Description
도 2는 재조합 pMT/BiP-MERS-RBD-V5-His 플라스미드의 모식도를 나타낸 것이다.
도 3은 재조합 pMT/BiP-MERS-RBD-V5-His 플라스미드로 형질전환된 드로소필라 멜라노개스터 S2 세포의 세포 단백질 추출물 (intracellular fraction) 및 배양 배지 (medium fraction)에서 재조합 MERS-RBD 단백질의 존재를 anti-V5 항체를 이용한 웨스턴블럿 분석 결과이다.
도 4는 재조합 pMT/BiP-MERS-RBD-V5-His 플라스미드로 형질전환된 드로소필라 멜라노개스터 S2 세포의 배양 배지에서 Ni-NTA 수지를 이용한 친화성 크로마토그래피 방법으로 재조합 MERS-RBD 단백질을 분리 정제하고 SDS-PAGE 후 은 염색법 (silver staining)으로 확인한 결과이다.
도 5는 하이브리도마 세포주에서 생산된 메르스 바이러스 단일클론 항체의 재조합 메르스 바이러스 항원단백질 MERS-RBD에 대한 결합 능력을 ELISA 방법으로 확인한 결과이다.
Claims (13)
- 메르스 바이러스 스파이크 단백질의 수용체 결합 도메인 (MERS-RBD)을 코딩하는 핵산을 포함하는 발현 벡터를 드로소필라 멜라노게스터 (Drosophila melanogaster) S2 세포에 도입하여 형질전환 세포를 제조하는 단계; 및
상기 형질전환 세포를 배양 배지에서 배양하는 단계를 포함하는, MERS-RBD 항원의 제조 방법. - 제1항에 있어서, 상기 MERS-RBD는 메르스 바이러스 스파이크 단백질의 377-588 번째 아미노산으로 이루어진 것인, 방법.
- 제1항에 있어서, 상기 MERS-RBD를 코딩하는 핵산은 서열번호 2의 염기서열로 이루어진 것인, 방법.
- 제1항에 있어서, 상기 발현 벡터는 단백질 분비 신호 (secretion signal) 서열을 추가로 포함하는 것인, 방법.
- 제1항에 있어서, 상기 형질전환 세포를 배양한 배양 배지로부터 MERS-RBD 항원을 회수하는 단계를 추가로 포함하는 것인, 방법.
- 메르스 바이러스 스파이크 단백질의 수용체 결합 도메인 (MERS-RBD)을 코딩하는 핵산을 포함하는 발현 벡터를 드로소필라 멜라노게스터 (Drosophila melanogaster) S2 세포에 도입하여 형질전환 세포를 제조하는 단계;
상기 형질전환 세포를 배양 배지에서 배양하는 단계;

상기 MERS-RBD 항원에 대한 단일클론항체를 생산하는 단계를 포함하는, MERS-RBD 항체의 제조 방법. - 제6항에 있어서, 상기 MERS-RBD는 메르스 바이러스 스파이크 단백질의 377-588 번째 아미노산으로 이루어진 것인, 방법.
