면에 고정되어 연결된 제1 기저 부분, 및 하우징의 내부면에 이동 가능하게 연결된 제2 기저 부분을 포함한다);
제1 기저 부분에서 제1 내부 구획으로 뻗어 있는 제1 니들 부분;
제1 기저 부분에서 제2 내부 구획으로 뻗어 있는 제2 니들 부분;
제2 기저 부분에서 제1 내부 구획으로 뻗어 있는 제3 니들 부분; 및
상기 하우징을 통해 뻗어 나와 제3 니들 부분에 연결되는 벤트
를 포함하는, 방법. - 이동 장치로서,
하우징 내에 배치된 제1 니들, 제2 니들, 및 기저 구조를 포함하고;
여기서, 상기 하우징이 제1 및 제2 개방 말단을 갖고, 상기 기저의 반대 말단에 제1 및 제2 내부 구획을 포함하며;
제1 니들이 상기 기저 구조의 제1 부분에 고정되고, 상기 하우징의 제1 내부 구획 내에 위치한 제1 말단, 및 상기 하우징의 제2 내부 구획 내에 위치한 제2 말단을 포함하며;
제2 니들이 상기 기저 구조의 제2 부분에 연결되고, 상기 하우징의 제1 구획 내에 위치한 제1 말단, 및 벤트 구조에 연결된 제2 말단을 포함하며;
상기 제2 니들이 상기 하우징의 제1 개방 말단을 기준으로 한 제1 말단의 위치가 조정 가능하도록 제1 위치에서 제2 위치로 이동 가능한, 이동 장치. - 청구항 12에 있어서, 상기 제2 말단이 비스듬하게 상기 벤트 구조에 연결되는, 장치.
